  • Ultrasensitive U-shaped fiber optic LSPR cytosensing for label-free and in situ evaluation of cell surface N-glycan expression

    摘要: Label-free and in situ detection of N-glycan expression on surface of cells is vital for understanding the progression of cancer. Herein, cytosensing based on U-shaped fiber optic local surface plasmon resonance (LSPR) was developed for cancer cell detection and N-glycan evaluation on cell surface. The U-shaped fiber optic LSPR has high refractive index sensitivity (RIS). Therefore, U-shaped fiber optic LSPR cytosensing afforded ultrasensitivity for cancer cell detection with the limit of detection (LOD) of 30 cells/mL and good linearity in a wide range of 1×102-1×106 cells/mL under the optimal conditions of modified AuNPs size and Con A concentration. The U-shaped fiber optic LSPR cytosensing shows more than 29 times lower limit of detection than the straight ones. More importantly, the prepared U-shaped cytosensing was applied to evaluate the N-glycan expression level on the surface of cancer cells under different concentration external stimuli of inhibitor tunicamycin (TM). Noticeably, U-shaped fiber optic LSPR cytosensing showed an acceptable reproducibility, satisfactory anti-interference and good selectivity, and was also applied in the assessment of N-glycan expression on surface of six kinds of cell lines. Therefore, the U-shaped fiber optic LSPR offers a feasible tool in biophysical research and clinical diagnosis for cancer.

  • Optimized light delivery probe using ball lenses for co-registered photoacoustic and ultrasound endo-cavity subsurface imaging

    摘要: An optimized hand-held photoacoustic and ultrasound probe suitable for endo-cavity tumor subsurface imaging was designed and evaluated. Compared to previous designs, the prototype probe, consisting of four 1 mm multi-mode optical fibers attached with 1.5 mm diameter ball-shaped fiber tips sandwiched between a transvaginal ultrasound transducer and a custom-made sheath, demonstrated a higher light output and better beam homogeneity on tissue subsurface. The output power and fluence profile were simulated for different design parameters. A camera recorded fluence profiles through calibrated intralipid solution at various imaging depths. The light delivery efficiency was experimentally compared with and without the ball lenses, based on ex-vivo imaging of human colorectal cancer and in-vivo imaging of a palmar vein proximal to the human wrist. The simulations and experiments demonstrated that ball-shaped fiber tip design can achieve homogeneous fluence distribution on tissue subsurface with acceptable light output efficiency, suggesting its clinical potential for in-vivo endo-cavity imaging.

  • Functionalized PdAu Alloy on Nanocones Fabricated on Optical Fibers for Hydrogen Sensing

    摘要: This paper reports the development of fiber optical hydrogen sensors using palladium and gold alloy nanostructures as sensor materials for hydrogen measurements using a D-shaped fiber as a platform. Using a maskless reactive ion etching technique, silica nanocone structures were formed on the surface of the D-shaped fiber. Palladium and gold alloys were deposited on the surface of nanostructured D-shaped fiber to form a nano-alloy sensor film. Evanescent interaction between guided light propagated in the fiber core and nano-alloy enabled highly sensitive hydrogen detection from concentrations that ranged from 0.25% to 10% in atmosphere pressure. The formation of nanostructured alloy enabled by the nanocone surface led to more than 3 times faster in sensor response time and significant improvements in sensor sensitivity and reversibility. The work presented here demonstrates that highly controllable VLSI microfabrication schemes can be applied to produce nanostructured sensor films on optical fibers for high-sensitivity chemical sensing.

  • All-optical modulator based on reduced graphene oxide coated D-shaped fiber waveguide

    摘要: An all-optical modulator of reduced graphene oxide coated D-shaped fiber structure, utilizing a convenient and controllable evanescent-field-induced deposition method, was designed in this study. By using the pump pulse at 980 nm, the signal light at 1550 nm was modulated with the rate of 50 kHz, and the maximum modulation depth of 8 dB was obtained using this device. Furthermore, this kind of modulator features strong mechanical stability, ease of fabrication, low cost and compatibility with optical fiber systems, which implies great potential in photonics applications such as all-optical switching and all-optical communication.
