  • Physical origin of higher-order soliton fission in nanophotonic semiconductor waveguides

    摘要: Supercontinuum generation in Kerr media has become a staple of nonlinear optics. It has been celebrated for advancing the understanding of soliton propagation as well as its many applications in a broad range of fields. Coherent spectral broadening of laser light is now commonly performed in laboratories and used in commercial “white light” sources. The prospect of miniaturizing the technology is currently driving experiments in different integrated platforms such as semiconductor on insulator waveguides. Central to the spectral broadening is the concept of higher-order soliton fission. While widely accepted in silica fibers, the dynamics of soliton decay in semiconductor waveguides is yet poorly understood. In particular, the role of nonlinear loss and free carriers, absent in silica, remains an open question. Here, through experiments and simulations, we show that nonlinear loss is the dominant perturbation in wire waveguides, while free-carrier dispersion is dominant in photonic crystal waveguides.

  • 100?GHz free spectral range-tunable multi-wavelength fiber laser using single–multi–single mode fiber interferometer

    摘要: Six single–multi–single (SMS) mode fiber-based Mach–Zehnder Interferometers (MZI) with different spooling radii are demonstrated as spatial mode filtering elements for multi-wavelength laser generation. Free spectral range (FSR) tuning of the multi-wavelength output generated is realized by alternating between the six MZIs, which have different macro-bending losses. Additional FSR tuning is also realized by changing the spooling radius of the two-mode step index fiber section within the MZI setups from 80 mm to 30 mm, giving an FSR tuning range of 0.89–0.99 nm. All generated multi-wavelength outputs show high stability over a test period of 100 min. The proposed multi-wavelength lasers are highly suitable for various microwave photonics applications the field of such as microwave signal source generation and microwave photonic filtering.

  • Ultrasensitive tantalum oxide nano-coated long-period gratings for detection of various biological targets

    摘要: In this work we discussed a label-free biosensing application of long-period gratings (LPGs) optimized in refractive index (RI) sensitivity by deposition of thin tantalum oxide (TaOx) overlays. Comparing to other thin film and materials already applied for maximizing the RI sensitivity, TaOx offers good chemical and mechanical stability during its surface functionalization and other biosensing experiments. It was shown theoretically and experimentally that when RI of the overlay is as high as 2 in IR spectral range, for obtaining LPGs ultrasensitive to RI, the overlay’s thickness must be determined with subnanometer precision. In this experiment the TaOx overlays were deposited using Atomic Layer Deposition method that allowed for achieving overlays with exceptionally well-defined thickness and optical properties. The TaOx nano-coated LPGs show RI sensitivity determined for a single resonance exceeding 11,500 nm/RIU in RI range nD=1.335-1.345 RIU, as expected for label-free biosensing applications. Capability for detection of various in size biological targets, i.e., proteins (avidin) and bacteria (Escherichia coli), with TaOx-coated LPGs was verified using biotin and bacteriophage adhesin as recognition elements, respectively. It has been shown that functionalization process, as well as type of recognition elements and target analyte must be taken into consideration when the LPG sensitivity is optimized. In this work optimized approach made possible detection of small in size biological targets such as proteins with sensitivity reaching 10.21 nm/log(ng/ml).

  • A Speckle-Free Angular Interrogation SPR Imaging Sensor Based on Galvanometer Scan and Laser Excitation

    摘要: A speckle-free fast angular interrogation surface plasmon resonance imaging (SPRi) sensor based on a diode-pumped all-solid-state laser and galvanometer is reported in this work. A bidirectional scan using a galvanometer realizes the fast scanning of the incidence angle. The experimental results showed that the time needed for completing an SPR dip measurement was decreased to 0.5 s. And through cascading an immovable diffuser and two diffusers rotating in opposite directions, laser speckle was eliminated. The dynamic detection range and the sensitivity reached 4.6 × 10?2 and 1.52 × 10?6 refractive index unit (RIU), respectively, in a 2D array sensor when the angle scanning range was set as 7.5°. More importantly, the results demonstrated that the angular interrogation SPR imaging sensor scheme had the capability to perform fast and high-throughput detection of biomolecular interactions at 2D sensor arrays.

  • Precise label-free leukocyte subpopulation separation using hybrid acoustic-optical chip

    摘要: Leukocyte subpopulations contain crucial physiological information; hence, precise and specific leukocyte separation is very important for leukemia diagnosis and analysis. However, conventional centrifugation and immunofluorescence-based separation methods are inaccurate and inconvenient due to the overlapping cell size and density or complex marking processes. Herein, we report a new label-free technology for precise leukocyte subpopulation separation by synergy of acoustic and optical technologies. Standing surface acoustic wave (SSAW) solved the problem of gentle and precise focusing of cells in optical systems. In addition, SSAW was used for the separation of granulocytes, which have evident size distinction from other components. In case of lymphocytes and monocytes, which have overlap in size/density, optical force could distinguish them accurately based on the RI difference, with the convenience of acoustic pre-focusing. In this experiment, separation of three types of leukocyte subtypes with considerable throughput and purity was conducted, through which we obtained 99% pure lymphocytes, 98% pure monocytes, and 95% pure granulocytes. Experimental results prove that the device has robust ability in separating leukocyte phenotypes and have the advantages of being non-invasive, label-free and precise. In the future, this convenient hybrid method will be a potential powerful tool for auxiliary clinical diagnosis and analysis.

