  • Immobilization of Detonation Nanodiamonds on Macroscopic Surfaces

    摘要: Detonation nanodiamonds (NDs) are a novel class of carbon-based nanomaterials, and have received a great deal of attention in biomedical applications, due to their high biocompatibility, facile surface functionalization, and commercialized synthetic fabrication. We were able to transfer the NDs from large-size agglomerate suspensions to homogenous coatings. ND suspensions have been used in various techniques to coat on commercially available substrates of pure Ti and Si. Scanning electron microscopy (SEM) imaging and nanoindentation show that the densest and strongest coating of NDs was generated when using 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide and N-hydroxysuccinimide (EDC/NHS)-mediated coupling to macroscopic silanized surfaces. In the next step, the feasibility of DNA-mediated coupling of NDs on macroscopic surfaces is discussed using fluorescent microscopy and additional particle size distribution, as well as zeta potential measurements. This work compares different ND coating strategies and describes the straightforward technique of grafting single-stranded DNA onto carboxylated NDs via thioester bridges.

  • Tuning Molecular Solar Thermal properties by modification of a promising norbornadiene photoswitch

    摘要: Carefully targeted modifications to the norbornadiene-quadricyclane couple should give molecules with great potential for solar energy storage. Here we report the synthesis of derivatives of a well-studied compound, 2-cyano-3-anisyl norbornadiene (NBD5). The conjugation was extended by conversion of the nitrile into acrylonitrile and acrylate derivatives over two steps. Despite red-shifting the absorbance properties, the metastable quadricyclanes exhibited extremely short lifetimes. Meanwhile, treatment of NBD5 with acetyl halides in the presence of aluminum trihalides led to the formation of 7-acetyl norbornenes through a Meerwein-Wagner rearrangement. Subsequent transformations made it possible to synthesise norbornadienes with varying degrees of steric bulk at the 7-position of the molecular scaffold. It was found that the bulkier this group was, the more stable the quadricyclane form. This reaction sequence allows for the synthesis of norbornadiene derivatives which are more suited to molecular solar thermal applications in domestic heating despite providing a slightly lower energy density.

  • Novel BODIPY-conjugated amino acids: Synthesis and spectral properties

    摘要: Three BODIPY-based conjugates with lysine, methionine, and tryptophan bound via amino group were synthesized and characterized. Spectral properties of the compounds were studied and a number of photophysical characteristics were obtained. Bioconjugates investigated are water soluble, which makes them promising markers for various bioobjects.

  • (Poly)terephthalates with Efficient Blue Emission in the Solid State

    摘要: We prepared dimethyl and diaryl 2,5-dialkoxyterephthalates from dimethyl 2,5-dihydroxyterephthalate in good-to-high yields via alkylation or a sequence of alkylation, hydrolysis, chlorination, and condensation. The absorption spectra of the dialkoxyterephthalates contain a small band at 332–355 nm, which could be assigned to intramolecular charge-transfer transition from the alkoxy to alkoxycarbonyl groups on the basis of theoretical calculations using density functional theory. The dialkoxyterephthalates exhibited blue fluorescence with moderate-to-excellent quantum yields not only in solution but also in the solid state, such as a poly(methyl methacrylate) (PMMA) film and a powder. The solid-state quantum yields of the diisopropoxy-substituted terephthalates were similar or considerably higher than those of the dimethoxy-substituted counterparts. Copolymerization of 2,5-diisopropoxyterephthaloyl chloride and 1,4-butanediol with or without terephthaloyl chloride gave brilliantly blue fluorescent polymers, whose quantum yields were 0.72 and 0.71 in toluene and 0.46 and 0.40 in the neat film, respectively. Furthermore, white emission was achieved when a fluorescent yellow 2,5-diaminoterephthalate was doped into the thin film of the blue fluorescent polymer at 0.4 wt %.

  • Polymorphic Pure Organic Luminogens with Through Space Conjugation and Persistent Room Temperature Phosphorescence

    摘要: Pure organic luminogens with persistent room temperature phosphorescence (p-RTP) have attracted increasing attention owing to their fundamental significance and potential applications in security inks, bioimaging, and photodynamic therapy. Previous p-RTP luminogens are normally with through bond conjugation. In this work, we report a pure organic luminogen, AN-MA, the Diels-Alder cycloaddition adduct of anthracene (AN) and maleic anhydride (MA), which possesses isolated phenyls and anhydride moiety. AN-MA exhibits aggregation-enhanced emission (AEE) characteristics, with efficiency of ~2% and up to 8.5% in solution and crystals, respectively. Two polymorphs of AN-MA are readily obtained, which can generate UV emission from individual phenyl rings together with bright blue emission owing to the effective through space conjugation. Moreover, p-RTP with lifetime of up to ~1.6 s is obtained in crystals. These results not only disclose a new system with both fluorescence and RTP dual emission, but also suggest an alternative through space conjugation strategy towards pure organic p-RTP luminogens with tunable emissions.

