  • Preparation of a star-shaped copolymer with porphyrin core and four PNIPAM-b-POEGMA arms for photodynamic therapy

    摘要: A series of thermosensitive star-shaped copolymers THPP-(PNIPAM-b-POEGMA)4 with 5, 10, 15,20-tetrakis-(4-hydroxyphenyl)-21H,23H-porphyrin (THPP) core and four poly(N-isopropylacrylamide)-b-poly(oligo(ethylene glycol) mono methyl ether methacrylate) (PNIPAM-b-POEGMA) arms were synthesized via reversible addition-fragmentation chain transfer (RAFT) polymerization. The low critical solution temperatures (LCSTs) of THPP-(PNIPAM-b-POEGMA)4 with different molecular weights were 37.5 °C, 39.9 °C and 41.9 °C respectively, which depending on the hydrophilic POEGMA contents of copolymers. The micellar structures of copolymers could be formed above the LCSTs. The MTT study indicated that THPP-(PNIPAM-b-POEGMA)4 and THPP showed no significant cytotoxicity toward HeLa cells and L929 cells. And THPP-(PNIPAM-b-POEGMA)4 under light irradiation showed photodynamic activity which photo-toxicity toward HeLa cells was stronger than THPP.

  • Synthesis of Temperature and Light Sensitive Copolymers with Controlled Aggregation during Phase Transitions

    摘要: Two thermosensitive copolymers of acrylates (PMP/PEP) and a thermo/light sensitive copolymer of acrylate with acrylamide (PMABz) are synthesized via a ring-opening reaction and subsequent radical polymerization using a facile one-pot method. The lower critical solution temperature (LCST) of PMP and PEP shows a dependence on the content of the structural unit and can be easily tuned to 18–35 °C by mercaptoethanol-mediated control of the molecular weight. The isomerization of PMABz can be reversibly induced by alternating irradiation at 365 nm to produce the cis form and at 254 nm to produce the trans form. The LCST of cis-PMABz (40.3 °C) is significantly higher than that of trans-PMABz (37.8 °C) because of its higher hydrophilicity. Furthermore, the particle size of aggregated PMABz can be controlled from 665 nm (for the cis form) to 996 nm (for the trans form) during the heating run. After quenching the PMABz solution, PMABz particles have repeating patterns, such as leaf veins or hollow containers, indicating their potential applicability.

  • Click synthesis of glycosylated porphyrin-cored PAMAM dendrimers with specific recognition and thermosensitivity

    摘要: Glycoporphyrin dendrimers have received increasing attentions due to their distinct physical and chemistry properties, such as fluorescence and water solubility. They also acted as a promising photodynamic therapy (PDT) sensitizer and used in the study of carbohydrate-protein interaction. In this study, different generations of porphyrin-cored dendrimers consisting of glycosylated siloxane-poly (amido amine) typed dendron (PP-Si-PAMAM-Dm-Lac, m = 1, 2, and 3), multifunctional specific-targeting sensitizers, had been synthesized via click reaction. The structure of PP-Si-PAMAM-Dm-Lac was characterized by 1H NMR, GPC, FT-IR, UV-vis and fluorescence analyses. This glycoporphyrin dendrimers, not only obtained high fluorescence quantum yield and singlet oxygen production efficiency, but also showed specific recognition of lectin and remarkable temperature-responsive property (20–80 °C). These properties proved a facile click synthetic strategy of glycosylated siloxane-poly (amido amine) typed dendrimers with multiple functions as a promising tumor-selective photsensitizer (PS) for PDT and also allow applications in highly sensitive fluorescent probes over a larger temperature window.
