摘要: To ensure the cost-competitiveness of Photovoltaic (PV) with integrated Battery Energy Storage Systems (PV-BESS), a highly reliable operation is demanded. The PV inverter has been reported as one of the reliability-critical components in the system, whose failures can lead to a severe negative impact on the cost of PV energy. Therefore, improving PV inverter reliability has high potential for the cost reduction of the PV-BESS. This paper thus investigates the impact of battery operation on the PV inverter reliability. A 6-kW residential PV-BESS in Germany is considered, where a self-consumption scheme is adopted. The evaluation results reveal that the reliability of the PV inverter is strongly affected by the battery operation. More specifically, the PV inverter loading is decreased significantly during the battery charging, which results in an improvement in the PV inverter reliability. In addition, the battery system parameters, such as battery capacity and battery converter power rating, also affect the PV inverter loading, whose impact on the PV inverter reliability has been analyzed.

[Methods in Molecular Biology] BCL-2 Family Proteins Volume 1877 (Methods and Protocols) || Rapid Imaging of BCL-2 Family Interactions in Live Cells Using FLIM-FRET
摘要: The Bcl-2 proteins control cell death via interchanging interactions within the Bcl-2 family. Fluorescence lifetime imaging microscopy (FLIM) is used to detect F?rster resonance energy transfer (FRET) between two fluorescent-fusion proteins in live cells. FLIM-FRET has been used to detect specific interactions and their disruption, for Bcl-2 family proteins. To date, this has been possible only in low throughput, due to the time required for serial data acquisition. We developed an automated optical system with eight parallel detectors for rapid and efficient data collection. Here we describe how to use this system for FLIM-FRET imaging of Bcl-2 family protein interactions in a 384-well plate format.

Label‐Free Metabolic Classification of Single Cells in Droplets Using the Phasor Approach to Fluorescence Lifetime Imaging Microscopy
摘要: Characterization of single cell metabolism is imperative for understanding subcellular functional and biochemical changes associated with healthy tissue development and the progression of numerous diseases. However, single-cell analysis often requires the use of ?uorescent tags and cell lysis followed by genomic pro?ling to identify the cellular heterogeneity. Identifying individual cells in a noninvasive and label-free manner is crucial for the detection of energy metabolism which will discriminate cell types and most importantly critical for maintaining cell viability for further analysis. Here, we have developed a robust assay using the droplet micro?uidic technology together with the phasor approach to ?uorescence lifetime imaging microscopy to study cell heterogeneity within and among the leukemia cell lines (K-562 and Jurkat). We have extended these techniques to characterize metabolic differences between proliferating and quiescent cells—a critical step toward label-free single cancer cell dormancy research. The result suggests a droplet-based noninvasive and label-free method to distinguish individual cells based on their metabolic states, which could be used as an upstream phenotypic platform to correlate with genomic statistics.

摘要: Multilevel inverters (MIs) have become widely used in photovoltaic (PV) systems for their high efficiency, reduced leakage currents, and reduced EMI levels. Although MIs suffer from unequal thermal stresses in power devices that lead to aging of power device with their continuous operation. Therefore, this paper proposes a new aging mitigation control (AMC) method to enhance the reliability of MIs against the aging effects in power devices. The operation of the proposed AMC method has three operating modes that are adapted according to the level and type of aging. In small and medium aging levels, the proposed method preserves the full output power ratings of the inverter with maximum energy extraction from PV modules. Whereas, in high aging level region and multiple devices failures, the proposed method reduces the output power so as to maintain continuous operation of the inverter without harmful consequences. The simulation and experimental results with comparisons to conventional methods show effective performance of the proposed AMC method.

摘要: Recently, introducing grid-tied PV systems is recommended in many countries. The inverter lifetime in PV applications is affected by their operating conditions. That is affected by the thermal loading of the power devices lifetime of electrolytic capacitors. Especially, the electrolytic capacitors at the DC-link to compensate the power ripple decreases when the ambient temperature increases. To improve the lifetime, a micro-inverter with an active buffer at the DC-link is introduced in this paper. In this micro-inverter, the DC-link electrolytic capacitor is replaced with a ceramic capacitor that has small capacitance. However, the reliability performance of the entire micro-inverter has not been evaluated. This paper thus assesses the lifetime of the micro-inverter considering the operational conditions (i.e., ambient temperature and solar irradiance, also referred to as mission profiles). A comparison between the proposed circuit with an active buffer and the conventional circuit based on a boost chopper is performed in this paper. It is revealed that the lifetime of the micro-inverter is very long. This is due to the over-designed power devices.
