  • [Methods in Molecular Biology] Autoantibodies Volume 1901 (Methods and Protocols) || Determination of CRP Autoantibodies by SPR Immunoassay

    摘要: Biosensors based on the principle of surface plasmon resonance (SPR) are surface-sensitive optical devices used for monitoring biomolecular interactions at the sensor surface in real time without any labeling. It is used in a wide variety of areas including proteomics, clinical diagnosis, environmental monitoring, drug discovery, and food analysis. C-reactive protein (CRP) is a marker of in?ammation, which undergoes conformation changes in local lesions, leading to the formation of mCRP. Autoantibodies against mCRP are frequently detected in systemic lupus erythematosus (SLE) and associated with disease activity and prognosis. An SPR immunoassay for CRP autoantibodies at complement factor H–CRP interface is described in this chapter.

  • [Methods in Molecular Biology] Autoantibodies Volume 1901 (Methods and Protocols) || Detection of Autoantibodies to Complement Components by Surface Plasmon Resonance-Based Technology

    摘要: The innate immune complement system is a powerful defense cascade against pathogens, but can induce host tissue damage when overactivated. In pathological conditions, mainly but not restricted to renal diseases, such as lupus nephritis, atypical hemolytic uremic syndrome, and C3 glomerulopathies, complement is overactivated or dysregulated by autoantibodies directed against its components and regulators. Among the key autoantibody targets are the initiator of the classical complement pathway C1q, the alternative pathway regulator Factor H, the components of the alternative pathway C3 convertase complex C3 and Factor B and the convertase complex itself. This methodological article describes our experience with a method for detection of anti-complement autoantibodies in real time using surface plasmon resonance-based technology. It allows label-free evaluation of the binding efficacy and stability of the formed antigen–antibody complexes.

  • Complement inhibition as a therapeutic strategy in retinal disorders

    摘要: Introduction: Dry age-related macular degeneration (AMD) and Stargardt Macular Dystrophy (STGD1) result in vision loss, due to progressive atrophy of the macula and lack of effective treatments. Numerous studies have implicated complement-associated inflammation as a contributor to both diseases. Areas covered: The complement factor D inhibitor, lampalizumab, failed to halt geographic atrophy (GA) progression in phase 3 studies. The complement factor 3 (C3) inhibitor, APL-2, has shown potential to reduce GA growth in a phase 2 trial, supporting advancement to phase 3 trials. The intravenous complement factor 5 (C5) inhibitor, eculizumab, failed to halt GA progression in a phase 2 study. Another C5 inhibitor, avacincaptad pegol, is delivered by intravitreal injection, and will be studied for safety and preliminary signs of efficacy for AMD and STGD1 patients in phase 2 trials. LFG316 (C5 inhibitor) and CLG561 (properdin inhibitor) failed to halt GA progression in phase 2 studies. A phase 1 trial is evaluating the effects of combining LFG316 and CL561. Complement inhibition by gene therapy will be explored in the phase 1 trial of HMR59 in AMD patients. Expert opinion: While complement inhibition has not yet demonstrated ability to halt GA progression in a phase 3 trial, further study is warranted.

  • New insight into the role of the complement in the most common types of retinopathy-current literature review

    摘要: Pathological neovascularisation, which is a critical component of diseases such as age-related macular degeneration (AMD), diabetic retinopathy (DR) and retinopathy of prematurity (ROP), is a frequent cause of compromised vision or blindness. Researchers continuously investigate the role of the complement system in the pathogenesis of retinopathy. Studies have confirmed the role of factors H and I in the development of AMD, and factors H and B in the development of DR. Other components, such as C2, C3, and C5, have also been considered. However, findings on the involvement of the complement system in the pathogenesis of ROP are still inconclusive. This paper presents a review of the current literature data, pointing to the novel results and achievements from research into the role of complement components in the development of retinopathy. There is still a need to continue research in new directions, and to gather more detailed information about this problem which will be useful in the treatment of these diseases.
