  • Application of experimental design and multivariate analysis in the on-line reaction monitoring of a Suzuki cross-coupling reaction by Raman spectroscopy and multivariate analysis

    摘要: A Suzuki-coupling reaction for the construction of CeC bond was monitored using Raman spectroscopy. The effects of temperature and concentration on the reaction yield were investigated following a central composite design. The percent yield for each data was measured off-line using GCeMS. On-line monitoring reactions using Raman spectroscopy coupled with partial least squares (PLS) were used for building a prediction model. Raman spectra collected from five experiments at different sampling times were used in building the model. The PLS model was then validated and results showed that the yield of reaction can be predicted well with high accuracy by the model, both within and outside the experimental limits. The results presented here have shown that such model is well suited for monitoring Suzuki-coupling reactions that are widely used in chemical and pharmaceutical industries.

  • Raman Spectroscopic Analysis of Signaling Molecules–Dopamine Receptors Interactions in Living Cells

    摘要: The selective interaction of signaling compounds including neurotransmitters and drugs with the dopamine receptors (DARs) is extremely important for the treatment of neurodegenerative diseases. Here, we report a method to probe the selective interactions of signaling compounds with D1 and D2 DARs in living cells using the combined approach of theoretical calculation and surface-enhanced Raman spectroscopy (SERS). When signaling compounds such as DA, amphetamine, methamphetamine, and methylenedioxypyrovalerone interact with D1 dopamine receptors (DRD1), the intracellular cyclic adenosine monophosphate (cAMP) level is increased. However, the intracellular level of cAMP is decreased when D2 dopamine receptors (DRD2) interact with the abovementioned signaling compounds. In our experiments, we have internalized the silica-coated silver nanoparticles (AgNP@SiO2) in living cells to adsorb biologically generated cAMP which was probed by using SERS. Besides adsorptions of cAMP, AgNP@SiO2 has a crucial role for the enhancement of Raman cross section of the samples. We observed the characteristic SERS peaks of cAMP when DRD1-overexpressed cells interact with the signaling compounds; these peaks were not observed for other cells including DRD2-overexpressed and DRD1?DRD2-coexpressed cells. Our experimental approach is successful to probe the intracellular cAMP and characterize the selectivity of signaling compounds to different types of DARs. Furthermore, our experimental approach is highly capable for in vivo studies because it can probe intracellular cAMP using a low input power of incident laser without significant cell damage. Our experimental results and density functional theory calculations showed that 780 and 1503 cm?1 are signature Raman peaks of cAMP. The SERS peak at 780 cm?1 is associated with C?O, C?C, and C?N stretching and symmetric and asymmetric bending of two O?H bonds of cAMP, whereas the SERS peak at 1503 cm?1 is contributed by the O9?H3 bending mode.

  • Improving clinical diagnosis of early-stage cutaneous melanoma based on Raman spectroscopy

    摘要: BACKGROUND: Clinical diagnosis of early melanoma (Breslow thickness less than 0.8 mm) is crucial to disease-free survival. However, it is subjective and can be exceedingly difficult, leading to missed melanomas, or unnecessary excision of benign pigmented skin lesions. An objective technique is needed to improve the diagnosis of early melanoma. METHODS: We have developed a method to improve diagnosis of (thin) melanoma, based on Raman spectroscopy. In an ex vivo study in a tertiary referral (pigmented lesions) centre, high-wavenumber Raman spectra were collected from 174 freshly excised melanocytic lesions suspicious for melanoma. Measurements were performed on multiple locations within the lesions. A diagnostic model was developed and validated on an independent data set of 96 lesions. RESULTS: Approximately 60% of the melanomas included in this study were melanomas in situ. The invasive melanomas had an average Breslow thickness of 0.89 mm. The diagnostic model correctly classified all melanomas (including in situ) with a specificity of 43.8%, and showed a potential improvement of the number needed to treat from 6.0 to 2.7, at a sensitivity of 100%. CONCLUSION: This work signifies an important step towards accurate and objective clinical diagnosis of melanoma and in particular melanoma with Breslow thickness <0.8 mm.

