    摘要: Computational models are useful for quantitative elucidation of the dynamical behavior of biological systems. Oftentimes, several competing models (i.e., hypotheses) are proposed to describe the underlying molecular mechanisms of a biological system. Selecting the most representative model is imperative for obtaining meaningful quantitative insights into the dynamics of the system of interest. However, the discrimination between competing models poses a significant challenge due to heterogeneity that is intrinsic to biological systems. This letter demonstrates the effectiveness of a probabilistic approach to optimal experiment design for model discrimination in the presence of time-invariant cell-to-cell differences within a cell population. The JAK2/STAT5 signaling pathway, which is involved in proliferation and differentiation of hematopoietic stem cells, is used as a case study.

  • Optimisation of laser beam cutting parameters of high density composite fibre cement flat board

    摘要: At the beginning of the research work we encountered two major problems. First, literature for laser beam machining of fibre cement is virtually zero. This fact does not exclude the research of the subject by others, of course, but it may in any case indicate its confidentiality (technological competitive advantage). Second, the exact composition of the fibre cement material is protected, but the boards we use are available from manufacturer by code number, so the experiment can be clearly reproduced. For this reasons, we have decided to use the rigid, classic Design of Experiments method. Based on our previous experience, the effect factors were chosen in the group of Laser power, Cutting velocity, Cutting gas pressure, Focal length, Laser frequency and Cutting gas type, at a total of 17 factor levels. The necessary and sufficient conditions are met by the 2-Factor-Interaction (2FI) fractional factorial experiments, instead of full factorial 6FI experiment series. Based on our very first experiments, it was found that the high-density composite fiber cement material can be cut with laser beam technology. Experiments and related visual and measurement responses clearly define the significant effect factors. The experiments proved our zero hypotheses, the "p-values", derived from the analysis of the response parameters, were below 0.05% (the significance limit usually is 0.1%), for each responses. We answered the questions of the fastest and cheapest technology lineup, to laser beam cut a composite fibre cement board to the desired size, with the expected quality.

  • Study of correlation between edge roughness and gas flow characteristics in laser beam fusion cutting

    摘要: Fiber laser fusion cutting trials on AISI 304 stainless steel sheets of 10 mm thickness were performed with the aim to gain further insights into the striation formation process on cut edges. The study is based on a factorial Design-of-Experiment (DoE) approach with consideration of laser power, focal plane position, gas pressure, nozzle stand-off and nozzle diameter as control factors. Analyzed responses include the achievable cutting speed, the cut kerf geometry and the cut edge roughness. In addition, numerical simulations of the cutting gas flow were carried out for adapted kerf geometries and gas parameters. The evaluation of these results reveals strong correlations between experimental roughness values and the numerically computed rear-directed shear stress component within the transition zone between cutting front and cut edge surface. This finding supports the hypothesis that the particular characteristics of the boundary layer zone of the cutting gas flow affects the structure and roughness of cut edges.

  • Experimental Investigation of Laser Surface Transformation Hardening of 4340 Steel Spur Gears

    摘要: This paper presents an experimental investigation of laser surface transformation hardening (LSTH) of 4340 steel spur gears using regression analysis. The experimental work is focused on the effects of various LSTH parameters on the hardness profile shape and the hardened depth variation. The investigations are based on a structured design of experiments and improved statistical analysis tools. The experimentations are carried out on AISI 4340 steel spur gears using a commercial 3 kW Nd:YAG laser system. Laser power, scanning speed, and rotation speed are used as process parameters to evaluate the variation of the hardened depth and to identify the possible relationship between the process parameters and the hardened zone physical and geometrical characteristics. Based on the experimental data and analysis of variance, the direct and interactive contributions of the process parameters on the variation of the hardness profile shape and the hardened depth are analyzed. The main effects and the interaction effects are also evaluated. The results reveal that all the process parameters are relevant. The cumulative contribution of the three parameters in the hardened depth variation represents more than 80% with a clear predominance of laser power. The contribution of the interactions between the parameters represents 12% to 16%. The resulting hardness values are relatively similar for all the experimental tests with about 60 HRC. The evaluation of the produced regression models for hardened depth prediction shows limited performance suggesting that the predictive modeling process can be improved.

  • Accurate and efficient analysis of the upward heat flow in InGaP/GaAs HBTs through an automated FEM-based tool and Design of Experiments

    摘要: This paper presents an extensive analysis aimed at quantifying the impact of all the key technology parameters on the upward heat flow in state‐of‐the‐art InGaP/GaAs heterojunction bipolar transistors (HBTs) for various emitter areas and shapes. Extremely accurate thermal simulations are conducted in a relatively short time with a tool relying on a commercial 3‐D finite‐element method (FEM) solver and an in‐house routine for automated geometry construction, optimized mesh generation, sequential solution, and data storing/processing. Design of Experiments is used to define a thermal resistance model as a function of the aforementioned parameters on the basis of a few FEM data.

  • Multivariate Spatio-temporal Solar Generation Forecasting: A Unified Approach to Deal With Communication Failure and Invisible Sites

    摘要: Short-term prediction of multivariate dynamical processes evolving over time when data are partially observable is a challenging task. The power from solar resources has reached grid parity and must be predicted based on real-time observations available up to the current time step to ensure efficient power systems operation. However, solar data in the form of a time series generated by a network of sensors are not always available. Invisible solar sites and communication failure are two causes that leave the energy management data acquisition system with an incomplete solar time series, thus leading to inaccurate forecasts. This paper addresses the impact of partially observable measurements on short-term solar power prediction. We present a low-rank tensor learning scheme to predict six-hour-ahead solar power generation. We use actual multivariate spatio-temporal National Renewable Energy Laboratory solar data in the state of Kansas presented in the form of tensors along with a photovoltaic power conversion model. A design of experiments experimental framework has been proposed to evaluate single and joint effects of spatio-temporal partially observable sites and the regulating parameters on forecasting accuracy. Numerical results show the capability of the framework to uncover detailed insight into the forecasting model behavior.
