  • A disposable microfluidic-integrated hand-held plasmonic platform for protein detection

    摘要: Healthcare is in the midst of a transformative shift from centralized care to point-of-care (POC). In this regard, recent efforts have focused on integration of biosensing technologies with clinical management and existing healthcare systems to improve the effectiveness and quality of care. Plasmonic technologies in particular, have been used for multiple applications in biosensing, pharmaceutical industry, food quality monitoring, and healthcare. However, bulky-sized platforms, expensive instrumentation, incomprehensive benchmarking, laborious protocols, and time-consuming processing steps remain challenges to adopt biosensing platforms to the POC settings. Here, we present a hand-held biosensing platform that integrates a plasmonic detection modality with a microfluidic chip. As a biological target model, we assess hemoglobin—an iron carrying protein in red blood cells. We comprehensively perform theoretical simulations and kinetic calculations to benchmark the platform performance. Overall, this miniaturized platform provides label-free detection, simple configuration for user-interface, facile sampling, assay-time down to 15–30 min, and inexpensive disposable chips. Therefore, this platform will potentially accelerate the deployment of portable biosensing systems for the POC and primary care settings.

  • Smartphone-based three-channel ratiometric fluorescent device and application in filed analysis of Hg2+, Fe3+ and Cu2+ in water samples

    摘要: Determination of metal ions in water samples, especially harmful heavy metal like mercury or essential elements like iron and copper, is an important work in environmental monitoring. Herein, we report a smartphone-based three-channel ratiometric fluorescence device for simultaneous determination of Hg2+, Fe3+ and Cu2+ in environmental samples on site. It was based on the fluorescence quench mechanism of Hg2+, Fe3+ and Cu2+ to three kinds of doped carbon quantum dots, which were prepared by pyrolysis methods using ammonium citrate, citric acid + 1,10-phenanthroline, and EDTA + thiourea as initial materials, respectively. By using a multivariate calibration method, the mutual interference among Hg2+, Fe3+ and Cu2+ can be corrected. Under the optimized conditions, the limits of detection for Hg2+, Cu2+ and Fe3+ are 3, 0.5, and 30 nM, respectively. The proposed device offers the advantage of portability, sensitivity and reliability for field analysis.

  • Detection of Semen Stains in Rape Cases by a Very High Powered UV-VIS Light Source, Facilitated Conviction of Accused Person

    摘要: Detection of semen in forensic cases mostly sexual assault or abuse is critical for evidence collection. A portable forensic light source, which emits wavelength from 430 nm to 700 nm (UV to VIS) and filters were used. The very high intensity light source detected semen in darkness and daylight using wavelength between 415 nm to 490 nm. ABO blood grouping method is an oldest method used in forensic biology for grouping purpose. But now a days, DNA fingerprinting is an advanced technique used to solve crimes like gang rape. Even though in this case, simplest and oldest ABO blood grouping method helped to solve gang rape case. In present case, victim was 5 years old girl who was raped by a person. Screening the semen stains using portable forensic light source and primary testing of semen stains from different places on victim’s undergarments was done. ABO blood grouping of each selected semen stains were done by absorption-elution method. Those selected stains showed blood group which matched with the accused person’s blood group. Thus the accused person was convicted in this heinous crime.

    摘要: Long-term monitoring of the fetal heart rate (fHR) is important to control the fetal well-being. One of the promising methods for fetal monitoring is fetal phonocardiography which is based on recording fetal heart sounds. This paper presents a compact low-cost device for multi lead fHR measurement. The device introduced allows monitoring of fHR non-invasively and with no side effects for the mother or the foetus. In this case, the device presented is used to measure HR of the adult to compare the performance of various stethoscope heads. In addition to this measurement, the LabVIEW application for signal processing and analysis was improved.

