  • Extremely Photostable Electron-Deficient Phthalocyanines That Generate High Levels of Singlet Oxygen

    摘要: A robust lead-mediated synthetic procedure for the generation of phthalocyanines substituted with electron-withdrawing groups has been developed. The free-base phthalocyanine and various metal complexes were prepared without discernible degradation of the peripheral electron-withdrawing substituents. Upon irradiation with red light, some of the thus obtained metal complexes generated high levels of singlet oxygen. Especially a palladium complex exhibited attractive photostability upon exposure to singlet oxygen as a bleaching agent. The photostability of such complexes that may manifest concomitantly to the generation of high levels of singlet oxygen was attributed to the presence of the electron-withdrawing groups, which results in energetically low-lying highest occupied molecular orbitals.

  • Adipogenic differentiation of murine bone marrow mesenchymal stem cells induced by visible light via photo- induced biomodulation

    摘要: Background: Bone marrow mesenchymal stem cells (BM-MSCs) are undifferentiated cells that can proliferate and differentiate into specialized cells for tissue self-repair. Low-level laser (LLL) can induce biomodulatory effects such as cellular proliferation, differentiation, and migration. We investigated the biomodulatory effects of the photoactive compound chloroaluminum phthalocyanine nanoemulsion (AlClPc/NE) on the adipogenic differentiation of BM-MSCs, when combined with LLL (AlClPc/NE-LLL). Methods: The BM-MSCs used in this work were isolated from green fluorescent protein-positive (GFP+) C57BL6 mice. Cells were first treated with AlClPc/NE, a well-designed photoactive nano-drug and were then subjected to in vitro expansion, morphological and immunophenotypic characterization, and cellular cytotoxicity analysis. Subsequently, BM-MSCs were induced to differentiate into adipocytes by photo-induced biomodulation with AlClPc/NE-LLL. Results: Our results showed that the isolated cell population was consistent with murine BM-MSCs. The cellular cytotoxicity analysis revealed that the optimal nanoemulsion dose to induce BM-MSC biomodulation was 5.0 μmol/L. Twenty-four hours following treatment with AlClPc/NE, BM-MSC were subjected to visible light irradiation of 20 mJ/cm2 at 670 nm. Six days after photo-induced biomodulation, cells maintained high GFP expression level, and expressed detectable mRNA levels of adipogenic genes (lipoprotein lipase and PPARγ); formation of lipid vacuoles was observed, and the cells did not show any tumorigenic potential in vivo. Conclusions: Our results indicated that photo-induced biomodulation via visible light using AlClPc/NE and LLL can induce adipogenic differentiation of murine BM-MSCs. Therefore, cell therapy with BM-MSCs and photo-induced biomodulation may contribute to the development of new therapeutic strategies that are faster and more effective than traditional methods to trigger MSC differentiation.

  • Complex Formation of Cobalt(II) Octakis(3,5-di-tert-butylphenoxy)phthalocyanine with 2′-(Pyridin-4-yl)-5′-(Pyridin-2-yl)-1′-(Pyridin-2-ylmethyl)-2′,4′-Dihydro-1′H-Pyrrolo[ 3′,4′:1,2][C60-Ih][5,6]fullerene

    摘要: The formation of the donor–acceptor complex of cobalt(II) octakis(3,5-di-tert-butylphenoxy)phthalocyanine with 2'-(pyridin-4-yl)-5'-(pyridin-2-yl)-1'-(pyridin-2-ylmethyl)-2',4'-dihydro-1'H-pyrrolo[3',4':1,2][C60-Ih][5,6]fullerene (Py3C60) in toluene was studied by chemical kinetics (thermodynamics) and spectroscopy methods in comparison with a similar system based on 4-picoline (Pic). In both cases, the reaction occurred in one step giving donor–acceptor complexes of 1 : 1 stoichiometry, which were characterized by key spectral (UV-vis, IR, 1H NMR) parameters and thermodynamic equilibrium parameters; in the case of reaction with pyridyl-substituted fullerene, full kinetic description was obtained. The stability constants for (Pic)CoPc(3,5-tBuPhO)8 and (Py3C60)CoPc(3,5-tBuPhO)8 are (1.01 ± 0.16) × 104 L/mol (logK = 4.00, Δ G = –22.84 kJ/mol) and (1.84 ± 0.16) × 105 L/mol (logK = 5.26, Δ G = –30.04 kJ/mol), respectively. The obtained results are relevant for the development of new efficient components for photovoltaic cells based on phthalocyanine–fullerene systems.

