  • Straw Degradation Behaviors under Different Conditions of Relative Air Humidity and Ultraviolet-A Irradiation

    摘要: In this study, straw was degraded continuously for 150 days under one of three levels of relative air humidity (RH) (90%, 60%, or 30%) to estimate the effect of humidity on straw biodegradation. Moreover, straw was treated with ultraviolet (UV)-A irradiation + 90% RH for 180 days to evaluate the interaction between photodegradation and biodegradation. The effects of 30% and 60% RH on straw degradation was inconspicuous. Straw mass losses at 90% RH and UV-A + 90% RH were 18.5% and 39.1%, respectively. BIOLOG analysis showed that filamentous fungi played a major role in straw biodegradation. Thermogravimetric analysis showed that treatment with UV-A + 90% RH tended to increase the maximum pyrolysis rate and decreased the initial pyrolysis temperature. Compared with 90% RH, infrared spectra analysis showed that functional groups of UV-A + 90% RH treatment, e.g., –CH, –C=O, and the benzene ring structure, clearly decreased. Straw-degrading bacteria were observed by scanning electron microscopy at the beginning and end of UV-A + 90% RH treatment. Results highlight the role of humidity in the degree of straw biodegradation by filamentous fungi. Straw degradation is accelerated by the combined action of photodegradation and biodegradation under high UV-A irradiation and high humidity.

  • Bactericidal action of ferulic acid with ultraviolet-A light irradiation

    摘要: The bactericidal activity of ferulic acid (FA) against various microorganisms was remarkably enhanced by ultraviolet-A (UV-A) irradiation (wavelength, 365 nm). However, the bactericidal mechanism in the photo-combination system has not been evaluated. In the present study, this combined treatment was characterized by investigating associated changes in cellular functions of Escherichia coli, including assessments of respiratory activity, lipid peroxidation, membrane permeability, and damage to DNA and the cell surface. FA adsorbed onto and was incorporated into bacterial membranes, and the affinity resulted in decreased respiratory activity and enhanced lipid peroxidation in the cytoplasmic membrane with low-fluence (1.0 J/cm2) UV-A irradiation. Flow cytometry analysis revealed that additional exposure (8 J/cm2) combined with FA (1 mg/mL) induced increased cell permeability, yielding a 4.8-log decrease in the viable cell count. Morphologically, the treated cells exhibited a bacterial membrane dysfunction, producing many vesicles on the cell surface. However, despite this effect on the cell surface, plasmid DNA transformed into FA-treated E. coli maintained supercoiled integrity with negligible DNA oxidation. Our data strongly suggested that FA functions inside and outside the bacterial membrane; UV-A exposure in the presence of FA then causes increased oxidative modification and subsequent disruption of the bacterial membrane, without causing detectable genotoxicity.

  • Visible light accelerates the ultraviolet A-induced degradation of eumelanin and pheomelanin

    摘要: Exposure to excess ultraviolet (UV) A radiation induces the degradation/modification of both eumelanin and pheomelanin that may be deleterious to pigmented tissues. Although the spectral distribution of solar energy comprises nearly half of visible light (VL), few studies have been conducted to examine the role of VL in the photodegradation of both types of melanin, either VL alone or in combination with UVA. In this study, we examined the effects of physiological doses of VL (150 to 300 J/cm2) alone or in combination with a physiological dose of UVA (20 J/cm2) in normal human epidermal melanocytes. The degradation/modification of melanin structures were evaluated by our chemical degradation – HPLC methods. The results show that VL accelerates UVA-induced changes in the structural features of both eumelanin and pheomelanin, although VL or UVA alone induced only minor changes in melanin structure. The differential spectral method provides support for the additive effects of VL.
