  • Green synthesis of multi-color emissive carbon dots from Manilkara zapota fruits for bioimaging of bacterial and fungal cells

    摘要: Natural resources have widely been used as precursors for the preparation of ultra-small carbon dots (C-dots) due to ease of availability, low cost and C-dots with high quantum yields (QYs). Herein, water dispersible multi-color emissive C-dots were obtained from Manilkara zapota fruits. The emission of C-dots was well tuned by sulphuric acid and phosphoric acids, which results to generate blue-, green- and yellow- C-dots. The fabricated C-dots exhibit blue, green and yellow color emissions when irradiated them under UV light at 365 nm. The emission/excitation peaks of blue-, green-, and yellow- C-dots were observed at 443, 515 and 563 nm when excited at 350, 420 and 440 nm, respectively. The QYs of blue-, green-, and yellow- C-dots are 5.7, 7.9 and 5.2 %. The average sizes of blue- green- and yellow- C-dots are 1.9±0.3, 2.9±0.7and 4.5±1.25 nm, respectively. Because of ultra-small size and biocompatibility, three C-dots act as promising bioimaging agents for imaging of cells (E. coli, Aspergillus aculeatus and Fomitopsis sp). The cytotoxicity on HeLa cells indicates that three C-dots have non-toxic nature, which confirms their biocompatibility. The ultra-small C-dots were effectively distributed in the cytoplasm of the cells, ensuring the potential applications in cell imaging and biomedical studies.

  • Synthesis of new zinc (II) phthalocyanine conjugates with block copolymers for cancer therapy

    摘要: Synthetic routes towards new conjugates of hydrophilic zinc(II) phthalocyanine (ZnPc) with poly(ethylene oxide)-b-poly(propylene oxide)-b-poly(ethylene oxide) (Pluronic P123) and poly(L-lactide) (PLLA), are described. The main semiproduct ZnPc was obtained by heating 4-nitrophthalimide with urea and zinc chloride, followed by the reduction step. Steglich esterification was used to synthesize two ZnPc-conjugated block copolymers, further utilized in fabrication of polymeric micelles (PMs) - functionalized with the zinc(II) phthalocyanine-type moiety. Biological evaluation of the PMs indicated an acceptable biocompatibility level in accord with requirements in the field of nanotheranostics and nanomedicine.

  • A Signal-On Fluorosensor Based on Quench-Release Principle for Sensitive Detection of Antibiotic Rapamycin

    摘要: An antibiotic rapamycin is one of the most commonly used immunosuppressive drugs, and also implicated for its anti-cancer activity. Hence, the determination of its blood level after organ transplantation or tumor treatment is of great concern in medicine. Although there are several rapamycin detection methods, many of them have limited sensitivity, and/or need complicated procedures and long assay time. As a novel fluorescent biosensor for rapamycin, here we propose “Q’-body”, which works on the fluorescence quench-release principle inspired by the antibody-based quenchbody (Q-body) technology. We constructed rapamycin Q’-bodies by linking the two interacting domains FKBP12 and FRB, whose association is triggered by rapamycin. The fusion proteins were each incorporated position-specifically with one of fluorescence dyes ATTO520, tetramethylrhodamine, or ATTO590 using a cell-free translation system. As a result, rapid rapamycin dose-dependent fluorescence increase derived of Q’-bodies was observed, especially for those with ATTO520 with a lowest detection limit of 0.65 nM, which indicates its utility as a novel fluorescent biosensor for rapamycin.

  • Fluorescent Penetrant Testing by Means of Excilamps

    摘要: The paper provides a brief review of modern sources of ultraviolet (UV) radiation used in fluorescent penetrant testing (FT). The differences in implementation of the method with different radiation sources (excilamps, mercury UV lamp and LED UV lamp) are revealed experimentally. It is shown that the XeCl excilamp is not inferior to other sources of UV radiation used in FT.

  • Ratiometric fluorescence imaging for sodium selenite in living cells

    摘要: Sodium selenite as a common selenium-containing agent has been extensively studied for human health, especially cancer prevention and treatment. However, precisely evaluating the dynamic changes of sodium selenite at cellular and molecular level in these pharmacological and pathophysiological studies is still challenging. Developing the chemical tools that enable the quantification of sodium selenite in vivo with high spatial and temporal fidelity thus can provide an efficient solution to this challenge. Here, we report the design, synthesis and biological evaluation of ratiometric sodium selenite fluorescent probe (HBTN-Se) for quantitative ratiometric fluorescence imaging of sodium selenite concentration fluctuations in living cells. A strategy for sodium selenite quantification has been achieved effectively through using a highly selective, rapid and biocompatible hydroxyl-deprotecting reaction. The ratiometric fluorescence imaging with HBTN-Se is constructed to precisely monitor changes in sodium selenite levels in living cells. Moreover, HBTN-Se is capable of visualizing differences in sodium selenite levels between normal cells and cancer cells, establishing the utility of this ratiometric detection platform for assessing the correlation between physiological effects of sodium selenite and its levels in living cells.