  • Raman spectroscopic detection of high-grade cervical cytology: Using morphologically normal appearing cells

    摘要: This study aims to detect high grade squamous intraepithelial cells (HSIL) by investigating HSIL associated biochemical changes in morphologically normal appearing intermediate and superficial cells using Raman spectroscopy. Raman spectra (n = 755) were measured from intermediate and superficial cells from negative cytology ThinPrep specimens (n = 18) and from morphologically normal appearing intermediate and superficial cells from HSIL cytology ThinPrep specimens (n = 17). The Raman data was subjected to multivariate algorithms including the standard principal component analysis (PCA)-linear discriminant analysis (LDA) and partial least squares discriminant analysis (PLS-DA) together with random subsets cross-validation for discriminating negative cytology from HSIL. The PCA-LDA method yielded sensitivities of 74.9%, 72.8%, and 75.6% and specificities of 89.9%, 81.9%, and 84.5%, for HSIL diagnosis based on the dataset obtained from intermediate, superficial and mixed intermediate/superficial cells, respectively. The PLS-DA method provided improved sensitivities of 95.5%, 95.2% and 96.1% and specificities of 92.7%, 94.7% and 93.5% compared to the PCA-LDA method. The results demonstrate that the biochemical signatures of morphologically normal appearing cells can be used to discriminate between negative and HSIL cytology. In addition, it was found that mixed intermediate and superficial cells could be used for HSIL diagnosis as the biochemical differences between negative and HSIL cytology were greater than the biochemical differences between intermediate and superficial cell types.

  • Quasi-1D TiS <sub/>3</sub> Nanoribbons: Mechanical Exfoliation and Thickness-Dependent Raman Spectroscopy

    摘要: Quasi-one-dimensional (quasi-1D) materials enjoy growing interest due to their unusual physical properties and promise for miniature electronic devices. However, the mechanical exfoliation of quasi-1D materials into thin flakes and nanoribbons received considerably less attention from researchers than the exfoliation of conventional layered crystals. In this study, we investigated the micromechanical exfoliation of representative quasi-1D crystals, TiS3 whiskers, and demonstrate that they typically split into narrow nanoribbons with very smooth, straight edges and clear signatures of 1D TiS3 chains. Theoretical calculations show that the energies required for breaking weak interactions between the two-dimensional (2D) layers and between 1D chains within the layers are comparable and, in turn, are considerably lower than those required for breaking the covalent bonds within the chains. We also emulated macroscopic exfoliation experiments on the nanoscale by applying a local shear force to TiS3 crystals in different crystallographic directions using a tip of an atomic force microscopy (AFM) probe. In the AFM experiments, it was possible to slide the 2D TiS3 layers relative to each other as well as to remove selected 1D chains from the layers. We systematically studied the exfoliated TiS3 crystals by Raman spectroscopy and identified the Raman peaks whose spectral positions were most dependent on the crystals’ thickness. These results could be used to distinguish between TiS3 crystals with thickness ranging from one to about seven monolayers. The conclusions established in this study for the exfoliated TiS3 crystals can be extended to a variety of transition metal trichalcogenide materials as well as other quasi-1D crystals. The possibility of exfoliation of TiS3 into narrow (few-nm wide) crystals with smooth edges could be important for the future realization of miniature device channels with reduced edge scattering of charge carriers.

  • Raman spectroscopic determination of hole concentration in undoped GaAsBi

    摘要: Raman spectra of the undoped GaAs1-xBix (0<x<0.037) grown on GaAs by molecular beam epitaxy were investigated. With an increase of Bi component, we find that the longitudinal optical phonon-hole-plasmon-coupled (LOPC) mode first appears in the vicinity of the unscreened longitudinal optical (ULO) phonon frequency, and then shifts towards near the transverse optical (TO) phonon frequency. A new vibrational modes (~287 cm-1) between the TO and the ULO phonons was verified by use of low temperature polarized Raman measurement and the corresponding scattering intensities are found to be linearly proportional to the composition of Bi in GaAsBi. The hole concentrations determined by using the LOPC/ULO Raman intensities ratio increase from ~6.5×1016 to ~2.8×1017 cm -3 with Bi content and the measured results are in agreement with Hall measurements. Furthermore, the influence of excitation laser power on the estimation of the hole densities is discussed with the help of power dependent Raman spectroscopy.

