A RAGE-Targeted Antibody-Drug Conjugate: Surface Plasmon Resonance as a Platform for Accelerating Effective ADC Design and Development
摘要: Antibodies, antibody-like molecules, and therapeutics incorporating antibodies as a targeting moiety, such as antibody-drug conjugates, offer significant potential for the development of highly efficacious drugs against a wide range of disorders. Despite some success, truly harnessing the superior targeting properties of these molecules requires a platform from which to effectively identify the best candidates for drug development. To streamline the development of antibody-drug conjugates targeting gynecological cancers within our laboratory, we incorporated surface plasmon resonance analysis (Biacore? T200) into our development toolkit. Antibodies, selected based on positive ELISA screens as suitable for development as antibody-drug conjugates, were evaluated using surface plasmon resonance to determine a wide range of characteristics including specificity, kinetics/affinity, the effect of linker binding, the impact of the drug to antibody ratio, and the effect of endosomal pH on antibody-antigen binding. Analysis revealed important kinetics data and information regarding the effect of conjugation and endosomal pH on our antibody candidates that correlated with cell toxicity and antibody internalization data. As well as explaining observations from cell-based assays regarding antibody-drug conjugate efficacies, these data also provide important information regarding intelligent antibody selection and antibody-drug conjugate design. This study demonstrates the application of surface plasmon resonance technology as a platform, where detailed information can be obtained, supporting the requirements for rapid and high-throughput screening that will enable enhanced antibody-drug conjugate development.

Molecularly imprinted based surface plasmon resonance nanosensors for microalbumin detection
摘要: Human serum albumin (HSA) is a major blood plasma protein also found in urine where its existence may be a marker of some types of liver or kidney dysfunction. Herein, we fabricated a novel surface plasmon resonance (SPR) nanosensor for selective, sensitive, and label-free microalbumin detection both in aqueous and urine sample solutions. Firstly, HSA-imprinted nanoparticles were synthesized, which consist of ethylene glycol dimethacrylate and N-methacryloyl-L-leucine as a cross-linker and functional monomer. The nanoparticles were characterized by zeta-size and scanning electron microscope analyses and were dropped onto the SPR chip surface to make HSA sensitive nanosensor. Characterization studies of HSA-imprinted SPR chip were carried out by atomic force microscopy, Fourier-transform infrared spectroscopy, contact angle, and ellipsometer. The limit of detection and limit of quantification values of HSA-imprinted SPR nanosensor were calculated as 0.7 pM and 1.9 pM for the concentration range of 0.15-500 nM. Selectivity studies of HSA-imprinted SPR nanosensor were achieved with hemoglobin and transferrin proteins which were chosen as competitor molecules. HSA-imprinted SPR nanosensor was displayed highly selective and sensitive to HSA.

Plasmon-enhanced upconversion luminescence in pyrochlore phase Yb<sub>x</sub>Er<sub>2-x</sub>Ti<sub>2</sub>O<sub>7</sub> thin film
摘要: Pyrochlore phase YbxEr2-xTi2O7 (YETO) thin films have been prepared by employing a facile sol-gel method combining with spin-coating technique and post-annealing treatment at 700 ℃. High concentration of Yb3+ ions can promote the transformation from Yb3+/Er3+ co-doped anatase phase TiO2 to pyrochlore phase YETO at 700 ℃ temperature. We find that the YETO thin film with 30 mol% Yb3+ ions exhibits the brightest upconversion (UC) emission. Moreover, introduction of Au nanorods (Au NRs) in the YETO thin film can further enhance the UC fluorescence. By adjusting the density of Au NRs, the UC emission intensity is increased by about 2.8-fold due to the excitation field enhancement caused by the localized surface plasmon resonance effect.

