研究目的
Investigating the interaction of cefepime hydrochloride with bovine serum albumin using fluorescence, synchronous fluorescence, three-dimensional fluorescence, and circular dichroism under different pH conditions.
研究成果
The study concluded that the binding reaction of cefepime hydrochloride with BSA is sensitive to pH changes, with pH 7.4 being the optimal condition. Denatured BSA showed weaker binding affinity towards cefepime hydrochloride. The presence of certain metal ions affected the binding constant, indicating their potential influence on the drug's medicinal effects.
研究不足
The study focused on the interaction under different pH conditions and the effect of metal ions, but other environmental factors that might affect the interaction were not considered.
1:Experimental Design and Method Selection:
The study utilized molecular spectroscopy techniques including fluorescence, synchronous fluorescence, three-dimensional fluorescence, and circular dichroism to investigate the interaction under different pH conditions.
2:Sample Selection and Data Sources:
Bovine serum albumin (BSA) and cefepime hydrochloride were used. BSA was purchased from Sigma (USA), and cefepime hydrochloride was of analytical grade.
3:List of Experimental Equipment and Materials:
F-4500 spectrofluorophotometer (Hitachi, Japan) for fluorescence measurements, Olis DSM 1000 automatic recording spectrophotometer for CD spectral measurements.
4:Experimental Procedures and Operational Workflow:
Titrations were performed manually by using trace syringes. The effects of metal ions on the interaction were investigated by the gradual addition of cefepime hydrochloride to the mixture of metal ions and BSA.
5:Data Analysis Methods:
The fluorescence quenching data were analyzed with the Stern–Volmer equation to confirm the quenching mechanism.
