1:Experimental Design and Method Selection:
Electroretinography, optokinetic response measurements (OKR), immunohistochemistry, and electron microscopy analyses were employed to assess ribbon synapse function, protein expression, and ultrastructure in 5-day-old zebra?sh larvae. Expression of wrb was investigated with real-time qRT-PCR and in situ hybridization.
2:Sample Selection and Data Sources:
Zebra?sh larvae at 5 days post fertilization (dpf) were used. The mutant wrbhi1482 was initially identi?ed in a forward genetic screen for mutants affecting ocular development or function.
3:List of Experimental Equipment and Materials:
Equipment included a commercial system (CFX96; Bio-Rad Laboratories) for real-time PCR, a ?uorescent microscope (Zeiss Axio Imager.Z2 with Apotome.2), and a digital electron microscope (Tecnai 20, 200 kV; Philips Corp.). Materials included antibodies for immunohistochemistry and immunoblotting, and morpholino antisense oligonucleotides for knockdown experiments.
4:Experimental Procedures and Operational Workflow:
Larvae were dark-adapted for 1 hour, euthanized, and eyes removed for ERG recording. For immunohistochemistry, larvae were ?xed, cryoprotected, and sectioned. For electron microscopy, larvae were prepared and sectioned for imaging.
5:Data Analysis Methods:
ERG sweeps were extracted and analyzed with custom scripts using data analysis software (IgorPro). Statistical analyses were performed using commercial software (Prism 6).
