  • Highly sensitive luminescent probe of aniline and trace water in organic solvents based on covalently modified lanthanide metal-organic frameworks

    摘要: A novel europium functionalized MOF named Eu3+@UiO-66-NH2-IM is robustly synthesized via covalent PSM (post-synthetic modification). The ligand of matrix MOF is covalent modified via imidazole-2-carboxaldehyde after the UiO-66-NH2 is synthesized. The successful transformation of amino groups to imine groups is confirmed by FTIR analysis. The modified MOF named UiO-66-NH2-IM shows outstanding crystalline stability. After Eu3+ doped into UiO-66-NH2-IM, the as-prepared fluorescent MOF displays characteristic emission line of Eu3+. Discussion of the tunable emission color of the modified MOF is included and white light integration could be achieved through altering the ratio of Eu3+ doped into the MOF. What’s more, the luminescent transparent film based on Eu3+@UiO-66-NH2-IM can emit white light under proper excitation wavelength, which may be helpful to some potential applications in lighting. In addition, Eu3+@UiO-66-NH2-IM shows different emission response in water and organic solvents, and it is further developed into sensitive probe for the detection of water content in organic solvents (ethanol, DMF) with low detection limits ( 0.088%, and 0.054%, respectively) by utilizing its quenching behavior in water. Meanwhile, Eu3+@UiO-66-NH2-IM displays greatly selective and sensitive response for detecting aniline with a low detection limit (0.072%), a broad linear range (0-3.75%), and the signal of the detection is obvious which can be seen clearly under the irradiation of UV light.

  • A curcumin-based NIR fluorescence probe for detection of amyloid-beta (Aβ) plaques in Alzheimer's disease

    摘要: Amyloid-beta (Aβ) peptide as one of the main components of senile plaques is closely related to the onset and progression of incurable Alzheimer's disease (AD). In recent years, it is reported that curcumin derivatives have been used as the near-infrared (NIR) fluorescence imaging probes of Aβ plaques for the early diagnosis of AD. To further develop a curcumin-based NIR fluorescent probe for Aβ plaques, in this work, we have synthesized three novel curcumin-based NIR Dyes. Among them, the curcumin derivative Dye 2 showed a significant enhancement in its fluorescence intensity (λem, 635 nm; 19.5-fold increase in quantum yield, Φ = 0.36; Kd, 1.13 μM) after binding to Aβ plaques. Additionally, in vitro and in vivo fluorescence imaging of Aβ plaques strained with Dye 2 confirmed that the compound was a potential probe to detect Aβ plaques in AD. This work opens a perspective to rationally design novel curcumin-based NIR compound for Aβ detection.

  • Visualizing Endogenous Sulfur Dioxide Derivatives in Febrile Seizure-Induced Hippocampal Damage by a Two-Photon Energy Transfer Cassette

    摘要: Febrile seizure (FS), a frequently encountered seizure disorder in pediatric populations, can cause hippocampus damage. It has been elucidated that sulfur dioxide (SO2) content is overproduced during the development of FS and related brain injury. Thus, in situ monitoring the level of endogenous SO2 in FS-related models is helpful to estimate the pathogenesis of FS-induced brain injury, but effect detection method remains to be explored. Herein, we developed a two-photon energy transfer cassette based on acedan-anthocyanidin scaffold, TP-Ratio-SO2, enabling to achieve this purpose. TP-Ratio-SO2 specifically responds to SO2 derivatives (HSO3-/SO32-) in ultrafast fashion (less than 3s), and HSO3-/SO32- can be sensitively determined with a detection limit of 26 nM. Moreover, it exhibits significant changes in two well-resolved ?uorescence emission (?λ = 140 nm) by reacting with HSO3-/SO32-, behaving as a ratiometric fluorescent sensor. Importantly, ratiometric imaging of endogenous SO2 derivatives generation in hyperpyretic U251 cells and as well as in rat model of FS-treated hippocampus damage were successfully carried out by TP-Ratio-SO2, demonstrating that it may be a promising tool for studying the role of SO2 in FS-associated neurological diseases.

  • A cyclometalated iridium(III) complex-based fluorescence probe for hypochlorite detection and its application by test strips

    摘要: A new cyclometalated iridium(III) complex-based fluorescence probe (IrCN) for hypochlorite (ClO?) has been synthesized and characterized. The probe displayed nonfluorescent around 577 nm, while a 54-fold enhancement in fluorescence emission intensity was observed after the addition of ClO? due to the removal of C=N isomerization effect. Such “turn-on” fluorescence probe worked excellently in wide pH range (5–12) with short response time (< 20 s) and the detection limit was as low as 0.11 μM. In addition, IrCN exhibited high selectivity even in the presence of other competing species. Furthermore, IrCN was successfully integrated in towards ClO? fluorescent test strips for real-time detection of ClO?.

