  • Electro-optic imaging enables efficient wide-field fluorescence lifetime microscopy

    摘要: Nanosecond temporal resolution enables new methods for wide-field imaging like time-of-flight, gated detection, and fluorescence lifetime. The optical efficiency of existing approaches, however, presents challenges for low-light applications common to fluorescence microscopy and single-molecule imaging. We demonstrate the use of Pockels cells for wide-field image gating with nanosecond temporal resolution and high photon collection efficiency. Two temporal frames are obtained by combining a Pockels cell with a pair of polarizing beam-splitters. We show multi-label fluorescence lifetime imaging microscopy (FLIM), single-molecule lifetime spectroscopy, and fast single-frame FLIM at the camera frame rate with 103–105 times higher throughput than single photon counting. Finally, we demonstrate a space-to-time image multiplexer using a re-imaging optical cavity with a tilted mirror to extend the Pockels cell technique to multiple temporal frames. These methods enable nanosecond imaging with standard optical systems and sensors, opening a new temporal dimension for wide-field low-light microscopy.

  • Temporal resolution of single photon responses in primate rod photoreceptors and limits imposed by cellular noise

    摘要: Sensory receptor noise corrupts sensory signals, contributing to imperfect perception and dictating central processing strategies. For example, noise in rod phototransduction limits our ability to detect light and minimizing the impact of this noise requires precisely tuned nonlinear processing by the retina. But detection sensitivity is only one aspect of night vision: prompt and accurate behavior also requires that rods reliably encode the timing of photon arrivals. We show here that the temporal resolution of responses of primate rods is much finer than the duration of the light response and identify the key limiting sources of transduction noise. We also find that the thermal activation rate of rhodopsin is lower than previous estimates, implying that other noise sources are more important than previously appreciated. A model of rod single-photon responses reveals that the limiting noise relevant for behavior depends critically on how rod signals are pooled by downstream neurons.

  • Technical note: Coupling infrared gas analysis and cavity ring down spectroscopy for autonomous, high-temporal-resolution measurements of DIC and <i>δ</i><sup>13</sup>C–DIC

    摘要: A new approach to autonomously determine concentrations of dissolved inorganic carbon (DIC) and its carbon stable isotope ratio (δ13C–DIC) at high temporal resolution is presented. The simple method requires no customised design. Instead it uses two commercially available instruments currently used in aquatic carbon research. An inorganic carbon analyser utilising non-dispersive infrared detection (NDIR) is coupled to a Cavity Ring-down Spectrometer (CRDS) to determine DIC and δ13C–DIC based on the liberated CO2 from acidi?ed aliquots of water. Using a small sample volume of 2 mL, the precision and accuracy of the new method was comparable to standard isotope ratio mass spectrometry (IRMS) methods. The system achieved a sampling resolution of 16 min, with a DIC precision of ±1.5 to 2 μmol kg?1 and δ13C–DIC precision of ±0.14 ‰ for concentrations spanning 1000 to 3600 μmol kg?1. Accuracy of 0.1 ± 0.06 ‰ for δ13C–DIC based on DIC concentrations ranging from 2000 to 2230 μmol kg?1 was achieved during a laboratory-based algal bloom experiment. The high precision data that can be autonomously obtained by the system should enable complex carbonate system questions to be explored in aquatic sciences using high-temporal-resolution observations.

  • Pulse dilation gas Cherenkov detector for ultra-fast gamma reaction history at the NIF (invited)

    摘要: The Cherenkov mechanism used in Gas Cherenkov Detectors (GCDs) is exceptionally fast. However, the temporal resolution of GCDs, such as the Gamma Reaction History diagnostic at the National Ignition Facility (NIF), has been limited by the current state-of-the-art photomultiplier tube technology to ~100 ps. The soon-to-be deployed Pulse Dilation Photomultiplier Tube (PD-PMT) at NIF will allow for temporal resolution comparable to that of the gas cell or ~10 ps. Enhanced resolution will contribute to the quest for ignition in a crucial way through precision measurements of reaction history and ablator areal density (ρR) history, leading to better constrained models. Features such as onset of alpha heating, shock reverberations, and burn truncation due to dynamically evolving failure modes may become visible for the ?rst time. Test measurements of the PD-PMT at Atomic Weapons Establishment con?rmed that design goals have been met. The PD-PMT provides dilation factors of 2 to 40× in 6 increments. The GCD-3 recently deployed at the NIF has been modi?ed for coupling to a PD-PMT and will soon be making ultrafast measurements.

  • Three-strip microchannel plate gated x-ray framing camera

    摘要: A microchannel plate (MCP) gated x-ray framing camera with three-strip is reported. The diameter of the MCP is 56 mm and the width of each microstrip line cathode deposited on the MCP is 8 mm. While the microstrip line cathode is driven by a gating electrical pulse with width of 200 ps and amplitude of ?1.9 kV overlapped with a ?200 V DC bias, the measured temporal resolution is about 68 ps. Furthermore, the gain uniformity of the microstrip line cathode is measured showing that there is a 3.6× drop in gain along the pulse propagation direction, whereas the variations in the gain transverse to the pulse propagation direction are within 10%.