  • Recent advances in linear and nonlinear Raman spectroscopy. Part XII

    摘要: This annual review is an overview of advances in the field of Raman spectroscopy as found in papers published in the previous calendar year in the Journal of Raman Spectroscopy (JRS), as well as in trends over the past decade across journals that have published papers important to the field of Raman spectroscopy. This information is gleaned from statistical data on article counts obtained from the Clarivate Analytic's Web of Science Core Collection by year and by subfield of Raman spectroscopy. Additional information is gleaned from presentations at the XXVI International Conference on Raman Spectroscopy (ICORS 2018) in Jeju Island, Korea in August 2018, and contributions on Raman scattering at SCIX 2018, organized by the Federation of Analytical Chemistry and Spectroscopy Societies (FACSS) in Atlanta, Georgia, USA in October 2018. Coverage is also provided for topics from the European Conference on Nonlinear Optical Spetroscopy (ECONOS 2018) held in April in Milan, Italy and GeoRaman 2018 in Catania, Italy in June. Finally, papers published in JRS in 2017 are highlighted and arranged by topics at the frontier of Raman spectroscopy. On the basis of this survey information, it is clear that Raman spectroscopy continues as in recent years as a rapidly expanding field of research across a wide range of novel disciplines and applications that provide sensitive photonic information of matter at the molecular level.

  • Time-Resolved Impulsive Stimulated Raman Spectroscopy with Synchronized Triple Mode-Locked Lasers

    摘要: A complete understanding of a photochemical reaction dynamics begins with real-time measurements of both electronic and vibrational structures of photoexcited molecules. Time-resolved impulsive stimulated Raman spectroscopy (TR-ISRS) with femtosecond actinic pump, Raman pump, and Raman probe pulses is one of the incisive techniques enabling one to investigate the structural changes of photoexcited molecules. Here, we demonstrate that such femtosecond TR-ISRS is feasible with synchronized triple mode-locked lasers without using any time-delay devices. Taking advantage of precise control of the three repetition rates independently, we could achieve automatic scanning of two delay times between the three pulses, which makes both rapid data acquisition and wide dynamic range measurement of the fifth-order TR-ISRS signal achievable. We thus anticipate that the present triple mode-locked laser-based TR-ISRS technique will be of critical use for long-term monitoring of photochemical reaction dynamics in condensed phases and biological systems.

  • Raman spectroscopy coupled with ambient ionization mass spectrometry: A forensic laboratory investigation into rapid and simple dual instrumental analysis techniques

    摘要: We present a laboratory experiment, developed around a pair of instrumental analysis techniques, Raman spectroscopy and ambient ionization mass spectrometry (MS), conducted by senior chemistry undergraduate students for bulk analysis of over-the-counter drugs using benchtop versions of the two instruments, as well as trace analysis of illicit drugs utilizing the corresponding portable instruments. The identification and confirmation of seized drugs is a routine operation in forensic laboratories that can be simplified using these two instrumental techniques in tandem. Bulk samples are readily analyzed using conventional Raman spectroscopy followed by paper cone spray ionization (PCSI) MS, while trace analysis is allowed by a simple single-substrate dual-analyzer method using surface enhanced Raman spectroscopy (SERS) from paper followed by paper spray ionization (PSI) MS. The introduction of these novel strategies to the students exposed them to potential career paths in forensic science by means of a hands-on experience that provided fundamental knowledge about this set of analytical methods, including intercomparisons of their performance, as well as practical issues like costs, regulations and databases. The use of portable instrumentation represents an additional learning experience for the students, considering that it is cutting-edge technology that is just now being transferred from academic to industrial labs and that it allows a valuable discussion of the advantages and disadvantages of bringing the laboratory to the field.

  • Effect of Ga introduction during the second stage of a coevaporation process of Cu(In,Ga)Se2 layers at low temperature on polyimide substrates

    摘要: A proper control of Ga concentration pro?le is mandatory to achieve high e?ciency Cu(In,Ga)Se2 (CIGS) solar cells. At low temperature, deep gradients, detrimental for carriers' di?usion, are obtained when CIGS is deposited with a standard three-stage process: an optimization of the process is needed. In this study, we show the impact of a modify three-stage process on the depth of the notch by introducing Ga ?ux during the second stage from 0 nm/min to 1.1 nm/min. A higher open circuit voltage compensated by a lower short current density is obtained due to higher band gap energy. The surface and the bulk of the CIGS layer was analyzed at the end of the second stage by coupling di?erent characterization techniques: glow discharge optical emission spectroscopy, Raman and X-ray photoelectrons spectroscopy. The presence of binary compounds as well as a Ga enrichment at the end of the second stage are observed when Ga is introduced during the second stage.
