  • Aggregation-Induced Absorption Enhancement for Deep Near-Infrared II Photoacoustic Imaging of Brain Gliomas In Vivo

    摘要: The delineation of brain gliomas margins still poses challenges to precise imaging and targeted therapy, mainly due to strong light attenuation of the skull and high background interference. With deep penetration and high sensitivity, photoacoustic (PA) imaging (PAI) in the second near-infrared (NIR II) window holds great potential for brain gliomas imaging. Herein, mesoionic dye A1094 encapsulated in Arg-Gly-Asp-modified hepatitis B virus core protein (RGD-HBc) is designed and synthesized for effective NIR II PAI of brain gliomas. An aggregation-induced absorption enhancement mechanism is discovered in vitro and in vivo. It is also demonstrated that A1094@RGD-HBc, with an enhanced absorption in the NIR II window, displays ninefold PA signal amplification in vivo, allowing for precise PAI of the brain gliomas at a depth up to 5.9 mm. In addition, with the application of abovementioned agent, high-resolution PAI and ultrasensitive single photon emission computed tomography images of brain gliomas are acquired with accurate co-localization. Collectively, the results suggest great promise of A1094@RGD-HBc for diagnostic imaging and precise delineation of brain gliomas in clinical applications.

  • Embedded Near-Infrared Sensor with Tunable Sensitivity for Nanoscale CMOS Technologies

    摘要: Near-infrared (NIR) sensors has become one of the key components in applications, such as temperature, proximity, and even ?ngerprint sensing. A novel full-CMOS (Complementary Metal-Oxide-Semiconductor) compatible near-infrared sensor is proposed and demonstrated in this article. This sensing device consists of a gateless transistor, enabling near-infrared photon absorption in the channel region. With a tunable channel barrier, this near-IR sensor also features a dynamic photo-response with an extending sensing range up to 60 dB.

  • Perspective: Prospects of non-invasive sensing of the human brain with diffuse optical imaging

    摘要: Since the initial demonstration of near-infrared spectroscopy (NIRS) for noninvasive measurements of brain perfusion and metabolism in the 1970s, and its application to functional brain studies (fNIRS) in the 1990s, the field of noninvasive optical studies of the brain has been continuously growing. Technological developments, data analysis advances, and novel areas of application keep advancing the field. In this article, we provide a view of the state of the field of cerebral NIRS, starting with a brief historical introduction and a description of the information content of the NIRS signal. We argue that NIRS and fNIRS studies should always report data of both oxy- and deoxyhemoglobin concentrations in brain tissue, as they complement each other to provide more complete functional and physiological information, and may help identify different types of confounds. One significant challenge is the assessment of absolute tissue properties, be them optical or physiological, so that relative measurements account for the vast majority of NIRS and fNIRS applications. However, even relative measurements of hemodynamics or metabolic changes face the major problem of a potential contamination from extracerebral tissue layers. Accounting for extracerebral contributions to fNIRS signals is one of the most critical barriers in the field. We present some of the approaches that were proposed to tackle this challenge in the study of cerebral hemodynamics and functional connectivity. Finally, we critically compare fNIRS and functional magnetic resonance imaging by relating their measurements in terms of signal and noise, and by commenting on their complementarity.

  • Near-infrared fluorescence laparoscopy of the ureter with three preclinical dyes in a pig model

