Analysis of Fluorescence and Biodegradability of Wastewater
摘要: Three-dimensional fluorescence spectrometer was adopted for the content analysis of different types of organics in coking wastewater before biochemical treatment and through biochemical treatment, and the model of parallel factors was employed to analyze fluorescence components and contents. It was found that tryptophan-like components were the most easily degraded by biology, while humic-like components were the least easily degraded. Meanwhile, it had been seen that the change trends over time of total fluorescence densities of proteinoid fluorescence, and degradable organic fluorescence were highly consistent with that of parameter values of COD, NH3-N in this wastewater after analyzing the trends of the two indexes. It was proved that the three-dimensional fluorescence spectrum method was appropriate for the accurate degradation analysis of wastewater components.

FDOM Conversion in Karst Watersheds Expressed by Three-Dimensional Fluorescence Spectroscopy
摘要: A karst system, formed by the dissolution of carbonate rocks, is usually susceptible to contamination. Little is known about the composition of natural dissolved organic matter (DOM) in groundwater systems, especially in karstic groundwater. To reveal the characteristics of DOM in a karst aquifer, the Yufuhe River Basin, a typical karst watershed in northern China, was selected. DOM fluorescence (FDOM) was measured with the excitation-emission matrices (EEMs) spectroscopy technique. Parallel factor analysis (PARAFAC) was used to analyze the karst hydrogeological factors that affect FDOM biogeochemical behavior. Three fluorescent components, i.e., tyrosine-like, tryptophan-like, and ultraviolet fulvic acid were found. Their fluorescence properties were closely related to human activity and subterranean hydrology. Fluorescence properties suggested that FDOM in the Yufuhe River karst aquifer was predominant from anthropogenic activity. In addition, due to the effect of karstic heterogeneous hydrological conditions, FDOM showed obvious differentiation in the recharge, flow path, and discharge systems. The FDOM fluorescence intensity (FI) was weak in surface water and groundwater at the upper reaches (recharge area). In the middle of the flow path area, the percentage of tyrosine-like and tryptophan-like substances degraded and fulvic acid rose gradually. However, after infiltrating into the lower reaches (discharge area) of the deep karst aquifer system, the fulvic acid matter was consumed and protein-like matter accumulated.

Studies on the Interaction of Cefepime Hydrochloride with Bovine Serum Albumin by Fluorescence, Synchronous Fluorescence, Three-Dimensional Fluorescence and Circular Dichroism
摘要: The investigation on the binding mode between drug and protein is extremely important to understand biopharmaceutics, pharmacokinetics and toxicity of the drug as well as the relationship of structure and function of the protein. It is well proved that biological activity is a function of the chemical structure or structural properties. There is a study on the interaction of cefepime hydrochloride with serum albumin using in-silico molecular docking. But up to date, there is hardly any interaction investigation of cefepime hydrochloride with serum albumin utilizing by fluorescence, synchronous fluorescence, three-dimensional fluorescence and circular dichroism. In this study, the interaction of cefepime hydrochloride with bovine serum albumin in aqueous solution has been investigated by molecular spectroscopy under different pH conditions. The quenching rate constant and binding constant calculated at pH 7.4 indicated the static quenching mechanism and medium binding force. The effect of cefepime hydrochloride on the conformation of bovine serum albumin was analyzed using fluorescence, synchronous fluorescence, three-dimensional fluorescence and circular dichroism. In addition, influence of pH on the binding of cefepime hydrochloride to bovine serum albumin was investigated and the binding ability of the drug to bovine serum albumin deceased under other pH conditions (pH 1.9, 3.5, and 9.0) as compared with that at pH 7.4. As compared with the binding ability of cefepime hydrochloride to native bovine serum albumin that of cefepime hydrochloride to denatured bovine serum albumin deceases dramatically. Furthermore, the effect of metal ions on the binding constant of cefepime hydrochloride with bovine serum albumin was investigated.
