The study found significant differences in reduced scattering coefficient and scattering anisotropy between bruised and non-bruised blueberry flesh, particularly in the near-infrared region (930-1400 nm), making it effective for non-destructive bruise detection using reflectance or transmittance methods. The skin's high absorption and scattering in the visible range limit detection effectiveness there. MCML simulations confirmed the feasibility of detecting early bruising in the near-infrared spectrum.

The study did not account for the curvature effect of blueberries in MCML simulation, and there were potential errors from sample preparation, such as skin wrinkling and thickness measurement inaccuracies. The spectral ranges were limited to 500-800 nm and 930-1400 nm due to signal-to-noise issues.
1:Experimental Design and Method Selection:
The study used a single integrating sphere-based spectroscopic system to measure total reflectance, total transmittance, and collimated transmittance spectra. The inverse adding-doubling (IAD) method was applied to calculate optical properties, and Monte Carlo multi-layered (MCML) simulation was used to model light propagation.
2:Sample Selection and Data Sources:
Blueberries of cultivars 'Farthing' and 'Meadowlark' were hand-picked from a commercial farm. Samples included non-bruised, 30-min bruised, and 24-h bruised flesh and skin tissues. A total of 150 blueberries were used.

Equipment included an integrating sphere (model 4P-GPS-060-SF, Labsphere), spectrometers (USB2000+ Ocean Optics, SNIR0018 Control Development), collimators (F240SMA-B and F240SMA-C, Thorlabs), light source (DC-950 Dolan-Jenner), optical fiber (M32L02 Thorlabs), microscope slides (Fisher finest Premium), digital micrometer (model 35-025 iGaging), Abbe refractometer (Abbe Mark II Reichert), digital refractometer (Digital Palette Refractometer PR-101α Atago), and microscope (BA410 Motic). Materials included Nigrosin, Naphthol Green B, Intralipid 20% (Sigma-Aldrich), and distilled water.
4:Experimental Procedures and Operational Workflow:
Samples were prepared by slicing flesh to 3 mm thickness and peeling skin. Spectra were collected in configurations for total reflectance, total transmittance, and collimated transmittance. System validation was done with liquid phantoms. Optical properties were calculated using IAD, and MCML simulation was performed for light propagation analysis.
5:Data Analysis Methods:
Data were processed using Savitzky-Golay filter, IAD algorithm, and t-tests for statistical significance. MCML simulation used programs for photon transport and convolution.
