  • Au nanoparticle preconcentration coupled with CE-electrochemiluminescence detection for sensitive analysis of fluoroquinolones in European eel ( <i>Anguilla anguilla</i> )

    摘要: In this work, a novel method based on gold nanoparticle preconcentration coupled with CE for electrochemiluminescent detection of ciprofloxacin, enrofloxacin, ofloxacin, and norfloxacin in European eels was developed. The addition of gold nanoparticles induced the rapid enrichment of fluoroquinolones, which was simpler than the conventional enrichment approaches such as solid phase extraction and solid-phase microextraction. More than 100 times enrichment was observed after gold nanoparticle aggregation-based preconcentration. The CE-electrochemiluminescent parameters that affected the separation and detection were optimized. Under the optimized conditions, the linear ranges for the four fluoroquinolones were 0.090—8.0 μmol/L with the detection limits between 0.020 to 0.050 μmol/L. The proposed approach showed the advantages of high sensitivity, high selectivity, wide linear range, and low detection limit. It was used to analyze fluoroquinolones in European eel, and the results showed that the developed method can satisfy the detection requirements for fluoroquinolones determination in aquatic products set by China and European Union.

  • Rapid screening detection of fluoroquinolone residues in milk based on turn-on fluorescence of terbium coordination polymer nanosheets

    摘要: We report a facile, effective and rapid screening method for the determination of total fluoroquinolones (FQs) in milk using lanthanide coordination polymer nanomaterials as the sensing platform. The non-luminescent coordination polymer nanosheets (CPNSs) used in this work are composed of adenosine monophosphate (AMP) and terbium (Tb3+) ions (AMP/Tb CPNSs). The as-prepared CPNSs were characterized by transmission electron microscopy, Fourier transform infrared spectroscopy and fluorescence techniques. Tb3+ acts as the center ion of the coordination polymer, with AMP assembled through phosphate and amine groups. However, the reaction between FQs and AMP/Tb CPNSs noticeably turns on the fluorescence due to the strong coordination interaction between FQs and Tb3+. Under optimized conditions, the fluorescence intensity of AMP/Tb at 545 nm shows a linear relationship with FQ concentrations between 0.04 μmol/L and 5.0 μmol/L. The relative standard deviation (RSD) of the intraday precision is below 1 %, and detection limits are as low as 0.01 μmol/L. The proposed method was successfully applied to the determination of FQs in milk samples.

  • Silver nanoprisms based plasmonic ELISA for sensitive detection of fluoroquinolones

    摘要: Fluoroquinolones are synthetic antibiotics that are commonly used in animal husbandry, and the consumption of animal products with fluoroquinolone residues has imposed serious threat to human health. Here, we report a plasmonic enzyme-linked immunosorbent assay (pELISA) method based on oxidative etching of silver nanoprism (AgNPR) for the quantitative and qualitative detection of danofloxacin (DAN), a fluoroquinolone antibiotic. AgNPRs that undergo colorimetric changes upon oxidative etching by H2O2 serve as the signal transducer in our design. The indirect competitive pELISA was constructed by introducing biotinylated monoclonal antibody (mAb), streptavidin and biotinylated glucose oxidase, which catalyzes the generation of H2O2 for etching AgNPRs. The quantitative detection limit of the proposed method was 0.24 ng/mL for DAN. The qualitative detection limit for DAN reached 0.32 ng/mL, which was 32-fold lower than that of the assay using 3, 3’, 5, 5’-Tetramethylbenzidine (TMB) as the signal transducer. The average recoveries of DAN in milk ranged from 103% to 121%, with a coefficient of variation of 0.6% - 3.41%. The recovery results were further confirmed using liquid chromatography-tandem mass spectrometry. In summary, the proposed AgNPR etching pELISA produces high sensitivity, good accuracy and excellent reliability for the quantitative and qualitative detection of DAN in milk.
