  • Potassium triiodide-quenched gold nanocluster as a fluorescent turn-on probe for sensing cysteine/homocysteine in human serum

    摘要: A fluorescent sensing platform using KI3-quenched bovine serum albumin stabilized gold nanoclusters has been designed and used as a fluorescent probe for the turn-on detection of homocysteine/cysteine (Cys/Hcy). The fluorescence of gold nanoclusters was quenched by iodine. The fluorescence of quenched gold nanoclusters was effectively switched on by Cys/Hcy devoid of the interference of glutathione. The transmission electron microscopy image, X-ray photoelectron spectroscopy analysis, time-correlated single photon counting analysis, and dynamic light scattering data confirmed the aggregation-induced quenching of fluorescence of gold nanoclusters by iodine. The turn-on response of Cys/Hcy shows two linear ranges from 0.0057 to 5 μM and from 8 to 25 μM, with a limit of detection of 9 nM for cysteine and 12 nM for homocysteine. Real samples were analyzed to monitor Cys/Hcy added to human serum. The fluorescence turn-on response of the probe on a paper strip in the presence of Cys/Hcy was studied.

  • A coumarin-based dual optical probe for homocysteine with rapid response time, high sensitivity and selectivity

    摘要: In this study, a new coumarin-based fluorescent and chromogenic dual channel probe (DC) was used for the selective detection of homocysteine (Hcy) over other amino acids, especially for cysteine (Cys) and glutathione (GSH). When Hcy is present in the solution, the remarkable fluorescence enhancement and obvious blue shift in UV–vis spectra can be observed. In addition, the color change from purple to yellow can be observed clearly by unaided eyes. This probe DC has fast response time, excellent sensitivity and selectivity to Hcy. A linear relationship exists between the ratio of emissions at 486 and 625 nm, and Hcy can be detected in a wide concentration range (0 to 200 μM). The signal-to-background ratio of fluorescence at 486 nm can reach 8.4, and the detection limit is calculated to be 3.5 μM. The response mechanism is proved to be the Michael addition reaction by Hcy. Preliminary results on cell imaging enable the practical application of Hcy tracing in living cells.

  • A series of BODIPY-based probes for the detection of cysteine and homocysteine in living cells

    摘要: Biothiols, such as glutathione (GSH), homocysteine (Hcy) and cysteine (Cys), are important biomarkers and play crucial roles in many physiological processes. Thus, the detection of biothiols is highly important for early diagnosis of diseases and evaluation of disease progression. Herein, new types of BODIPY-based fluorescent probes (probe 1, probe 2 and probe 3) capable of cysteine (Cys)/homocysteine (Hcy) sensing with high selectivity over other amino acids were developed. In addition, we further studied the influence of different electronegativity substituents on these probes to sensing Cys/Hcy. Ultimately, we concluded that the electron withdrawing group on probe 1 can accelerate the probe response to Cys/Hcy, and probe 1 was successfully applied for selective imaging Cys/Hcy in living cells.
