FRET-based dual channel fluorescent probe for detecting endogenous/exogenous H2O2/H2S formation through multicolor images
摘要: We have developed a FRET-based fluorescent probe (PHS1) as a combination of two different fluorophores (coumarin and naphthalimide); which can detect both exogenous and endogenous H2S and H2O2 in live cells through multicolor images. The precise overlap between UV-absorption of naphthalimide and the emission band of coumarin in probe PHS1 allows the acquisition of the self-calibrated information of dual analytes through FRET-based imaging. The UV–Vis absorption (λabs 390 nm) and fluorescence emission (λem 460 nm) of probe PHS1 in the presence of H2O2 are increased ∽35- fold and ∽15-fold respectively. It also allows the estimation of the levels of H2S through enhancement of emission intensity at 550 nm. The probe PHS1 exhibits high stability against various analytes, including various pH (4–9.5). The cell viability assay data indicate that the probe is not harmful to the cancer cells. The nontoxic nature of the probe PHS1 encourages application for cancer cell labeling. The probe PHS1 can detect the level of endogenous H2O2, H2S, and H2O2/H2S in cancer cells through blue, green and FRET-based green channel imaging. PHS1 is a unique probe, has potential application for diagnosing cancer by providing information on the level of dual analytes (H2S, H2O2) in cancer cells.

Novel rhodamine-based colorimetric and fluorescent sensor for the dual-channel detection of Cu2+ and Co2+/trivalent metal ions and its AIRE activities
摘要: A rhodamine hydrazone 1 bearing coumarin moiety was designed and prepared. Compound 1 exhibited high selectivity toward Co2+ and trivalent metal ions with fluorescence enhancement in CH3OH solution. However, 1 selectively responded to Al3+ in nearly pure H2O media and was further applied to monitor Al3+ in live cells. Moreover, 1 could also act as a colorimetric probe toward Cu2+ in either CH3OH or H2O solution. In addition, sensor 1 displayed aggregation-induced ratiometric emission (AIRE) activities in mixed H2O/CH3OH solution.

Photostable coumarin containing azo dyes with multifunctional property
摘要: A series five coumarin azo disperse dyes with thiophene bridge are synthesized using diazotized aniline derivatives which are confirmed by FT-IR, 1H NMR, 13C NMR, and CHN analysis. The dyes are applied on polyester and nylon material. The multifunctional properties (lightfastness, washing fastness, sublimation fastness, K/S, UPF, and antimicrobial activity) of the dyed fabric are determined. The dye 3a (-NO2) show red shifted absorption (649 nm) and blocking 95.5-97.4% of UV radiations with 96% antimicrobial property. The geometries of all the azo dyes are optimized using Density Functional Theory (DFT). The Global Reactivity Descriptors (GRD) are evaluated at the same method of theory. The GRD obtained shows a linear relation with lightfastness and UPF ratings. The antimicrobial activity of all synthesized dyes are determined by AATCC 100 test method and it is correlated with the HOMO-LUMO energy band gap.

New Coumarin-benzoxazole derivatives: Synthesis, photophysical and NLO properties
摘要: In this work, a series of novel D-π-A structure coumarin-benzoxazole derivatives have been synthesized. The third-order NLO properties of the coumarin derivatives in DMSO are investigated by Z-scan measurement, and the photophysical properties of the compounds are studied through experimental and theoretical calculation methods. The results showed that the introducing specific functional groups at multiple active sites of coumarin reduced the π-π intermolecular stacking, while the formation of push-pull structure decreased E-gap of entire molecule. The excellent third-order NLO responses based on chromophore 4c with maximum nonlinear absorptive coefficients is 50.64 × 10-13 esu and the largest second-order NLO hyperpolarizability is 7.12 × 10-30 esu. Experimental results have also been confirmed through DFT, TDDFT and ZINDO calculations. Our results indicate that the coumarin fluorescent dyes are promising candidate materials for NLO applications.

Fluorescent sensor for copper(II) ion based on coumarin derivative and its application in cell imaging
摘要: A novel fluorescent sensor CHT was designed based on the excellent optical properties of coumarin derivatives. It was synthesized by Schiff base reaction connected by 7-(N,N-diethylamino) coumarin-3-aldehyde and 2-hydrazinobenzothiazole. The sensor CHT displayed exhibited a significant fluorescence quenching at 540 nm upon the addition of Cu2+ ions. And the sensor exhibited good sensitivity, fast response time, high selectivity for Cu2+ ions in the presence of other important relevant metal ions. In addition, CHT fluorescent sensor was successfully applied for fluorescent imaging of Cu2+ ions in A549 and MCF-7 cells, demonstrating its potential applications in live cell imaging.

A coumarin-based colorimetric fluorescent probe for rapid response and highly sensitive detection of hydrogen sulfide in living cells
摘要: Hydrogen sulfide (H2S) plays a vital role in numerous biological processes in living organisms. To better understand its functions, a fluorescent probe to fast and sensitively detect H2S is imminently needed. Keep this in mind, we reasonably designed probe DHC for detecting H2S based on α, β-unsaturated ethanoylcoumarin fluorophore. The limit of detection (LOD) is found to be as low as 5 × 10?8 M, which is superior to most reported fluorescent probes to detect H2S. Furthermore, the wide pH range of 4–11 makes it capable of application in biological systems. Most importantly, MTT assays and cell imaging experiments indicate that probe DHC has hypotoxicity and outstanding membrane permeability, which makes DHC successful imaging of H2S in Baby Hamster Syrian Kidney (BHK) cells.

