To examine the comparative effect of He–Ne laser irradiated seeds of moringa from Egypt on agro-morphological criteria, genomic DNA stability using ISSR markers and protein profiling using SDS-PAGE.

He–Ne laser treatments of wetted seeds achieve biostimulation in many aspects of agro-morphological criteria. ISSR analysis is a highly sensitive method for the detection of DNA alteration induced by laser treatments. The exposure of dry and wetted seeds to He–Ne laser at different times affected the genomic template stability percentage (GTS %). Also, the laser treatments have a great effect on gene expression by switching on or off some genes that may lead to the presence or absence of certain proteins, respectively.

The study focused on short-term effects of laser treatment on moringa seeds and seedlings. Long-term effects on mature plants and yield were not investigated. The study was conducted under controlled conditions, and field trials may yield different results.
1:Experimental Design and Method Selection:
The study investigated the effect of He–Ne laser irradiation on dry and wetted moringa seeds at different exposure times (0, 25, 45, and 90 min). Agro-morphological criteria, SDS-PAGE protein banding patterns, and ISSR markers were used to assess the influence of laser treatment.
2:Sample Selection and Data Sources:
Moringa oleifera L. seeds were collected from the farm of the Egyptian Scientific Society of Moringa, National Research Centre, Giza, Egypt.
3:List of Experimental Equipment and Materials:
A helium–neon (He–Ne) laser with an average power density of 10 mW cm2 (wavelength of
4:8 nm and beam diameter = 5 cm) was used. Experimental Procedures and Operational Workflow:
6 Seeds were divided into dry and wetted groups and irradiated for 25, 45, and 90 min. Agro-morphological criteria were recorded after 50 days from planting. Molecular analysis included ISSR-PCR and SDS-PAGE.
5:Data Analysis Methods:
Data of agro-morphological criteria were statistically analyzed using the Co-State statistical program. ISSR profiles were analyzed by Total Lab program to find out the molecular size of each band. SDS-PAGE bands were analyzed by Total Lab program to find out the molecular mass of each band.