  • Influence of Iodine Doping on the Structure, Morphology and Physical Properties of Manganese Phthalocyanine Thin Films

    摘要: Doping with halide ions is a popular method to alter the properties of metal phthalocyanines (MPcs), particularly magnetic and electrical nature of organic semiconductors for applications in spintronic or electronic devices. Doping can cause a structural rearrangement in MPc packing and the physical properties may be correlated with molecular packing. Films of a planar and magnetic MPc, manganese(II)phthalocyanine (MnPc) is chosen for iodine doping study. The optical, magnetic and the electrical properties of pristine and iodine doped MnPc thin films are investigated and can be directly associated with their molecular structure. 2D grazing incidence Synchrotron X-ray diffraction reveals structural disorder in MnPc films upon iodine infusion induced by the reorientation of ordered, edge-on molecular configuration to tilted and face-on configurations in a random fashion. The film morphology changes accordingly, where in the uniform crystallites reorganize in a disordered manner. The ferromagnetic nature of the pristine film gets weakened due to iodine species and favors antiferromagnetic coupling. The study of electrical properties at room temperature by conducting atomic force microscopy reveals that the conductance is enhanced independently of the film thickness due to disorder induced by iodine inclusion.

  • Photodynamic Effect of some Phthalocyanines on Enveloped and Naked Viruses

    摘要: Activity of three photosensitizing phthalocyanine derivatives was tested for photodynamic inactivation towards two coated and two non-enveloped viruses – bovine viral diarrhea virus (BVDV), influenza virus A(H3N2), poliovirus type 1 (PV-1) and human adenovirus type 5 (HAdV5). In the case of coated viruses, a combination of virucidal and irradiation effects was registered by octa-methylpyridyloxy-substituted Ga phthalocyanine (Ga8) toward BVDV, whereas tetra-methylpyridyloxy-substituted Ga phthalocyanine (Ga4) revealed a marked photoinactivation only. No such effect was observed towards influenza A virus. In contrast, the photoinactivating potential of Ga4 and Ga8 marked very high values on naked viruses, especially on HAdV5, at which both the virucidal as well as the irradiation effects became combined.

  • Temperature and frequency effect on the electrical properties of bulk nickel phthalocyanine octacarboxylic acid (Ni-Pc(COOH)8)

    摘要: The AC conductivity of nickel phthalocyanine octacarboxylic acid was investigated from 100 Hz to 1 MHz and temperature from 290 to 423 K. The AC conductivity was found to vary with frequency (σ(f)) and form two dispersion regions; the associated exponent factor “s” values were found to vary from 1.17 to 1.34 and from 0.42 to 0.67 (< 1). The value and temperature dependent of s are found in agreement with conduction mechanism models of large-polaron tunneling and the correlated barrier hopping, at the first and the second regions, respectively. The real and the imaginary parts of the dielectric constant were observed to decrease as the frequency increases indicating the pronounce contribution of low-frequency polarization mechanisms. Furthermore, the activation free energy ?F, enthalpy ?H, and entropy ?S of the sample were calculated as well.