Cu nanoclusters in ion exchanged soda-lime glass: Study of SPR and nonlinear optical behavior for photonics
摘要: Nanomaterials with large optical nonlinearities have received considerable attention in the field of modern science and nanotechnology. In this paper, we have studied nonlinear optical and surface plasmon resonance properties and behavior of Cu nanoclusters formed in ion exchanged soda-lime glass. The soda-lime glasses were successfully doped with Cu nanoclusters in the ion-exchange process. The size of the clusters estimated from the optical absorption spectroscopy results closely matched with those obtained from the transmission electron microscopy data. The results revealed that spherical shaped Cu nanoclusters were homogeneously distributed in the glass matrix and the size of the Cu nanoclusters varied from 4 to 10 nm. The structure and chemical state were further analyzed by X-ray diffraction and X-ray photoelectron spectroscopy. The nonlinear optical behavior of the materials was analyzed using femtosecond Z-scan technique. The nonlinear refraction index (n2), nonlinear absorption coefficient (β) and the third-order nonlinear optical susceptibility (χ(3)) were estimated to be -1.72 × 10?17 m2/W, 9.96 × 10?11 m/W, and 0.56 × 10?11 esu, respectively, which shows possible application in the field of photonics.

Plasmonic Metasurfaces with Tunable Gap and Collective SPR Modes
摘要: Optical properties of a plasmonic metasurface made of a monolayer of gold nanoparticles in close proximity to an aluminum thin film were studied numerically and experimentally. Extinction spectra of the plasmonic metasurface were studied as functions of the thickness of a dielectric spacer between the monolayer of gold nanoparticles and the aluminum film in the visible wavelength range. The goal was to understand the excitation of a collective surface plasmon resonance (SPR) mode and a gap plasmon mode as well as their dependence on the spacer thickness, nanoparticles spacing and their size. By using finite-difference-time-domain (FDTD) calculations we find that the SPR extinction peak first red-shifts and then splits into two peaks. The first extinction peak is associated with the collective SPR mode of the monolayer and it shifts to shorter wavelengths as the spacer layer decreases. As the spacer layer decreases from 35 nm to 7.5 nm, the second peak gradually appears in the extinction spectra of the metasurface. We assign the second peak to the gap mode. The gap mode first appears at around 620 nm or greater and it shifts to larger wavelength for larger nanoparticle spacing and size. The FDTD simulations are confirmed by an experimental examination of the dispersion curves of a similar multilayer system. The computational results match the experimental results and confirm the excitation of the two modes.

Laser-driven structural transformations in dextran- <i>graft</i> -PNIPAM copolymer/Au nanoparticles hybrid nanosystem: the role of plasmon heating and attractive optical forces
摘要: Laser induced structural transformations in a dextran grafted-poly(N-isopropylacrylamide) copolymer/Au nanoparticles (D-g-PNIPAM/AuNPs) hybrid nanosystem in water have been observed. The laser induced local plasmonic heating of Au NPs leads to Lower Critical Solution Temperature (LCST) phase transition in D-g-PNIPAM/AuNPs macromolecules accompanied by their shrinking and aggregation. The hysteresis non-reversible character of the structural transformation in D-g-PNIPAM/AuNPs system has been observed at the decrease of laser intensity, i.e. the aggregates remains in solution after the turn-off the laser illumination. This is an essential difference comparing to the case of usual heating–cooling cycles when there is no formation of aggregates and structural transformations are reversible. Such a fundamental difference has been rationalized as the result of action of attractive optical forces arising due to the excitation of surface plasmons in Au NPs. The attractive plasmonic forces facilitate the formation of the aggregates and counteract their destruction. The laser induced structural transformations have been found to be very sensitive to matching conditions of the resonance of the laser light with surface plasmon resonance proving the plasmonic nature of observed phenomena.

Tailoring optical cross sections of gold nanorods at a target plasmonic resonance wavelength using bromosalicylic acid
摘要: In many applications, the optical cross sections of gold nanorods (AuNRs) are required to be tailored at a fixed target longitudinal surface plasmon resonance (LSPR) wavelength depending on the excitation source and the photodetector. In this work, we demonstrate the fine tailoring of optical cross sections of AuNRs at a fixed target resonance wavelength, on the basis of AuNR overgrowth using a binary surfactant mixture consisting of 5-bromosalicylic acid (BSA) and cetyltrimethylammonium bromide (CTAB). A systematic study was performed on the sum effects of the BSA concentration and the volume of the growth solution, which gives a formula for quantitative instructions. Based on the formula, we gave examples for the successful synthesis of AuNRs with different optical cross sections at target LSPR wavelengths. From simulation, a nonlinear relationship was further derived to understand the relationship between the aspect ratio and the width of the AuNRs at a target LSPR wavelength for the dimension design of AuNRs. The ratio of optical against physical cross sections was calculated and plotted as a function of the width. The results clearly indicate that AuNRs with a width of 30 nm possess the highest efficiency in terms of optical per physical cross section. Our study provides reliable methods for the synthesis, as well as guidelines for the dimension design of AuNRs, for use in a variety of applications.

