  • Fine Fabrication and Optical Waveguide Characteristics of Hexagonal tris(8-hydroxyquinoline)aluminum(a?¢) (Alq3) Crystal

    摘要: Herein, we reported on the precise growth and optical waveguide characteristics of hexagonal tris(8-hydroxyquinoline)aluminum(III) (Alq3) micro-crystals (MCs). The hexagonal Alq3 MCs were prepared using surfactant-assisted assembly growth with the help of cetyltrimethylammoniumbromide (CTAB), in which the crystallization occurred as a result of molecular assembly and packing. Also, we adjusted the molar ratio of Alq3 and CTAB for the control degree of crystallization. The formation and structure of Alq3 MCs were investigated using field-emission scanning electron microscopy and X-ray diffraction pattern experiments, respectively. The solid-state laser confocal microscope-photoluminescence spectra and charge-coupled device images for the Alq3 MCs were measured to study the luminescence efficiency and colors, respectively. The optical waveguide performance of the hexagonal Alq3 MCs was measured for each side direction. According to our results, crystalline Alq3 micro-crystals are promising materials for application to the development of optical communication devices.

  • High tension electrocution death: New histopathological cardiac tools by Confocal Laser Scanning Microscope

    摘要: In deaths due to electrocution intraepidermal separation, vacuolation of epidermal cells, “swiss cheese” aspect of the superficial part of epidermis (swollen and with multiple vescicles), metallization, necrosis of collagenous fibers, cardiomyocytes alterations are microscopically described. No cardiac nerve damage due to electricity is actually reported in literature. In this work we tried to find new morphological signs in the hearts of deaths due to electrocution. In three cases of deaths due to electrocution, in which forensic autopsies were performed within 36 h of the death, heart specimens were taken at the level of common trunk of the left coronary artery. The myocardium histological examination at optical microscope and Confocal Laser Scanning Microscope (CLSM) revealed fragmentation of cardiomyocytes, nerve trunks damage with hydropic swelling of the nerve fibers, interstitial and sub-nerve-sheath edema, very dishomogenous distribution of the natural fluorescence of the neurofilaments, coarctation of epicardial gangliar cells with cytoplasmic cleft and irregular fluorescence pattern. Identification of S-100 protein by immunohistochemistry can help to better observe the hydropic swelling of the nerve fibers and the central cytoplasmic clefts. These alterations could be used in future as specific signs of the passage of the electrical current through the heart. We recommend heart specimens at the level of common trunk of the left coronary artery in all the cases of suspected electrocution, to better evaluate cardiac nerve trunks damages and alterations.

  • Color Reproduction Accuracy Promotion of 3D-Printed Surfaces Based on Microscopic Image Analysis

    摘要: Full-color 3D printing technology is a powerful process to manufacture intelligent customized colorful objects with improved surface qualities; however, poor surface color optimization methods are the main impeding factors for its commercialization. As such, the paper explored the correlation between microstructure and color reproduction, then an assessment and prediction method of color optimization based on microscopic image analysis was proposed. The experimental models were divided into 24-color plates and 4-color cubes printed by ProJet 860 3D printer, then impregnated according to preset parameters, at last measured by a spectrophotometer and observed using both a digital microscope and a scanning electron microscope. The results revealed that the samples manifested higher saturation and smaller chromatic aberration (ΔE) after post-processing. Moreover, the brightness of the same color surface increased with the increasing soaked surface roughness. Further, reduction in surface roughness, impregnation into surface pores, and enhancement of coating transparency effectively improved the accuracy of color reproduction, which could be verified by the measured values. Finally, the chromatic aberration caused by positioning errors on different faces of the samples was optimized, and the value of ΔE for a black cube was reduced from 8.12 to 0.82, which is undetectable to human eyes.

  • Alignment of liquid crystals using Langmuir?Blodgett films of unsymmetrical bent-core liquid crystals

    摘要: The properties of the thin films of liquid crystal (LC) molecules can be governed easily by external fields. The anisotropic structure of the LC molecules has a large impact on the electrical and optical properties of the film. The Langmuir monolayer (LM) of LC molecules at the air–water interface is known to exhibit a variety of surface phases which can be transferred onto a solid substrate using the Langmuir?Blodgett (LB) technique. Here, we have studied the LM and LB films of asymmetrically substituted bent-core LC molecules. The morphology of LB film of the molecules is found to be a controlling parameter for aligning bulk LC in the nematic phase. It was found that the LB films of the bent-core molecules possessing defects favour the planar orientation of nematic LC, whereas the LB films with fewer defects show homeotropic alignment. The defect in LB films may introduce splay or bend distortions in the nematic near the alignment layer which can govern the planar alignment of the bulk LC. The uniform layer of LB film facilitates the molecules of nematic to anchor vertically due to a strong van der Waals interaction between the aliphatic chains leading to a homeotropic alignment.