  • Portable Multiplex Optical Assays

    摘要: Global health issues are increasingly becoming critical with high fatality rate due to chronic and infectious diseases. Emerging technologies aim to address these problems by understanding the causes of lethal conditions and diagnosing symptoms at early stage. Existing commercial diagnostics primarily focus on single-plex assays due to ease-of-use, simplicity in analysis, and amenability to mass manufacturing. Many research grade devices have utilized only a few molecular and morphological signatures in bodily fluids. However, multiplex devices can improve accuracy, sensitivity, and scalability of research and diagnostic devices. This review presents multiplex assays that utilize optical, electrical, and chemical methods and materials that have the potential to improve portable point-of-care diagnostics. The critical role of emerging optical and complementary assays with multiple contrast mechanisms is investigated to enable highly multiplex analysis in field settings. Multiparameter portable devices for field applications toward health monitoring, food testing, air quality monitoring, and microanalysis in other extreme conditions are examined. Current trends indicate the need for validation of health diagnosis based on a large number of biomarkers in randomized clinical trials. Advanced digital analysis, crowd-sourced solutions, and robust user interfaces will become an integral part of the connected global health systems and personalized monitoring platforms.

  • Measuring heavy metal content in bone using portable X-ray fluorescence

    摘要: The ability of inorganic-based analytical chemistry techniques to quantify trace amounts of heavy metals in skeletal remains has been integral for understanding health and social status in human populations. Low detection limits and the sensitivity of inductively coupled plasma-mass spectrometry (ICP-MS) and other techniques to most elements on the periodic table are ideally suited for the quantification of lead (Pb) and other heavy metals in bone. However, the time required for sample preparation and analysis, expense, destructive analytical process, and availability of instrumentation often limit researchers’ ability to utilise these techniques for archaeological applications. This paper explores the use of portable X-ray fluorescence (XRF) instrumentation for heavy metal analysis of bone as an alternative to more traditional analytical techniques. XRF has been shown to be an extremely useful tool for archaeologists seeking to conduct quantitative analyses of cultural materials such as obsidian and metals. However, little research has been undertaken to assess the usefulness of portable XRF for measuring heavy metals found in low concentrations in archaeological bone. This paper compares data derived from ICP-MS and portable XRF analyses of bone. Results demonstrate that XRF analyses of bone are problematic due to diagenesis and variability of Pb content in bone.

    摘要: Folate is an essential vitamin to the development of a fetus in early pregnancy. Maternal folate supplementation around the time of conception has been shown to decrease the risk of neural tube defects (NTDs), a class of serious birth defects. The closure of the neural tube before the 28th day after conception necessitates that the folate intake must take place before most women know that they are pregnant. Therefore, screening women of reproductive age for folate status would allow for an improved understanding of the need for supplementation in women who could become pregnant as well as the effectiveness of current supplementation and fortification recommendations. Current folate assessment is limited to lab-based assays which require expensive equipment, trained personnel, and are time-intensive. Our point-of-care diagnostic test quantifies levels of folate in human serum with the use of a lateral flow assay and a portable imaging device. We have designed an assay which uses fluorescent particles, folate binding protein, and antibodies to measure serum folate. This test could be used in resource-limited settings, where access to laboratory infrastructure is limited and where knowledge of folate status in women of reproductive age is lacking. By increasing our understanding of folate status around the world, we can improve implementation of folic acid supplementation and fortification and therefore reduce the risk of NTDs.

  • A field-deployable surface-enhanced Raman scattering (SERS) method for sensitive analysis of silver nanoparticles in environmental waters

