The developed NIR light-responsive SO2 gas therapy platform based on RUCSNs-DM is effective for in vivo anti-tumor therapy, showing high biocompatibility, efficient SO2 generation, induction of apoptosis via ROS increase and DNA damage, and significant tumor growth inhibition with prolonged survival in mice.

1:Experimental Design and Method Selection:
Designed a platform using rattle-structured upconversion@silica nanoparticles (RUCSNs) loaded with a SO2 prodrug (DM) to enable NIR light-triggered SO2 release. Utilized upconversion nanoparticles to convert NIR to UV light for prodrug activation.
2:Sample Selection and Data Sources:
Used cancer cell lines (HeLa, MCF-7, S180) and tumor-bearing mice models for in vitro and in vivo studies.

Included transmission electron microscopy (TEM), UV-vis spectroscopy, fluorescence spectrometry, confocal laser scanning microscopy (CLSM), flow cytometry, inductively coupled plasma optical emission spectrometer (ICP-OES), and various chemicals like Y(ac)3, Yb(ac)3, Tm(ac)3, oleic acid, TEOS, etc.
4:Experimental Procedures and Operational Workflow:
Synthesized RUCSNs and loaded with DM; characterized nanoparticles; conducted in vitro cytotoxicity, ROS, apoptosis assays; performed in vivo biodistribution and therapy studies with NIR irradiation.

Used statistical methods (mean ± s.d., n=3 or n=5), fluorescence intensity measurements, and imaging analysis.
