  • Turbidimetric inhibition immunoassay revisited to enhance its sensitivity via an optofluidic laser

    摘要: Turbidimetric inhibition immunoassay (TIIA) is a classic immunodiagnostic method that has been extensively used for biomarker detection. However, the low sensitivity of this technique hinders its applications in the early diagnosis of diseases. Here, a new concept, optofluidic laser TIIA (OFL-TIIA), is proposed and demonstrated for sensitive protein detection. In contrast to the immunoreaction in traditional TIIA, in which the single-pass laser loss is detected, the immunoreaction in the OFL-TIIA method takes place in a laser cavity, which considerably increases the loss induced by antigen-antibody complexes (AACs) via the amplification effect of the laser. A commercial IgG TIIA kit was selected as a demonstrative model to characterize the performance of OFL-TIIA. A wide dynamic range of five orders of magnitude with an exceptional limit of detection (LOD) (1.8×10-10 g/L) was achieved. OFL-TIIA is a fast, sensitive, and low-cost immunoassay with a simple homogeneous and wash-free process and low-volume sample consumption, thus providing a new detection platform for disease diagnostics.

  • Hierarchical Core-Shell Structure of 2D VS2@VC@N-Doped Carbon Sheets Decorated by Ultrafine Pd Nanoparticles: Assembled in 3D Rosette-like Array on Carbon Fiber Microelectrode for Electrochemical Sensing

    摘要: The development of two-dimension (2D) nanohybrid materials with heterogeneous component in nanoscale and three-dimension (3D) well-ordered assembly in microscale has been regarded as an effective way to improve their overall performances by synergistic coupling of the optimized structure and composition. In this work, we reported the design and synthesis of a new type of hierarchically core-shell structure of 2D VS2@VC@N-doped carbon (NC) sheets decorated by ultrafine Pd nanoparticles (PdNPs), which were vertically grown on carbon fiber (CF) and assembled into a unique 3D rosette-like array. The resultant VS2@VC@NC-PdNPs modified CF microelectrode integrated the structural and electrochemical properties of the heterogeneous hybridization of core-shell VS2@VC@NC-PdNPs sheets with unique rosette-like array structure, and gave rise to a significant improvement in terms of electron transfer ability, electrocatalytic activity, stability and biocompatibility. Under the optimized conditions, the VS2@VC@NC-PdNPs modified CF microelectrode demonstrated excellent electrochemical sensing performance towards biomarker hydrogen peroxide (H2O2) including high sensitivity of 152.7 μA cm-2 mM-1, low detection limit of 50 nM (a signal-to-noise ratio of 3:1), as well as good reproducibility and anti-interference ability, which could be used for real-time in situ electrochemical detection of H2O2 in live cancer cells and cancer tissue. The remarkable performances of the proposed nanohybrid microelectrode will have a profound impact on the design of diverse 2D layered materials as promising candidate for electrochemical biosensing applications.

  • Diagnostic Biosensors in Medicine- a Review

    摘要: Biosensors consist of a biocatalyst that can detect a biological element and a transducer which can convert the combination event of the biocatalyst and the biological element into a detectable parameter. The biocatalyst can be biomolecules like, enzyme, DNA, RNA, metabolites, cells, oligonucleotides etc., and the transducers can be electrochemical, optical, piezoelectric, acoustics, calorimetric etc. In disease diagnostics biosensors utilizing immobilized cells, enzymes, nucleic acids have come into the field in recent years. Nanobiosensors exploiting the ultrasmall size and unique properties have also been used for engineering disease diagnostic biosensors. The use of biosensors can rapidly assess the health status, onset of the disease and its progression and can help to plan treatment for many diseases with the aid of multidisciplinary combination of chemistry, medical science and nanotechnology. The devices are cost effective, highly sensitive, rapid, user friendly and can be produced in bulk for human use. This review focuses on the different biosensors for the diagnosis of three major diseases like diabetes, cardiovascular disease and cancer.

    摘要: This article describes the fabrication and characterization of aluminium interdigitated electrode hybrid with ZnO for cardiac troponin T biomarker detection. The device was modified with one type of probe for the detection of cardiac troponin T (cTnT) biomarker. The fabrication process involved the deposition of ZnO thin film on the aluminium interdigitated electrode (Al IDE) using sol-gel method. The device was then characterized using scanning electron microscope (SEM), atomic force microscope (AFM), and 3D surface (Nano) profilometer. The electrical characterization was performed using current-voltage (I-V) measurement. The results showed that the device was able to detect cTnT biomarker with high sensitivity and selectivity.
