  • A mitochondria targetable and viscosity sensitive fluorescent probe and its applications for distinguishing cancerous cells

    摘要: The determination of mitochondrial viscosity is of great importance owing to its crucial roles in the diffusion-mediated processes. It has demonstrated that the viscosity in cancer cells is higher than normal cells. Developing of viscosity sensitive fluorescent probes for distinguishing normal cells and cancer cells is necessary. Herein, we present a mitochondrial-targeting fluorescent probe PFV for the detection of viscosity in live cells. It exhibited outstanding sensitivity to viscosity, free from disturbing by ROS/sulfuret even at high concentrations. Equipped with mitochondria targeting ability, PFV was then applied to mitochondrial viscosity detection. Most importantly, by viscosity monitoring PFV successfully differentiated normal hepatic cells and cancerous hepatic cells. The present study would offer a broadly applicable for the determination of viscosity in complex systems.

  • The Impact of Photobleaching on Microarray Analysis

    摘要: DNA-Microarrays have become a potent technology for high-throughput analysis of genetic regulation. However, the wide dynamic range of signal intensities of fluorophore-based microarrays exceeds the dynamic range of a single array scan by far, thus limiting the key benefit of microarray technology: parallelization. The implementation of multi-scan techniques represents a promising approach to overcome these limitations. These techniques are, in turn, limited by the fluorophores’ susceptibility to photobleaching when exposed to the scanner’s laser light. In this paper the photobleaching characteristics of cyanine-3 and cyanine-5 as part of solid state DNA microarrays are studied. The effects of initial fluorophore intensity as well as laser scanner dependent variables such as the photomultiplier tube’s voltage on bleaching and imaging are investigated. The resulting data is used to develop a model capable of simulating the expected degree of signal intensity reduction caused by photobleaching for each fluorophore individually, allowing for the removal of photobleaching-induced, systematic bias in multi-scan procedures. Single-scan applications also benefit as they rely on pre-scans to determine the optimal scanner settings. These findings constitute a step towards standardization of microarray experiments and analysis and may help to increase the lab-to-lab comparability of microarray experiment results.

  • Effects of physical orientation of dye molecules and molecular orbitals on performance of solid-state dye sensitized solar cells

    摘要: Performance of Dye-sensitized devices depends on the photon absorption and carrier injection properties of the sensitizer (dye). The orientation of the dye molecule affects the photon absorption cross-section, injection efficiency and carrier transport. These effects are studied, using three variants of cyanine dyes in n-TiO2/Dye/p-CuSCN heterojunction. The results show correlation of dye-molecule’s orientation on the short-circuit-photocurrent (Isc). The open-circuit-voltage (Voc) is also subjective. The orientation of the dye molecule influence the photon-harvesting efficiency and obstruct the hole-conductor penetrating onto the working-electrode. Additionally, Cumulative effects of e-e, e-h, spin-coupling and HOMO/LUMO distribution are identified.

  • Aggregation States of Organic Dye Molecules in Organic – Inorganic Hybrid Films Studied by Near-Field and Far-Field Fluorescence Spectroscopy

    摘要: Near-field fluorescence spectroscopy in conjunction with far-field fluorescence and Raman spectroscopy has been applied to investigate the aggregation states of cyanine dye of 2-[5-(1,3-Dihydro-3,3-dimethyl-1-octadecyl-2H-indol-2-ylidene)-1,3-pentadienyl]-3,3-dimethyl-1-octadecyl-3H-indolium perchlorate (NK3175) molecules in the depth direction, from the nanometer-scale top surface to the bulk, in the hybrid films consisted of NK3175 and a clay compound (SWN). The far-field Raman spectra reveal that these hybrid films contained both NK3175 molecules which did not take part in the adsorption and those adsorbed on SWN. The near-field photoluminescence (PL) spectra in the outermost surface within ca. 30 nm exhibit a significant blue shift for the hybrid films as compared to their respective PL spectra in the bulk, implying that the top surface of these hybrid films becomes more polar and/or rigid in terms of the microenvironment around NK3175 molecules compared to the bulk of them, in contrast to the case for the other hybrid film which was prepared by the alternative method. These results provide new information on the aggregation states of organic dye molecules not only in the bulk but in the nanometer-scale top surface of organic-inorganic hybrid systems.

  • Targeted Methotrexate Prodrug Conjugated With Heptamethine Cyanine Dye Improving Chemotherapy and Monitoring Itself Activating by Dual-Modal Imaging

    摘要: Theranostic prodrug plays a vital role in reducing the side effects and evaluating the therapeutic efficiency of prodrug in vivo. In particular, small conjugate-based theranostic prodrugs have attracted much attention because of their clear and simple structures. In this work, we synthesized a novel tumor-targeting and glutathione-activated conjugate-based theranostic prodrug (Cy-SS-MTX). The prodrug was constructed by conjugating Cy (IR780) to methotrexate (MTX) via a disulfide bond. The Cy dye as targeting molecule bring prodrug to cancer cells and then the prodrug was activated by the high levels of glutathione in tumor. In cell experiments, the results showed the excellent ability of prodrug to target tumor. Meanwhile, the prodrug apparently improved the anti-tumor ability and hugely reduced toxicity of free MTX on normal cells. Furthermore, owing to intramolecular charge transfer between Cy and MTX, the Cy structure in the prodrug showed an absorption peak at 654 nm in UV-Vis spectroscopy. However, when the disulfide bond of prodrug was broken by glutathione, a new UV-Vis absorption peak at 802 nm of Cy structure in prodrug was arised. At the same time, the fluorescence (FL) emission peak at 750 nm (excitation at 640 nm) would turn into 808 nm (excitation at 745 nm). What’s more, the photoacoustic (PA) signal with excitation at 680 and 808 nm also changed. The experimental results in vivo showed that the prodrug has been successfully utilized for real-timely tracking MTX activation by FL and PA imaging upon near infrared laser excitation and cancer targeting therapy. Our studies further encourage application of small conjugate-based prodrug based on tumor-targeted heptamethine cyanine dye as reporter group for targeted therapy and real-timely tracking activation of drug.