    摘要: The proliferation of silver nanoparticles (AgNPs) in the environment and resultant transport into aquatic systems have raised concerns regarding their potential toxicity to various organisms. These environmental and ecological concerns demand reliable AgNP detection methods which can measure environmentally relevant quantities of AgNPs in real aquatic systems. This study developed a method that couples a rapid vacuum filtration technique with a portable Raman spectrometer to achieve on-site detection of ultra-low levels of AgNPs in typical and complex aquatic systems. To extract and detect AgNPs, aluminum chloride and ferbam were added for AgNP aggregation and labelling, respectively. The AgNP aggregates were filtered through a membrane, and their presence and quantity were determined based upon the surface-enhanced Raman scattering (SERS) peak intensity of ferbam. Under the optimized conditions, the extraction efficiencies are 99 ± 0.001% in ultrapure water and 98 ± 0.025% in marine water for 1 mg/L AgNPs. This method enables simple volume adjustment and improves the consistency of AgNP distribution on the membrane. The performance of the method was evaluated in different environmental waters, including marine water, fresh waters (pond water, river water, and reservoir outlet water) and drinking waters (municipal tap water and well water), with highest signal intensity in marine water and lowest signals in fresh waters. The signal intensity difference was suggested to be caused by the amount of natural organic matter (NOM) in these environmental waters. Using pond water as an example, the interference was minimized by changing the aggregating salt from AlCl3 to MgCl2, and AgNPs as low as 5 μg/L were reliably detected with a volume of 100 mL. At the same volume, the developed method was sensitive enough to detect 1 μg/L AgNPs in marine water and also holds promise for assessing the time-dependent transformation of AgNPs.

  • Observation of short period ionospheric disturbances using a portable digital ionosonde at Sanya

    摘要: A number of ionosondes have been operated in China to detect ionospheric disturbances for many years. These ionosondes, however, are not very suitable for the short period (<15 min) disturbances due to their poor time resolutions (>5 min). During recent years, the Institute of Geology and Geophysics, Chinese Academy of Sciences (IGGCAS), together with the South Central University for Nationalities (SCUN) have been developing a portable digital ionosonde (PDI) equipped with the capability to detect and characterize small-scale/short period ionospheric disturbances, and to be quickly assembled and set up at temporary field stations for low-latitude campaign coordinated observations. The PDI uses a set of technologies (for example code multiplexing and antenna/transmitter matching) that allow it to obtain quality Doppler-ionograms at a good time resolution with small transmitting antennas. A preliminary analysis of observations by the PDI at Sanya (18.3°N, 109.6°E) shows the presence of ionospheric disturbances with periods ranging between several and tens of minutes. Interestingly, the disturbances (with different periods) were found to simultaneously occur at different F region altitudes, for example with periods of ~5 and 20 min below and above ~180 km, respectively. The absence of shorter period disturbance at higher altitude is consistent with acoustic gravity waves through the region with intrinsic periods above the Brunt-V?is?l? period. The short period disturbances observed in F region bottomside are not evident in TEC. The results demonstrate the capability of PDI to detect ionospheric disturbances with temporal scales down to a few minutes in routine ionogram mode. Future prospects of PDI are outlined.

  • Toward Point-of-Care Drug Quality Assurance in Developing Countries: Comparison of Liquid Chromatography and Infrared Spectroscopy Quantitation of a Small-Scale Random Sample of Amoxicillin

    摘要: Substandard antibiotics are thought to be a major threat to public health in developing countries and a cause of antimicrobial resistance. However, assessing quality outside of a laboratory setting, using simple equipment, is challenging. The aim of this study was to validate the use of a portable Fourier transform infrared (FT-IR) spectrometer for the identification of substandard antibiotics. Results are presented for amoxicillin packages from Haiti, Ghana, Sierra Leone, Democratic Republic of Congo, India, Papua New Guinea, and Ethiopia collected over the course of 6 months in 2017, including two field trips with the FT-IR to Ghana and Sierra Leone. Canadian samples were used as a control. Regarding drug quality, of 290 individual capsules of amoxicillin analyzed, 13 were found to be substandard with total active pharmaceutical ingredients (API) lying outside the acceptable range of 90–110%. Of these 13, four were below 80% API. The FT-IR reliably identified these outliers and was found to yield results in good agreement with the established pharmacopeia liquid chromatography protocol. We conclude that the portable FT-IR may be suitable to intercept substandard antibiotics in developing countries where more sophisticated techniques are not readily available.
