  • A Fluorescent Probe with Aggregation-Induced Emission for Detecting Alkaline Phosphatase and Cell Imaging

    摘要: Alkaline phosphatase (ALP) is associated with many diseases, and its accurate detection is of great significance. Fluorescent compounds with aggregation-induced emission (AIE) feature show beneficial advantages for serving as fluorescent probes. Herein, an AIE-active “turn on” probe for ALP detection was synthesized through incorporating a strong electron-withdrawing group (cyano) in the middle and the recognition moiety phosphate group at the end, thereby rendering a D–A–D structure with a relatively high conjugation degree and good water solubility. It was found that the probe TPE-CN-pho is highly sensitive to ALP in aqueous solution. In the presence of ALP, the hydrophilic phosphate group on the probe is rapidly removed, resulting in a decrease in water solubility and subsequent formation of aggregates, thereby achieving aggregation-induced emission. Moreover, the probe TPE-CN-pho has also been successfully applied to imaging ALP in living cells.

  • Aggregation-Induced Emission (AIE) Polymeric Micelles for Imaging-Guided Photodynamic Cancer Therapy

    摘要: Photodynamic therapy (PDT) is a noninvasive treatment for selectively killing malignant tumor cells. The photosensitizer is a necessary component of photodynamic nanomedicine. Many efforts have been made to develop new photosensitizers for efficient cancer photodynamic therapy. In this work, we report a novel nano photosensitizer, polymeric micelles (AIE-M) with aggregation induced emission characteristic, for photodynamic cancer therapy. AIE-M with sub-20 nm particle size is prepared by the self-assembly of salicylaldazine-incorporated amphiphilic polymer (AIE-1), which can produce reactive oxygen species (ROS) with light irradiation in solution. After uptake by cancer cells, AIE-M can specially sojourn in plasma membranes of cancer cells at the early stage and predominantly accumulate in the mitochondria of cancer cell at the late stage. The phototoxicity of AIE-M, resulting from the generation of intracellular ROS with light irradiation, can efficiently cause cancer cells death by apoptosis and necrosis. The advantages of AIE-M as a nano photosensitizer include the small size, highly colloidal stability in the process of preparation and storage, and high cell penetration. The ultra-low Critical Micelle Concentration (CMC) of AIE-1, negligible dark toxicity and super phototoxicity of AIE-M suggest its promising potential for image-guided PDT.

  • Dual sites fluorescence probe for hydrogen sulfide: AIEE activity and supramolecular assembly with β-cyclodextrin

    摘要: One aggregation-induced emission enhancement (AIEE) active compound based on α–cyanostilbenes and maleic anhydride group were synthesized. The “Turn-On” fluorescence recognition for H2S was evaluated both in aqueous solution and cells. It was found that cyanostilbenes group acts as the first reactive site and maleimide group acts as the second site to H2S. Supramolecular assembly with β-cyclodextrin successfully revealed the reaction mechanism of 3-state model.

  • Preparation of silica/polymer nanocomposites with aggregation-induced emission properties as fluorescent responsive coatings

    摘要: Fluorescent materials in the recent past have found numerous novel applications in various fields such as medicine, environment, electronics and coatings. For smart coating applications, durability, responsiveness and repeatable functionalities are desirable especially in their solid state. However, due to the known aggregation-caused quench (ACQ) effect, many traditional fluorescent molecules’ intensity tends to diminish at aggregated state. This ultimately affects the traditional fluorescent materials’ use in these applications. To develop smart coatings with suitable properties and responsiveness, herein, a novel aggregation-induced emission (AIE)-based composite fluorescent latex was prepared and systematically studied. Briefly, the approach involved first grafting tetraphenylethene (TPE) derivant onto the silica sol. The resultant modified silica sol could then emit strong fluorescent light courtesy of TPE’s restricted intramolecular rotation. Further, the modified silica sol was used as the surface to emulsion polymerize acrylic latex, forming a fluorescent composite core-shell structure. As a smart and responsive fluorescent composite coating, when cured, it showed fluorescent responsiveness to the external stimuli, such as water and toluene vapor, demonstrating possible application in smart paints.

  • Triphenylethylene- and Tetraphenylethylene-Functionalized 1,3-Bis(pyrrol-2-yl)squaraine Dyes: Synthesis, Aggregation-Caused Quenching to Aggregation-Induced Emission, and Thiol Detection

    摘要: Three novel pairs of 1,3-bis(pyrrol-2-yl)squaraine dyes, N-alkylated SQ1a?1b, and N-phenylated SQ2a?2b in which triphenylethylene moieties functionalized at 5-position of pyrrole, as well as SQ3a?3b with tetraphenylethylene (TPE) moieties attached at N-position of pyrrole, were synthesized. All six dyes were found not to exhibit aggregation-induced emission (AIE) properties. Spectrophotometric studies showed that N-TPE-functionalized SQ3a?3b exhibited much larger molar extinction coefficients (ε: 1.36?2.14 × 105 M?1 cm?1) than 5,5′-triphenylethylene-functionalized SQ1a?2b (ε: 2.17?8.22 × 104 M?1 cm?1). Surprisingly, SQ2b showed a remarkable red-shifted maximum absorption (λmax: 723 vs 631?652 nm) compared to that of other squaraine dyes. All six squaraine dyes selectively responded to the addition of thiol-containing biomolecules, such as cysteine and gluthatione, with the disappearance of λmax in the near-infrared region in their respective absorption spectra. Interestingly, the thiolated species of SQ3a?3b were AIE active, with the characteristic AIE emission of TPE at λmax = 484?490 nm upon addition of water. Further thiol sensing on solid supports was examined, indicating the potential applications of TPE-functionalized squaraine dyes as bioprobes for the detection of important thiol-containing biomolecules, with a clear change from aggregation-caused quenching to AIE.