  • Robust Photodynamic Therapy Using 5-ALA-Incorporated Nanocomplexes Cures Metastatic Melanoma through Priming of CD4 <sup>+</sup> CD8 <sup>+</sup> Double Positive T Cells

    摘要: Advanced melanoma can rarely be cured. Photodynamic therapy (PDT) readily eradicates the primary melanoma but has limited ability to destroy the spreading tumor cells unless supported by other combinative interventions to augment systemic antitumor immunity. Based on the previously synthesized penetration-enhancing biomaterials, a topically administered nanoformulation is developed, which profoundly assists 5-aminolevulinic acid (5-ALA) in circumventing skin barrier to be selectively delivered to tumor cells. After endocytosis, accumulated 5-ALA is efficiently metabolized to a photosensitizer protoporphyrin IX (PpIX) which stimulates a large production of cytotoxic reactive oxygen species (ROS) under illumination. Accompanied by the robust inflammatory responses followed by primary tumor destruction, CD4+CD8+ double positive T cells are highly boosted to harness host immunity to purge metastases in lymphoid organs. Compared with dacarbazine and programmed death 1 (PD-1) antibody, this treatment in advanced melanoma murine models, achieves a striking curable rate of 90% without melanoma prognostic markers LDH and S-100B detection, followed by a relapse-free survival rate of 83.33% in 300 days. Moreover, the cured mice’s immune system function recovers to an extent similar to healthy mice without prolonged or exaggerated inflammation. This study using the synergistic biomaterials approach may thus render 5-ALA-mediated PDT a potentially curative therapy for advanced melanoma in clinic.

  • A Double-Hybridization Approach for the Transcription- and Amplification-Free Detection of Specific mRNA on a Microarray

    摘要: A double-hybridization approach was developed for the enzyme-free detection of specific mRNA of a housekeeping gene. Targeted mRNA was immobilized by hybridization to complementary DNA capture probes spotted onto a microarray. A second hybridization step of Cy5-conjugated label DNA to another section of the mRNA enabled specific labeling of the target. Thus, enzymatic artifacts could be avoided by omitting transcription and amplification steps. This manuscript describes the development of capture probe molecules used in the transcription- and amplification-free analysis of RPLP0 mRNA in isolated total RNA. An increase in specific signal was found with increasing length of the target-specific section of capture probes. Unspecific signal comprising spot autofluorescence and unspecific label binding did not correlate with the capture length. An additional spacer between the specific part of the capture probe and the substrate attachment site increased the signal significantly only on a short capture probe of approximately 30 nt length.

  • Electric field assisted spray coated lead free bismuth iodide perovskite thin film for solar cell application

    摘要: Solution-processed Methylammonium iodo bismuthate (MBI) perovskite solar cell is fabricated by spray technique with changed applied voltages from 0 to 1000 V during the deposition of MBI thin film. The morphology and surface roughness of MBI films are influenced significantly by the electric field during film deposition. It is attributed to improve the atomization of spray droplets due to process of coulomb fission. The surface roughness of MBI film is reduced from 39 to 19 nm with increased applied voltages during the deposition from 0 V and 1000 V, respectively. A strong absorption band is observed ~500 nm for all MBI films. The MBI perovskite solar cell is showed enhancement in the efficiency with the maximum current density 2.33 mA/cm2 at 1000 V applied voltage during the deposition. The improvement in photovoltaic characteristics with applied voltage during the film deposition is attributed to the formation of more uniform film with improved surface morphology and roughness, resulting in efficient electron transfer and reduced recombination of charge carrier at grain boundaries.

  • Photo-Modification of Melanin by a Mid-Infrared Free-Electron Laser

    摘要: Melanin is rigidly constructed by several nitrogen-containing aromatic rings, and its excess accumulation in skin tissue is closely associated with melanosis. Although visible lasers (wavelength: 600–1000 nm) are conventionally used for the photo-thermolysis of melanocyte, several pigmented nevi are difficult to be treated. Here, we propose an alternate method for targeting the molecular structure of melanin using an infrared free electron laser (FEL) tuned to 5.8 μm that corresponds to the stretching vibrational mode of carboxylate group. A drastic morphological change on the black-colored surface of melanin powder were observed after the pulse irradiation with a power of 500 mJ/cm2, and the minimum irradiation time for damage to the morphology was 1.4 seconds. Analyses by mass spectroscopy, infrared spectroscopy, and 13C-nuclear magnetic resonance implied that a pyrrole group was removed by the FEL irradiation. In addition, the FEL irradiation dispersed almost all of the melanoma cells from a culture solution without any influence on other ingredients in the medium, and one-cell analysis by infrared microscopy showed that the structure of melanoma could be substantially damaged by the irradiation. This study proposes the potency of intense mid-infrared laser as novel alternative way to reduce melanin.

  • Crystallographically oriented porous ZnO nanostructures with visible-blind photoresponse: controlling the growth and optical properties

    摘要: We have grown catalyst-free crystallographically oriented porous ZnO nanostructures by pulsed laser deposition (PLD) method. The deposition was performed in two stages for each sample. In the first stage, self-seeding of ZnO was performed on the quartz substrate, and the angle of deposition (seeding-angle) was varied. Thus, the growth of seeds is different here. In the second stage, the deposition was performed at a glancing angle (at 85?) for the fixed duration of time to grow the nanostructures. These PLD-grown nanostructures acquire highly oriented wurtzite structure. We find that the seeding-angle during the first stage is the determining deposition parameter which influences the growth and other properties of these nanostructures in a controllable manner. The variation in seeding-angle systematically tunes the crystallographic orientation and porosity, which in turn influences the visible-blindness and ultraviolet (UV) photoresponse of these nanonetworks. Here we report the growth of completely defect-free crystallographically oriented nanostructures with necessary porosity for application in visible-blind UV photodetection.
