To develop and characterize bifunctional liposomes (GdLip) that serve as carriers for a gadolinium-based MRI contrast agent and a zinc phthalocyanine photosensitizer for cancer theranostics, and to evaluate their physicochemical properties, relaxivity, singlet oxygen production, cytotoxicity, and photodynamic activity.

Bifunctional liposomes (GdLip) were successfully developed and characterized, showing high relaxivities for MRI contrast and effective photodynamic activity with ZnPc. Formulations with anionic lipids (PG) exhibited more desirable properties than cationic ones. Pegylation improved relaxivity but reduced singlet oxygen production. GdDTPA1 did not affect photocytotoxicity. These liposomes hold promise for theranostic applications, with recommendations to use more stable gadolinium chelates in future studies.

The study is in vitro, so in vivo efficacy and toxicity are not addressed. The stability of GdDTPA1 chelate may be lower than macrocyclic alternatives, potentially limiting clinical use. Liposome size polydispersity was not fully optimized due to omission of purification steps. The mechanism behind ZnPc's effect on relaxivity is unclear and requires further investigation. Passive targeting via EPR effect may not be sufficient for specificity in vivo.

The study designed various liposome formulations with different lipid compositions (anionic, cationic, pegylated) to incorporate GdDTPA1 and ZnPc. 4. Experimental Procedures and Operational Workflow: Liposomes were prepared by thin-film hydration and extrusion. Size was measured over time with NTA. Thermal behavior was assessed by DSC. Singlet oxygen production was measured using DPBF quenching. Relaxivity was determined via NMR. Cytotoxicity was tested with MTT assay after 24h incubation and light exposure. Cellular localization was imaged with confocal microscopy after staining.
5:Data Analysis Methods:
Data were analyzed using standard fitting for NMR relaxation times, statistical analysis for cytotoxicity (triplicates with standard deviation), and qualitative assessment for microscopy images.