  • Phase Conjugation and Mode Conversion in Stimulated Parametric Down-Conversion with Orbital Angular Momentum: a Geometrical Interpretation

    摘要: We report on an experiment that investigates the spatial mode conversion in the process of parametric down-conversion seeded by a light beam in a superposition of orbital angular momentum modes. This process is interpreted in terms of a geometric representation of first-order spatial modes in a Poincaré sphere, providing an intuitive image of the phase conjugation and the topological charge conservation. We also make a comparison with the analogous phenomenon for optical parametric oscillators.

  • Antimicrobial photodynamic inactivation of fungal biofilm using amino functionalized mesoporus silica-rose bengal nanoconjugate against Candida albicans

    摘要: Candida albicans is an opportunistic fungal pathogen that causes both superficial and systemic infection and an important candidate that contribute to high morbidity and mortality rates in immunocompromised patients. The ability of C. albicans to switch from yeast to filamentous form and thereby forming biofilms make them resistant to most of the antifungal drugs available today. Thus the development of more effective antifungal drugs are essential and crucial at this point of time. Antimicrobial photodynamic therapy is an alternative modality to treat such biofilm forming resistant strains. This study aims to investigate the enhanced efficiency of newly synthesized MSN-RB conjugate as an antimicrobial photosensitizer for antimicrobial photodynamic therapy against C. albicans. Functionalization of MSN with amino groups was performed to increase the dye loading capacity. Conjugation process of MSN-RB was confirmed using different techniques including UV–Vis spectroscopy, Fluorescent spectroscopy and FTIR analysis. A low power green laser 50 mW irradiation was applied (5 min) for activation of MSN-RB conjugate and RB against C. albicans biofilm and planktonic cell. The comparative study of MSN-RB conjugate and free RB on aPDT was evaluated using standard experimental procedures. Antibiofilm efficacy was determined using biofilm inhibition assay, cell viability, EPS quantification and CLSM studies. The results revealed that MSN-RB conjugate has a significant antimicrobial activity (88.62 ± 3.4%) and antibiofilm effect on C. albicans when compared to free dye after light irradiation. The MSN-RB conjugate based aPDT can be employed effectively in treatment of C. albicans infections.

  • Photoimmunoconjugates: novel synthetic strategies to target and treat cancer by photodynamic therapy

    摘要: Photodynamic therapy (PDT) combines a photosensitizer (PS) with the physical energy of non-ionizing light to trigger cell death pathways. PDT has potential as a therapeutic modality to be used in alternative or in combination with other conventional cancer treatment protocols (e.g. surgery, chemotherapy and radiotherapy). Still, due to the lack of specificity of the current PSs to target the tumor cells, several studies have exploited their conjugation with targeting moieties. PSs conjugated with antibodies (Abs) or their fragments, able to bind antigens overexpressed in the tumors, have demonstrated potential in PDT of tumors. This review provides an overview of the most recent advances on photoimmunoconjugates (PICs) for cancer PDT, which involve the first and second-generation PSs conjugated to Abs. This is an update of our previous review “Antibodies armed with photosensitizers: from chemical synthesis to photobiological applications”, published in 2015 in Org. Biomol. Chem.

  • Autofocusing with out-of-band phase conjugation

    摘要: Electromagnetic phase conjugation provides an appealing path towards communication and target localization in dynamic media such as the Earth’s atmosphere. However, realizing phase conjugation-based devices has proven difficult because of the inherent complexity of systems that employ receivers and transmitters working on the same band. In this letter we propose the concept of out-of-band phase conjugation in which the frequency of the phase conjugated wave is an integer multiple of the incident wave frequency. Remarkably, the out-of-band phase conjugated wave maintains the autofocusing property that makes phase conjugation so powerful and opens the door to imaging technology effective in random and dynamic media.

  • Site-specific chelator-antibody conjugation for PET and SPECT imaging with radiometals

    摘要: Antibodies and their derivatives radiolabelled with positron- and gamma-emitting radiometals enable sensitive and quantitative molecular Positron Emission Tomography (PET) and Single Photon Emission Computed Tomography (SPECT) imaging of antibody distribution in vivo. Chelators that are covalently attached to antibodies allow radiolabelling with metallic PET and SPECT radioisotopes. Conventional strategies for chelator-protein conjugation generate heterogeneous mixtures of bioconjugates that can exhibit reduced affinity for their targets, and undesirable biodistribution and pharmacokinetics. Recent advances in bioconjugation technology enable site-specific modification to generate well-defined constructs with superior properties. Herein we survey existing site-specific chelator-protein conjugation methods. These include chelator attachment to cysteines/disulfide bonds or the glycan region of the antibody, enzyme-mediated chelator conjugation, and incorporation of sequences of amino acids that chelate the radiometal. Such technology will allow better use of PET and SPECT imaging in the development of antibody-based therapies.
