To review the recent advances in mitochondria-targeted materials for cancer phototherapy (PT), including photodynamic therapy (PDT) and photothermal therapy (PTT), and to summarize the combination of mitochondria-targeted PT with other anticancer strategies.

Mitochondria-targeted PT holds great promise for clinical use to combat cancer due to its high cancer selectivity, ability to overcome drug resistance, and potential to enhance the immune response. However, challenges such as potential cytotoxicity and the need for further development of targeting ligands remain.

The potential toxicity of lipophilic cations and peptides when they accumulate in mitochondria, the degradation of peptides in vivo leading to failed mitochondria-targeting, and the large molecular weights of aptamers that may affect the properties of the target materials.