  • A water-soluble benzoxazole-based probe: real-time monitoring PPi via situ reaction by two-photon cells imaging

    摘要: Pyrophosphate (PPi) played crucial roles in various fundamental physiological processes. Herein, a two-photon absorption (TPA) “On-Off-On” type benzoxazole-based fluorescence probe BN was designed and synthesized, which detected PPi through Cu2+ displacing method in situ system in aqueous medium. The on-off-on process of BN recognizing PPi was verified by mass spectra and theoretical calculations, which was successfully applied in TPA cells imaging.

  • Colorimetric and NIR fluorescence probe with multiple binding sites for distinguishing detection of Cys/Hcy and GSH in vivo

    摘要: Although some progress has been made in distinguishing detection of biothiols, NIR biothiol fluorescent probes for simultaneous distinguishing detection of Cys, Hcy and GSH in vivo have not been reported. The design of these probes involves the introduction of NIR fluorophores and multiple binding sites, so the integrated design of probes remains a challenge. Although Cys, Hcy and GSH have the common functional group: a sulfydryl group and an amino group, due to their differences in spatial structure, they may react with multiple binding sites probes to produce different reaction products in different bonding mechanisms, resulting in the different colors and fluorescent signals changes of the system. Therefore, multiple binding sites fluorescent probes can realize their discrimination detection. For NIR fluorescent probe, it is easier to realize in vivo imaging to promote the research of biothiols in clinical diagnosis. In our work, not only multiple binding sites were constructed in the compound, but also NIR fluorophores were introduced. This enables the probe to not only efficiently distinguish detection of Cys/Hcy and GSH but also achieve fluorescence imaging in vivo. We believe this result is a milestone in the discrimination detection of biothiols.

  • Visual colorimetric and fluorescence turn-on probe for Cu(II) ion based on coordination and catalyzed oxidative cyclization of ortho amino azobenzene

    摘要: 2,2′-diaminoazobenzene (L1), a visual colorimetric and fluorescence turn-on detection probe for Cu2+ ion, has been discovered successfully. The yellow L1 solution changed into light-purple solution rapidly with dropping of Cu2+ ion observed by the naked eyes, for the formation of 1:1 complex of L1 and Cu2+ ion calculated by Job-plot method. Non-emissive L1 transformed into 2-(2-aminophenyl) benzotriazole (L1-BTA) with strong fluorescence at the peak around 475 nm, owing to the Cu2+ catalyzed oxidative cyclization of L1, accompanied by the fluorescence intensity gradually increasing within 20 min. And L1 was applied for quantitative detection of Cu2+ with good linear relationship between its UV absorbance and Cu2+ concentration, and exhibited high fluorescence selectivity toward Cu2+ over other metal ions (Fe3+, Zn2+, Cd2+, Co2+, Ni2+, Mn2+, Na+, Al3+ and Ca2+) in EtOH solution.

  • A novel OFF-ON-OFF fluorescence probe based on coumarin for Al3+ and F? detection and bioimaging in living cells

    摘要: A novel fluorescence probe L2 based on coumarin has been designed and synthesized. The probe L2 can be used for relay recognition of metal ions Al3+ and anion F? in the aqueous HEPES buffer (0.05 M, pH=7.4), and build a OFF-ON-OFF detection system. The probe showed high selectivity and sensitivity to target ions in the process of relay recognition, and the corresponding detection limit could be as low as 0.014 μM (Al3+) and 0.03 μM (F?). Besides, the geometry optimizations of probe L2 and [L2+Al3+] complex were carried out using the Gaussian 16 program based on DFT, and the identification mechanism of the probe was also discussed by the mass spectrometry and theoretical calculations. Moreover, the probe have also been successfully applied to detection of target ions in living cells.

  • A simple and rapid fluorescent approach for flavonoids sensor based on gold nanoclusters

    摘要: The development of simple, easy-to-operate and real-time detection methods for the active ingredients in herbal medicines has aroused growing interest owing to their pivotal health benefits. In this study, a qualitative and quantitative detection method for the flavonoids was developed based on the specific interaction between flavonoids and bovine serum albumin (BSA). A fluorescent gold nanocluster was imbedded into the cavity enclosed in the tertiary structure of BSA, the fluorescence of which can be quenched by the flavonoids with fast response (< 5 s). This decrease in fluorescence intensity of BSA-AuNCs as output signal enables the real-time visual inspection of flavonoids. We demonstrated that the present approach was capable of detecting quercetin in serum, plasma, and monitoring the content of flavonoids in proprietary Chinese medicine Rutin Tablets. BSA-AuNCs was the first fluorescent probe for the specific determination of active ingredients in herbal medicines. Hence the reported protein-AuNCs sensing platform can serve as a convenient detection strategy in pharmaceutical analysis.