  • Rapid Detection of Aromatic Pollutants in Water Using Swellable Micelles of Fluorescent Polymers

    摘要: We constructed a kind of novel swellable fluorescent micelles for rapid and sensitive detection of toxic aromatic pollutants (APs) in water based on capture-report strategy. An amphiphilic triblock copolymer capped with an aggregation induced emission (AIE) chromophore self assembled into micelles with core-shell structures in aqueous solution. Hydrophobic segments of block polymers organized into cores with the AIE chromophore near the core/shell interface, which were stabilized by a corona of water-soluble polymer segments. Water-soluble polymer segments captured APs in water. The captured pollutants were subsequently transported to hydrophobic cores of micelles. The cores swelled after absorbing APs, leading to fluorescence quenching of the AIE chromophores. The fluorescent micelles allowed rapid detection of APs in the order of seconds at a concentration of 1 ug/L. Compared with commercial GC-MS, the fluorescent micelles require a much smaller amount sample and are much quicker with comparable sensitivity. With the merits of high sensitivity, rapid response, simple operation procedures, and low cost, the fluorescent micelles provide an ideal candidate for the facile detection of toxic aromatic pollutants in water.

  • Bis[1]benzothieno[1,4]thiazines - Planarity, Enhanced Redox Activity and Luminescence by Thieno-Expansion of Phenothiazine

    摘要: Twofold Buchwald-Hartwig aminations selectively furnish all three regioisomers of bis[1]benzothieno[1,4]thiazines and X-ray structure analyses and DFT calculations were corroborated for correlation of their electronic properties. All regioisomers outscore the parent compound phenothiazine with respect to a low lying oxidation potential and reversible redox activity. The anti-anti bis[1]benzothieno[3,2-b:2',3'-e][1,4]thiazines possess lowest oxidation potentials in this series and display pronounced green luminescence in solution (?F ≈ 20%) and in the solid state. Syn-anti regioisomers are only weakly luminescent in solution, but show aggregation induced emission enhancement and solid state luminescence. Most interestingly, found by X-ray structure analyses anti-anti derivatives reveal an amazingly coplanar structure of the pentacyclic anellated 1,4-thiazine system, emphasizing a structural similarity to heteroacenes. The calculated theoretical nucleus-independent chemical shifts additionally suggest that the 8?-electron core systems can be considered as first electronically unbiased anellated 1,4-thiazines with antiaromatic character.

  • AIEgen Nanoparticles of Arylamino Fumaronitrile Derivative with High Near-infrared emission for Two-photon Imaging and in vivo Cell Tracking

    摘要: Developing of two-photon materials for live-cell imaging and in vivo analysis in-deep have been received great attention, and it is still urgent so that such microscopy technique could be promoted and advanced into new progress using the powerful probs. Herein, a new arylamino fumaronitrile derivative NPAPF was synthesized and transferred as aggregation-induced emission luminogen (AIEgen) fluorescent nanoparticles (AF-NPs) via assembly technique. This AF-NPs exhibited two-photon absorption cross-section at 2.6×106 GM with 19.5% of fluorescence quantum yield. Moreover, utilizing the great potential of AF-NPs, two-photon imaging of live cells with good cytocompatibility is realized upon two-photon microscopy. By in vivo long-term tracing studies of mesenchymal stem cells, we demonstrated the tremendous advantage of AF-NPs tracer in monitoring the stem cells transplant. Therefore, our unique AF-NPs provided an efficient two-photon-absorbing probe for investigating biological mechanism and behavior, and also opened a new avenue for spatiotemporal visualization of transplanted stem cells.

  • Construction of a Layered Hydrogen-Bonded Organic Framework Showing High-Contrast Mechanoresponsive Luminescence Turn-On

    摘要: This paper probes the mechanoresponsive luminescence and aggregation-induced emission (AIE) properties of N-phenylcarbazole-based thiophene/dicyanovinyl (T?DCV) derivatives. In bis-T?DCV planar molecule 1, a layered hydrogen-bonded organic framework (HOF) structure is formed with the assistance of multiple C?H···N and strong interlayer interactions. The formation of such a layer packing HOF structure can quench the solid fluorescent emission when aggregated as nanosuspensions but guarantees the high-contrast luminescence on?off switching (1.2 × 103 fold) in the solid state. Moreover, the grinding-induced destruction of HOF 1 is reflected by powder X-ray diffraction measurements, in which the destruction of the α and β rings in the planar HOF stripe and the close interlayer stacking could be observed. This work achieves the successful construction of long-wavelength emission AIE molecules (2 and 3) by introducing sufficient steric hindrance rotors to disturb the intermolecular π?π stacking and/or dipole?dipole interactions.

  • Bio‐orthogonal AIE Dots Based on Polyyne‐Bridged Red‐emissive AIEgen for Tumor Metabolic Labeling and Targeted Imaging

    摘要: In this work, we aim to develop cancer cell-targeting AIE dots based on the polyyne-bridged red-emissive AIEgen, 2TPE-4E, through the combination of metabolic engineering and bio-orthogonal reactions. Azide groups on tumor were efficiently produced by intravenous injection of Ac4ManNAz and glycol-metabolic engineering. These bio-orthogonal azide groups could facilitate the specific targeting of DBCO-AIE dots to the tumor cells undergoing metal-free click reaction in vivo. The efficiency of this targeting strategy could be improved along with further development of new bio-orthogonal chemical groups with higher reactivity and sufficient AIEgens could be delivered to tumor for diagnosis.