    摘要: Background Ureteric injury is reported to occur in 1–7.6% of colorectal surgeries. To reduce the incidence of ureteral injury, it is essential to identify the ureters. The use of near-infrared fluorescence (NIRF) imaging with intravenously administered dyes might be of added value for ureteral visualization during laparoscopy. The aim of this study is to assess the performance of three preclinical dyes; IRDye? 800BK, IRDye? 800NOS and IRDye? 800CW, for near-infrared fluorescence laparoscopy of the ureter in pigs. Methods In three female Dutch landrace pigs, the new dyes were evaluated. In each pig, 1 dye was tested using a 6-mg intravenous dose in a concentration of 1?mg/ml. Imaging was performed in fluorescence mode and white light mode with a laparoscopic imaging system. In order to further evaluate the dyes, an ex?vivo imaging experiment was performed, in which 8 decreasing concentrations per dye, diluted in PBS, were evaluated in a transparent test tube with NIRF mode at a distance of 1, 5 and 10?cm from the laparoscope. Results All three dyes were effective in allowing the identification of the ureter with NIRF imaging. The ureter became fluorescent after 35, 45 and 10?min, respectively, for IRDye? 800BK, IRDye? 800NOS and IRDye? 800CW with a maximum target-to-background ratio (TBR) of 2.14, 0.66 and 1.44, respectively. In the ex?vivo imaging experiment, all three dyes produced a strong fluorescence signal at all concentrations and all distances evaluated. Conclusions Intravenous administration of the preclinical dyes IRDye? 800CW, IRDye? 800 BK and IRDye? 800NOS facilitated successful identification of the anatomical course of the ureter in living pig models. The highest measured TBR occurred with the use of IRDye? 800BK. Ex?vivo, a correlation was observed between the fluorescence intensities of the signal with the concentration of the dye and with the distance to the object.

  • Glutathione Triggered Near Infrared Fluorescence Imaging-guided Chemotherapy by Cyanine Conjugated Polypeptide

    摘要: Precise detection of tumor environment for cancer diagnosis was strongly demanded for further therapies. Here, a redox-responsive fluorescence switch off/on system PCQ was designed and synthesized conjugated with near infrared (NIR) cyanine dyes (Cy5.5) and relevant quencher (FQ) in mixed polymeric micelles (PCy and PFQ). The mixed PCQ micelles was prepared with two kinds of polymer with poly (oligo (ethylene glycol) methacrylate) (POEGMA) as the hydrophilic shell, in which fluorescence emission was quenched by fluorescence resonance energy transfer (FRET) effect. The FQ was conjugated with POEGMA by disulfide linkage, which could be broke with redox environment such as high glutathione (GSH) concentration in tumor cells. After the PCQ micelles got into tumor cells, PFQ block in PCQ would be disassembled to recompose PCy micelles. During that process, drugs like doxorubicin (DOX) could be loaded inside and formed PCQ@DOX nanoparticles and then released for accurate NIR bioimaging and drug delivery instantly.

  • Enhanced photoacoustic and photothermal effect of functionalized polypyrrole nanoparticles for near-infrared theranostic treatment of tumor

    摘要: Functionalized nanomaterials with near-infrared (NIR) responsive capacity are quite promising for theranostic treatment of tumors, but formation of NIR responsive nanomaterials with enhanced theranostic ability and excellent biocompatibility is still very challenging. Herein, PEGylated indocyanine green (ICG)-loaded polypyrrole nanoparticles (PPI NPs) were designed and successfully formed through selecting polydopamine as the linkage between each component, demonstrating enhanced NIR responsive theranostic ability against tumor. Combining in vitro cell study with in vivo assay, the formed PPI NPs were proved being fantastic biocompatible while effectively internalizing in HeLa cells and retaining in HeLa tumor demonstrated by in vitro flow cytometry/confocal measurement and in vivo photoacoustic imaging assay. With the guidance of photoacoustic imaging, successful photothermal ablation of tumor was achieved when treating with PPI NPs plus laser, which was much more effective than the group treated with NPs free of ICG. The greatly combined enhanced photoacoustic and photothermal effect is mainly ascribed to the functionalized polypyrrole nanoparticles, which could accumulate in tumor site more effectively with a relative longer retention time taking advantage of the nanomaterial-induced endothelial leakiness phenomenon. All these results demonstrate the designed PPI NPs possess enhanced NIR responsive property are to hold a great promise for tumor NIR theranostic applications.

