To evaluate the effects of low-level laser therapy (LLLT) on the proliferation, migration, and tube formation of human umbilical vein endothelial cells (HUVECs) and their related mechanisms.

LLLT contributes to the enhancement of HUVEC proliferation, migration, and tube formation. The activation of the PI3K/Akt signaling pathway is critical in promoting LLLT-induced cell proliferation, migration, and angiogenesis as PI3K inhibitor effectively decreases the effects inducing by LLLT exposure.
研究不足
The research focused mainly on in vitro cell experiments. It will be more credible if identical results are obtained from in vivo experiments and clinical trials. An in-depth investigation about molecules eNOS, VEGFA and HIF-1α involving the effect of LLLT on angiogenesis is required.
1:Experimental Design and Method Selection:
HUVECs were exposed to laser irradiation under different laser parameters to choose the optimal parameters. The effects of LLLT on HUVEC proliferation, migration, and tube formation were examined, along with the levels of angiogenesis-related genes and the PI3K/Akt signaling pathway.
2:Sample Selection and Data Sources:
HUVECs were provided by Shanghai Fuheng Biology Co., Ltd., and passed the inspection of short tandem repeat (STR) genotyping.
3:List of Experimental Equipment and Materials:
A portable semiconductor based GaAlAs laser was used for laser irradiation. The PI3K signaling pathway inhibitor (LY294002) was from Abcam Inc.
4:Experimental Procedures and Operational Workflow:
HUVECs were cultured and treated with LLLT under different parameters. Cell viability, proliferation, migration, and tube formation were assessed. The PI3K/Akt signaling pathway was explored using Western blot assay.
5:Data Analysis Methods:
Statistical analyses were processed with SPSS 21.0 statistical software. Comparisons among groups were analyzed using one-way analysis of variance (ANOVA) or repeated measurement ANOVA.
