  • Automated Quantification of Extranuclear ERα using Phosphor-integrated Dots for Predicting Endocrine Therapy Resistance in HR+/HER2? Breast Cancer

    摘要: In addition to genomic signaling, Estrogen receptor alpha (ERα) is associated with cell proliferation and survival through extranuclear signaling contributing to endocrine therapy (ET) resistance. However, the relationship between extranuclear ERα and ET resistance has not been extensively studied. We sought to measure extranuclear ERα expression by immunohistochemistry using phosphor-integrated dots (IHC-PIDs) and to assess its predictive value for ET resistance. After quantitative detection of ERα by IHC-PIDs in vitro, we developed “the nearest-neighbor method” to calculate the extranuclear ERα. Furthermore, tissue sections from 65 patients with HR+/HER2- BC were examined by IHC-PIDs, and the total ERα, nuclear ERα, extranuclear ERα PIDs score, and ratio of extranuclear-to-nuclear ERα (ENR) were measured using the novel method. We demonstrate that quanti?cation of ERα using IHC-PIDs exhibited strong correlations to real-time qRT-PCR (r2 = 0.94) and ?ow cytometry (r2 = 0.98). High ERα ENR was signi?cantly associated with poor overall survival (p = 0.048) and disease-free survival (DFS) (p = 0.007). Multivariate analysis revealed that the ERα ENR was an independent prognostic factor for DFS [hazard ratio, 3.8; 95% CI, 1.4–11.8; p = 0.006]. Our automated measurement has high accuracy to localize and assess extranuclear ERα. A high ERα ENR in HR+/HER2- BC indicates decreased likelihood of bene?ting from ET.

  • Determination of 17β-estradiol by surface-enhanced Raman spectroscopy merged with hybridization chain reaction amplification on Au@Ag core-shell nanoparticles

    摘要: The authors describe an aptamer-based assay for 17β-estradiol. It relies on the combined use of surface enhanced Raman scattering (SERS) and hybridization chain reaction (HCR). The aptamer against 17β-estradiol is applied as the recognition probes, and this results in excellent specificity. Specific recognition of target 17β-estradiol induce the freedom of DNA 2, which will open the stem-loop structure of probe 1 on the Au@Ag and form the partial dsDNA structure. With the nicking enzyme, the partial dsDNA will be hydrolyzed and the reside ssDNA on Au@Ag will form a small stem-loop structure. With the help of the other probe 2 modified Au@Ag and pre-immobilized probe 3 on the well of the microplate, an enzyme-free HCR can occur and tremendous Au@Ag can be assembled along the formed dsDNA in HCR, which can act as the excellent substrate for Raman measurement and greatly amplify the Raman signal of R6G on the Au@Ag. Afterwards, the key factor, ratio between probe 2-Au@Ag (P2) and probe1-Au@Ag (P1), affects the detection sensitivity is systematically optimized for the best sensing performance. The SERS signal of R6G, best measured at 1651 cm?1, increases linearly in the wide range from 1 pM to 10 nM. The detection limit can be as low as 0.1 pM.

  • Core-shell hollow spheres of type C@MoS2 for use in surface-assisted laser desorption/ionization time of flight mass spectrometry of small molecules

    摘要: Mesoporous carbon hollow spheres coated with MoS2 (C@MoS2) were synthesized to obtain a material with large specific surface area, fast electron transfer efficiency and good water dispersibility. The composite material was applied as a matrix for the analysis of small molecules by surface-assisted laser desorption/ionization time-of-flight mass spectrometry (SALDI-TOF MS). The use of a core-shell C@MoS2 matrix strongly reduces matrix background interferences and increases signal intensity in the analysis of sulfonamides antibiotics (SAs), cationic dyes, emodin, as well as estrogen and amino acids. The composite material was applied to the SALDI-TOF MS analysis of selected molecules in (spiked) real samples. The ionization mechanism of the core-shell C@MoS2 as a matrix is discussed. The method exhibits low fragmentation interference, excellent ionization efficiency, high reproducibility and satisfactory salt tolerance.

  • Measuring Estrogen Receptor Functionality using Progesterone Receptor PET Imaging: Rising to the (Estradiol) Challenge!

    摘要: Prognosis and therapy choice for breast cancer greatly depend upon pathohistological evaluation of biopsies and surgical samples. More specifically, the Estrogen Receptor (ER) – Progesterone Receptor (PR) – human epidermal growth factor receptor 2 (HER2) triad provides crucial information on which clinicians guide their intervention. Indeed, positivity for one or several of those receptors indicates that the use of hormone- or immuno-therapy against their respective targets is likely to benefit the afflicted patient.