  • Muscle oxygenation dynamics in response to electrical stimulation as measured with near-infrared spectroscopy: A pilot study

    摘要: Neuromuscular electrical stimulation (NMES) is used for preventing muscle atrophy and improving muscle strength in patients and healthy people. However, the current intensity of NMES is usually set at a level that causes the stimulated muscles to contract. This typically causes pain. Quantifying the instantaneous changes in muscle microcirculation and metabolism during NMES before muscle contraction occurs is crucial, because it enables the current intensity to be optimally tuned, thereby reducing the NMES-induced muscle pain and fatigue. We applied near-infrared spectroscopy (NIRS) to measure instantaneous and deoxygenation changes in 43 healthy young adults during NMES at 10, 15, 20, 25, 30, and 35 mA. Having been stabilized at the NIRS signal baseline, total hemoglobin concentration increased immediately after stimulation in a dose-dependent manner (p < 0.05) until stimulation was stopped at the level causing muscle contraction without pain. Tissue deoxygenation appeared relatively unchanged during NMES. We conclude that NIRS can be used to determine the optimal NMES current intensity by monitoring oxygenation changes.

  • Development of Near-Infrared Reflectance Spectroscopy (NIRS) Calibrations for Traits Related to Ethanol Conversion from Genetically Variable Napier Grass (Pennisetum purpureum Schum.)

    摘要: Napier grass (Pennisetum purpureum Schum.) is one of the highest-yielding feedstocks for bio-based products and biofuel in semi-tropical areas of the USA and the world. Thirty genetically diverse Napier grass accessions were selected from a germplasm nursery in Tifton, GA and analyzed for fiber, ash, nitrogen (N) concentration, and biochemical conversion to ethanol. A near-infrared reflectance spectroscopy (NIRS) calibration was developed from this material to predict ethanol production, xylans, N concentration, and ash by separating leaves and stems and correlating with wet chemistry analyses. The high diversity of material from dwarf material with high leaf and stem digestibility to taller and more productive Napier grass cultivars resulted in high correlations with predicted results for in vitro dry matter digestibility (2 = 0.93), neutral detergent fiber (r2 = 0.83), acid detergent fiber (r2 = 0.95), ethanol (r2 = 0.90), nitrogen (r2 = 0.99), and ash (r2 = 0.98). This information will allow faster evaluation of Napier grass biomass for use by industry or geneticists.

    摘要: This paper presents a battery-free wearable eye tracker that tracks both the 2D position and diameter of a pupil based on its light absorption property. With a few near-infrared (NIR) lights and photodiodes around the eye, NIR lights sequentially illuminate the eye from various directions while photodiodes sense spatial patterns of reflected light, which are used to infer pupil’s position and diameter on the fly via a lightweight inference algorithm. The system also exploits characteristics of different eye movement stages and adjusts its sensing and computation accordingly for further energy savings. A prototype is built with off-the-shelf hardware components and integrated into a regular pair of glasses. Experiments with 22 participants show that the system achieves 0.8-mm mean error in tracking pupil position (2.3 mm at the 95th percentile) and 0.3-mm mean error in tracking pupil diameter (0.9 mm at the 95th percentile) at 120-Hz output frame rate, consuming 395μW mean power supplied by two small, thin solar cells on glasses side arms.

  • Variety Identification of Raisins Using Near-Infrared Hyperspectral Imaging

    摘要: Different varieties of raisins have different nutritional properties and vary in commercial value. An identification method of raisin varieties using hyperspectral imaging was explored. Hyperspectral images of two different varieties of raisins (Wuhebai and Xiangfei) at spectral range of 874–1734 nm were acquired, and each variety contained three grades. Pixel-wise spectra were extracted and preprocessed by wavelet transform and standard normal variate, and object-wise spectra (sample average spectra) were calculated. Principal component analysis (PCA) and independent component analysis (ICA) of object-wise spectra and pixel-wise spectra were conducted to select effective wavelengths. Pixel-wise PCA scores images indicated differences between two varieties and among different grades. SVM (Support Vector Machine), k-NN (k-nearest Neighbors Algorithm), and RBFNN (Radial Basis Function Neural Network) models were built to discriminate two varieties of raisins. Results indicated that both SVM and RBFNN models based on object-wise spectra using optimal wavelengths selected by PCA could be used for raisin variety identification. The visualization maps verified the effectiveness of using hyperspectral imaging to identify raisin varieties.