Study of the Effect of Optical Illumination on Resistive Switching in ZrO2(Y) Films with Au Nanoparticles by Tunneling Atomic Force Microscopy
摘要: The effect of optical illumination on the resistive switching in ultrathin (~4 nm) ZrO2(Y) films with embedded single-layer Au nanoparticle arrays 2–3 nm in size is studied via tunneling atomic force microscopy. The ZrO2(Y) films with Au nanoparticles are grown by layerwise magnetron deposition onto glass substrates with a conductive indium-tin-oxide sublayer, followed by annealing at 450°C. An increase in hysteresis due to bipolar resistive switching in the ZrO2(Y) films is observed on the cyclic current–voltage curves of the microscope probe-to-sample contact. The effect is found to manifest itself in a dense Au nanoparticle array (~660 nm) when the contact area is photoexcited through a transparent substrate exposed to the radiation of a semiconductor laser at the plasmon-resonance wavelength. The effect is attributed to the photon-assisted field emission of electrons from Au nanoparticles to the conduction band of ZrO2(Y) in a strong electric field between the microscope probe and the indium-tin-oxide substrate under plasmon-resonance conditions.

LSPR Biosensing Approach for the Detection of Microtubule Nucleation
摘要: Microtubules are dynamic protein ?laments that are involved in a number of cellular processes. Here, we report the development of a novel localized surface plasmon resonance (LSPR) biosensing approach for investigating one aspect of microtubule dynamics that is not well understood, namely, nucleation. Using a modi?ed Mie theory with radially variable refractive index, we construct a theoretical model to describe the optical response of gold nanoparticles when microtubules form around them. The model predicts that the extinction maximum wavelength is sensitive to a change in the local refractive index induced by microtubule nucleation within a few tens of nanometers from the nanoparticle surface, but insensitive to a change in the refractive index outside this region caused by microtubule elongation. As a proof of concept to demonstrate that LSPR can be used for detecting microtubule nucleation experimentally, we induce spontaneous microtubule formation around gold nanoparticles by immobilizing tubulin subunits on the nanoparticles. We ?nd that, consistent with the theoretical model, there is a redshift in the extinction maximum wavelength upon the formation of short microtubules around the nanoparticles, but no signi?cant change in maximum wavelength when the microtubules are elongated. We also perform kinetic experiments and demonstrate that the maximum wavelength is sensitive to the microtubule nuclei assembly even when microtubules are too small to be detected from an optical density measurement.

Plasmonic Nanocavity Metasurface Based on Laser-Structured Silver Surface and Silver Nanoprisms for the Enhancement of Adenosine Nucleotide Photoluminescence
摘要: A reliable photoluminescence (PL) spectroscopy and imaging of biomolecules at room temperature is a challenging and important problem of biophysics, biochemistry, and molecular genetics. A unique effect of strong plasmonic enhancement of the PL by metal nanostructures is one of the most effective approaches for this purpose. The highest enhancement is provided by metal nanostructures with densely packed sharp tips, periodically arranged metal nanostructures, and plasmonic cavities. All of these features have been realized in the plasmonic cavity metasurface based on the silver (Ag) laser-induced periodic surface structure and Ag triangular nanoprisms studied in the present work. The strong plasmon-enhanced PL of 5′-deoxyadenosine monophosphate deposited on such metasurfaces has been revealed at room temperature. The observed enhancement of more than 1000-fold has been interpreted as a result of synergetic action of the generation of a high concentration of hot spots near the sharp edges of the laser-induced surface structure and nanoprisms together with excitation of the collective gap mode of the cavity due to strong near-field plasmonic coupling. Correspondingly, the plasmonic cavity metasurfaces consisting of metal laser-induced periodic surface structures and nonspherical metal nanoparticles with sharp edges have been shown to be crucial for the highly sensitive detection and imaging of biomolecules at room temperature without consuming any dye labels.