  • Redox-state dependent blinking of single photosystem I trimers at around liquid-nitrogen temperature

    摘要: Efficient light harvesting in a photosynthetic antenna system is disturbed by a ragged and fluctuating energy landscape of the antenna pigments in response to the conformation dynamics of the protein. This situation is especially pronounced in Photosystem I (PSI) containing red shifted chlorophylls (red Chls) with the excitation energy much lower than the primary donor. The present study was conducted to clarify light-harvesting dynamics of PSI isolated from Synechocystis sp. PCC6803 by using single-molecule spectroscopy at liquid?nitrogen temperatures. Fluorescence emission at around 720 nm from the red Chls in single PSI trimers was monitored at 80–100 K. Intermittent variations in the emission intensities, so-called blinking, were frequently observed. Its time scale lay in several tens of seconds. The blinking amplitude depended on the redox state of the phylloquinone (A1). Electrochromic shifts of Chls induced by the negative charge on A1 were calculated based on the X-ray crystallographic structure. A Chl molecule, Chl-A839 (numbering according to PDB 5OY0), bound near A1 was found to have a large electrochromic shift. This Chl has strong exciton coupling with neighboring Chl (A838) whose site energy was predicted to be determined by interaction with an arginine residue (ArgF84) [Adolphs et al., 2010]. A possible scenario of the blinking was proposed. Conformational fluctuations of ArgF84 seesaw the excitation-energy of Chl-A838, which perturbs the branching ratio of excitation-energy between the red Chl and the cationic form of P700 as a quencher. The electrochromic shift of Chl-A839 enhances the effect of the conformation dynamics of ArgF84.

  • Advanced methods for the optical quality assurance of silicon sensors

    摘要: We describe a setup for optical quality assurance of silicon microstrip sensors. Pattern recognition algorithms were developed to analyze microscopic scans of the sensors for defects. It is shown that the software has a recognition and classification rate of >90% for defects like scratches, shorts, broken metal lines etc. We have demonstrated that advanced image processing based on neural network techniques is able to further improve the recognition and defect classification rate.

  • A miniature multi-contrast microscope for functional imaging in freely behaving animals

    摘要: Neurovascular coupling, cerebrovascular remodeling and hemodynamic changes are critical to brain function, and dysregulated in neuropathologies such as brain tumors. Interrogating these phenomena in freely behaving animals requires a portable microscope with multiple optical contrast mechanisms. Therefore, we developed a miniaturized microscope with: a fluorescence (FL) channel for imaging neural activity (e.g., GCaMP) or fluorescent cancer cells (e.g., 9L-GFP); an intrinsic optical signal (IOS) channel for imaging hemoglobin absorption (i.e., cerebral blood volume); and a laser speckle contrast (LSC) channel for imaging perfusion (i.e., cerebral blood flow). Following extensive validation, we demonstrate the microscope’s capabilities via experiments in unanesthetized murine brains that include: (i) multi-contrast imaging of neurovascular changes following auditory stimulation; (ii) wide-area tonotopic mapping; (iii) EEG-synchronized imaging during anesthesia recovery; and (iv) microvascular connectivity mapping over the life-cycle of a brain tumor. This affordable, flexible, plug-and-play microscope heralds a new era in functional imaging of freely behaving animals.

    摘要: This paper presents a method for the determination of optimal curing conditions for the imaging of single lung cancer cells by atomic force acoustic microscope (AFAM). The cellular morphology, height and surface roughness of the cells treated with different concentrations of paraformaldehyde and methanol and durations were observed using an AFAM. The experimental results showed that the A549 cells solidified with 4% paraformaldehyde for the period from 30min to 60min were close to the profiles of living cells. The cells solidified with 4% paraformaldehyde for 10min were beneficial to obtain subsurface structures.

    摘要: Nanomanipulation system based on scanning electron microscope(SEM) with good real-time visual feedback and nanoscale observation resolution had high operability in a vacuum working environment. Different nanomanipulation tasks of carbon nanotubes (CNTs) are carried out through the nanomanipulation system in SEM. Nanomanipulation platform existed inherent manufacture errors, installation errors and other errors, and imprecise nanomanipulation system were also time-consuming and laborious for operators. This paper presentes a method of combining the visual feedback and feedforward control to detect and compensate the motion error of the multi-dimensional SmarAct nanomanipulation platform in the nanomanipulation system in SEM. This method reduces the motion error in the X-Y direction and achieved higher operating accuracy. At the different step speed, the motion error in the X direction and Y direction is 135.7nm and 112.9nm respectively. After the feedforward compensation, the motion error in the X direction and Y direction reduces to 61.3nm and 54.1nm respectively.

    摘要: The monitoring of Escherichia coli (E. coli) bacteria in food and water is an effective preventive measure in controlling the outbreaks of serious food-related diseases. Moreover, the accurate detection of its early stage of the bacteria can ensure the public health and safety. In this study, a target-selective aptamer-based platform is proposed as a reliable procedure for the detection of the E. coli in Phosphate-buffered saline (PBS) using fluorescent microscopy. The results show that the aptamer sequences, immobilized on an array of Au-micro-squares, are very high selective towards the target bacteria cells. This platform, presented for the first time, can be potentially used to detect different kinds of cells simultaneously. Also, an important cell-counting application, based on Neubauer Lam high accuracy method, is proposed and implemented and validated by experimental results.