Prediction of coumarin and ethyl vanillin in pure vanilla extracts using MID-FTIR spectroscopy and chemometrics
摘要: Fourier transform mid-infrared (MID-FTIR) spectroscopy coupled with chemometric analysis was used to identify and quantify coumarin (CMR) and ethyl vanillin (EVA) adulterations in pure vanilla extracts. Forty samples adulterated with CMR (0.25-10 ppm) and forty with EVA (0.25-10%) were prepared from pure vanilla extracts and characterized by MID-FTIR spectroscopy to develop chemometric models. Additionally, six commercial vanilla samples were analyzed. A soft independent modeling of class analogy (SIMCA) model was developed to identify and classify the purity from EVA-adulterated or CMR-adulterated samples. Prediction models for CMR or EVA content were developed using the principal component regression (PCR), partial least squares with single y-variables (PLS1), and with multiple y-variables (PLS2) algorithms. Moreover, the predictions of the best quantification chemometric model were compared with the results of a high-performance liquid chromatography-diode array detector (HPLC-DAD) method to evaluate the accuracy of the prediction. The PLS1 algorithm had better performance using 3 and 8 factors for EVA and CMR, respectively. The SIMCA model showed 100% recognition and rejections rates. The results demonstrate that adulteration of pure vanilla with EVA and CMR could be successfully predicted by the developed technique.

Fluorescence Quenching-Based Mechanism for Determination of Hypochlorite by Coumarin-Derived Sensors
摘要: A fluorescence quenching-based mechanism for the determination of hypochlorite was proposed based on spectroscopic and chromatographic studies on the hypochlorite-sensing potency of three structurally similar and highly fluorescent coumarins. The mode of action was found to rely upon a chlorination of the coumarin-based probes resulting from their reaction with sodium hypochlorite. Importantly, the formation of chlorinated derivatives was accompanied by a linear decrease in the fluorescence intensities of the probes tested. The results obtained suggest the applicability of a coumarin-dependent hypochlorite recognition mechanism for the detection of, as well as for quantitative determination of, hypochlorite species in vitro.

Quantum chemical investigation of modified coumarin-based organic efficient sensitizers for optoelectronic applications
摘要: One of the most major features in the improvement of dye-sensitized solar cells (DSSCs) is the survey and plan of high-efficiency and low-cost dyes. In this paper, a new series of metal-free coumarin-based (NKX-2311) dye molecules, namely (D1–D4) are investigated for use in DSSC applications. The dye molecules contain three parts, electron-donor (D) connected by the π-conjugated linker as a spacer and electron-acceptor (A)/anchoring group. Molecular structure, electronic absorption spectra and photovoltaic (PV) parameters of the D1–D4 dyes have been performed by using density functional theory (DFT) and time-dependent DFT (TD-DFT) approaches are employed. The effects of chemical modification on the spectra and PV properties of the coumarin-modified organic dyes have been revealed. The performance of three functionals (B3LYP, CAM-B3LYP and ωB97XD) has been evaluated to get an experimental absorption wavelength of NKX-2311. From the functionals, B3LYP method was better matched with the absorption wavelength of NKX-2311. Therefore, D1–D4 dye molecules excited state calculations of the absorption spectra have been investigated by using B3LYP functional with 6-31G(d) basis set. The results show that D1, D3 dyes are smaller energy gap and absorption spectra are systematically red-shifts. The theoretical results have a display that the lowest occupied molecular orbitals (LUMOs) and highest occupied molecular orbitals (HOMOs) of the D1–D4 dyes can be confirming positive response on the electron injection and dye regeneration process. The NLO property of the D1–D4 dyes can be derived as polarizability and first-order hyperpolarizability. The calculated values of D1 and D3 dyes are the best applicants for NLO performance. The conclusion of this study will deliver a useful reference to the future proposal and beneficial to get DSSCs with superior power conversion efficiency (PCE).

Coumarin Caged Compounds of 1-Naphthaleneacetic Acid as Light-Responsive Controlled-Release Plant Root Stimulators
摘要: Six coumarin caged compounds of NAA comprising different substituents on the coumarin moiety were synthesized and evaluated for their photo-physical and chemical properties as light-responsive controlled-release plant root stimulators. The 1H-NMR and HPLC techniques were used to verify the release of NAA from the caged compounds. After irradiation at 365 nm, the caged compounds exhibited the fastest release rate at t1/2 of 6.7 days and the slowest release rate at t1/2 of 73.7 days. Caged compounds at high concentration (10-5 M and 10-6 M) significantly stimulate secondary root germination while free NAA at the same level is toxic and leads to inhibition of secondary root germination. Cytotoxicity of the caged compounds against Fibroblasts and Vero cells were evaluated and the results suggested that at 10-5 – 10-6 M, caged compounds exhibit no significant cytotoxic to the cells. Thus, the caged compounds of NAA in this study can be of great benefits as efficient agrochemicals.