  • Synthesis and electrochemical properties of peripheral, non-peripheral tetra [2-(3,5-diphenyl-1H-1,2,4-triazol-1-yl)ethoxy] substituted cobalt(II), manganese(III) phthalocyanines

    摘要: In this study, 4-[2-(3,5-diphenyl-1H-1,2,4-triazol-1-yl)ethoxy]phthalonitrile ET-CN, 3-[2-(3,5-diphenyl-1H-1,2,4-triazol-1-yl)ethoxy]phthalonitrile n-ET-CN and cobalt(II), manganese(III) phthalocyanines bearing [2-(3,5-diphenyl-1H-1,2,4-triazol-1-yl)ethoxy] group at peripheral, non-peripheral positions were synthesized for the first time. The new compounds have been characterized by FT-IR, NMR (only for ET-CN, n-ET-CN), mass and UV–Vis spectroscopy (except ET-CN, n-ET-CN) techniques. Electrochemistry of cobalt(II), manganese(III) phthalocyanines bearing [2-(3,5-diphenyl-1H-1,2,4-triazol-1-yl)ethoxy] group at peripheral, non-peripheral positions were examined by using cyclic voltammetry.

  • Benzyl ester dendrimer silicon phthalocyanine based polymeric nanoparticle for in vitro photodynamic therapy of glioma

    摘要: A novel second generation benzyl ester dendrimer silicon phthalocyanine (D-SiPc) was synthesized and encapsulated into amphiphilic block copolymers to form polymeric nanoparticles (D-SiPc@m). The cellular uptake and subcellular localization of D-SiPc@m in U251 glioma cells were studied by confocal laser scanning imaging. The cellular uptake of D-SiPc@m into U251 glioma cells reached the peak after being treatment for 6 h, and D-SiPc@m was localized into both mitochondria and lysosomes. The photodynamic efficacy of D-SiPc@m against U251 glioma cells was evaluated. D-SiPc@m exhibited high photo-cytotoxicity toward U251 glioma cells under laser irradiation. This polymeric nanoparticle could be a promising candidate for therapy of glioma.

  • Analyzing the Influence of Substituents on Proton Tautomerization - a Comparison of tetra- <i>tert</i> -Butyl Phthalocyanine Isomers

    摘要: We present a comparative study of tetra-tert-butyl phthalocyanine (ttbPc) isomers on a Ag(111) surface by means of low temperature scanning tunneling microscopy to analyze the influence of substituents on the tautomerization, a proton transfer reaction, in single molecules. By comparing ttbPc with the well studied phthalocyanine (H2Pc) molecule, we demonstrate a decoupling from the surface by the tert-butyl substituents. A comparison between the four ttbPc isomers, which naturally exist due to different bonding positions of the tert-butyl groups on the macrocycle, reveals a significant influence of the structural differences on their tautomerization behavior, as evidenced by a switching rate which varies by up to a factor of four between ttbPc isomers. These findings can be understood by an energetic landscape of the proton switch which links the binding distance of tert-butyl groups with the height of potential barriers. This model is supported by the analysis of two types of deprotonated ttbPc molecules with the molecular nanoprobe (MONA) technique.

  • Photodynamic therapy in infected venous and mixed leg ulcers: a pilot experience

    摘要: Objective: To assess the tolerability and efficacy of photodynamic therapy (PDT) with RLP068, a novel phthalocyanine-derived photosensitiser, in controlling the bacterial load in different leg ulcers, due to vascular pathophysiology. Method: An observational study of patients with infected leg ulcers of different pathophysiology, admitted to the hospital for a skin grafting procedure, were included. All patients underwent two sessions of PDT at time zero and after 72 hours. A semi-quantitative swab was taken before and 30 minutes after both sessions of PTD. During the time interval between the two treatments the ulcer was covered with foam dressing and compression therapy was applied to all patients. No systemic or topical antibiotics were administered. Statistical analysis of results was performed. Results: A total of 36 patients (13 males, 23 females; aged 72.4±8.6 years, range: 55-85 years) were recruited; two had ulcers on both legs. PDT was shown to be effective in reducing bacterial load after the first treatment. Before the second PDT treatment (72 hours after first PDT treatment), a slight increase of the bacterial load was observed in all ulcers. However, after the second PDT session, bacterial swab results were negative in all but two ulcers. The procedure was well tolerated in all but four patients, who reported a very severe pain at baseline, which increased during treatment. Conclusion: In this study, PDT was effective in reducing bacterial load in patients with infected vascular leg ulcers, and allowed successful skin grafting to take place in all patients. The treatment was generally well tolerated. Studies with a greater number of patients and a control group are planned to confirm these results.