  • Synthesis of 2-methyl-5-(5-phenyl substituted-1,3,4 oxadiazole-2-yl) quinazolin-4-one fluorescent brightening agent: Computational and experimental comparison of photophysical structure

    摘要: Report is about the synthesized new range of oxadiazole substituted quinazoline and studied its electronic distribution to attribute fluorescent properties. B3LYP Density Functional Theory (DFT) computational optimization was studied to observe the effect of electron donor and acceptor substituent's on photophysical properties, electronic state and energy level. DFT computational optimization was performed by Polarizable Continuum Model (PCM) of solvation strictly in the gas phase and DMF maintaining C1 symmetry in ground state geometry structure. UV-vis and fluorescence spectroscopic methods help in understanding the relationship between the electron donor and acceptor functional groups on the photophysical properties. Eventually comparing experimental spectral emission and DFT computations were envisage understanding the changes of the electronic transition, energy levels, and electronic orbital distribution in the substituted quinazoline structure. These compounds have good fluorescent brightening properties hence studied and applied as fluorescent brightening agent on polyester fiber.

  • Tuning of carbon dots emission color for sensing of Fe3+ ion and bioimaging applications

    摘要: Herein, we report a facile one-step synthetic strategy for fabrication of three (blue, green and yellow) fluorescent color carbon dots (CDs) from tomato (Solanum lycopersicum). The as-synthesized CDs showed emission peaks at 450, 520 and 560 nm for blue, green and yellow color CDs when excited at 370, 420 and 460 nm, respectively. Using tomato as a carbon source, the fabricated three fluorescent color CDs showed good water dispersity and high quantum yield. The analytical performances of three fluorescent color CDs are evaluated by detecting Fe3+ ion in biofluids and iron tablets. Upon the addition of Fe3+ ion under optimal conditions, the fluorescence intensity of three fluorescent color CDs was quenched linearly over the range of 0.1 to 2.0 μM. This method opens a new analytical strategy to quantify Fe3+ ion in iron tablets and biofluids with high sensitivity. Further, the uptake of three fluorescent color CDs into HeLa cells was confirmed by confocal laser scanning microscopy. Intracellular experiments demonstrated that the three fluorescent color CDs were effectively internalized the cells and show excellent biocompatibility and low toxicity, suggesting that the CDs can be used as good candidates for biomedical applications.

  • Medium viscosity effect on fluorescent properties of Sn(IV)-tetra(4-sulfonatophenyl)porphyrin complexes in buffer solutions

    摘要: Supramolecular triads consisting of hydrophilic Sn(IV)-tetra(4-sulfophenyl)-porphyrin and two axial guests such as propylphenol, tyrosine, and 2-(2-hydroxyphenyl)-benzoxazole were synthesized. The structures of synthesized complexes were identified by experimental spectroscopic and quantum-chemical simulation methods, and their fluorescent properties were studied in various viscosity media (mixed phosphate buffer-glycerin solvents of different composition). The effect of axial ligand structure on the fluorescent properties of these triads (due to the hydrogen bonding or π-π stacking between the components of ?host-guest? systems) is discussed. The potential use of synthesized complexes as environmental probes of local viscosity is proposed.

  • A selective fluorescent chemosensor for Cd2+ based on 8-hydroxylquinoline-benzothiazole conjugate and imaging application

    摘要: A 8-hydroxylquinoline-benzothiazole conjugate (HQ-BT) was facilely synthesized by two steps with more than 60% total reaction yield. The HQ-BT showed a weak fluorescence that could be strongly enhanced by coordination with various metal ions such as Al3+, Cd2+, Zn2+ in methanol containing 1% water. Interestingly, the selectivity toward Cd2+ was achieved by increasing water fraction to 30% aqueous methanol solution. Thus, the HQ-BT was developed as a new and selective fluorescent chemosensor for Cd2+ in aqueous solution with a broad pH region 4–12. A good linear relationship between the fluorescence intensity and the Cd2+ concentration was found in the range of 0–5 μM with a detection limit of 0.1 μM (S/N = 3). It was also succesfully used for fluorescence imaging of Cd2+ in living cells.

  • A high-performance fluorescent probe for dopamine detection based on g-C3N4 nanofibers

    摘要: A novel fluorescent sensor based on g-C3N4 nanofibers for the sensitive detection of dopamine (DA) has been proposed. We synthesized g-C3N4 nanofibers by directly hydrolyzing bulk g-C3N4 in the alkaline atmosphere (3 M NaOH). The obtained ultrathin g-C3N4 nanofibers were verified by characterizations of Transmission electronic microscope (TEM), X-ray diffractometer (XRD), Fourier transformation-infrared (FT-IR) and X-Ray photoelectron spectroscopy (XPS). It was found that the fluorescence intensity of g-C3N4 nanofibers was obviously quenched by DA. Fluorescence resonance energy transfer (FRET) between DA and g-C3N4 nanofibers led to the fluorescence reduction of g-C3N4 nanofibers. The fluorescent probe based on g-C3N4 nanofibers exhibits linear responses to the concentration of DA in the range from 0 to 4 μM and 4 to 20 μM, the limit of detection is 17 nM. The fluorescent probe shows excellent stability, good selectivity with its application in serums.
