  • Turn-On Fluorescence Aptasensor on Magnetic Nanobeads for Aflatoxin M1 Detection Based on an Exonuclease III-Assisted Signal Amplification Strategy

    摘要: In order to satisfy the need for sensitive detection of Aflatoxin M1 (AFM1), we constructed a simple and signal-on fluorescence aptasensor based on an autocatalytic Exonuclease III (Exo III)-assisted signal amplification strategy. In this sensor, the DNA hybridization on magnetic nanobeads could be triggered by the target AFM1, resulting in the release of a single-stranded DNA to induce an Exo III-assisted signal amplification, in which numerous G-quadruplex structures would be produced and then associated with the fluorescent dye to generate significantly amplified fluorescence signals resulting in the increased sensitivity. Under the optimized conditions, this aptasensor was able to detect AFM1 with a practical detection limit of 9.73 ng kg?1 in milk samples. Furthermore, the prepared sensor was successfully used for detection of AFM1 in the commercially available milk samples with the recovery percentages ranging from 80.13% to 108.67%. Also, the sensor performance was evaluated by the commercial immunoassay kit with satisfactory results.

  • Even-odd effect in higher-order holographic production of electron vortex beams with nontrivial radial structures

    摘要: Structured electron beams carrying orbital angular momentum are currently of considerable interest, both from a fundamental point of view and for application in electron microscopy and spectroscopy. Until recently, most studies have focused on the azimuthal structure of electron vortex beams with well-defined orbital angular momentum. To unambiguously define real electron-beam states and realize them in the laboratory, the radial structure must also be specified. Here we use a specific set of orthonormal modes of electron (vortex) beams to describe both the radial and azimuthal structures of arbitrary electron wavefronts. The specific beam states are based on truncated Bessel beams localized within the lens aperture plane of an electron microscope. We show that their Fourier transform set of beams can be realized at the focal planes of the probe-forming lens using a binary computer-generated electron hologram. Using astigmatic transformation optics, we demonstrate that the azimuthal indices of the diffracted beams scale with the order of the diffraction through phase amplification. However, their radial indices remain the same as those of the encoding beams for all the odd diffraction orders or are reduced to the zeroth order for the even-order diffracted beams. This simple even-odd rule can also be explained in terms of the phase amplification of the radial profiles. We envisage that the orthonormal cylindrical basis set of states could lead to new possibilities in phase contrast electron microscopy and spectroscopy using structured electron beams.

  • Coulomb interaction-induced jitter amplification in RF-compressed high-brightness electron source ultrafast electron diffraction

    摘要: We have theoretically and experimentally demonstrated an RF compression-based jitter-amplification effect in high-brightness electron source ultrafast electron diffraction (UED), which degrades the temporal resolution significantly. A detailed analysis and simulations reveal the crucial role of the longitudinal and transverse Coulomb interaction for this jitter-amplification effect, which accord very well with experimental results. An optimized compact UED structure for full compression has been proposed, which can suppress the jitter by half and improve the temporal resolution to sub-100 fs. This Coulomb interaction-induced jitter amplification exists in nearly the whole ultrafast physics field where laser-electron synchronization is required. Moreover, it cannot be suppressed completely. The quantified explanation for the mechanism and optimization provides important guidance for photocathode accelerators and other compression-based ultrashort electron pulse generation and precise control.

  • Nonlinear Noise of Low Transmission Penalty Dual-Stage Discrete Raman Amplifier

    摘要: We experimentally characterise the linear and nonlinear performance of a >70nm, dual-stage, 19.5dB average net gain discrete Raman amplifier using different nonlinear fibres in the second stage. We propose an architecture built with a combination of IDF and SMF, and compare its performance with amplifiers built with conventionally used nonlinear fibre types (IDF-IDF, IDF-DCF). The measured FWM product power shows the IDF-SMF architecture to generate less nonlinear interference when compared to other schemes. We test the amplifiers with 5x120Gb/s DP-QPSK WDM signals in a recirculating loop at 10 recirculations of 93.4km SMF fibre, where the power sweep shows up to 2dB optimum launch power difference, with the maximum Q2 factor varying by up to 1.6dB. Using the optimum transmission point we measure a Q2=8.8dB at 35 recirculations of 93.4km transmission (3269km) with the proposed IDF-SMF scheme, which is >460km further than the other tested architectures. All characterised schemes performed similarly in the linear noise regime.

  • [Springer Series in Optical Sciences] Fano Resonances in Optics and Microwaves Volume 219 (Physics and Applications) || Storage and Retrieval of Electromagnetic Waves in Metamaterials by Dynamical Control of EIT-Like Effect

    摘要: The storage of light is one of the most significant applications of electromagnetically induced transparency (EIT) effects in atomic systems. It is also possible to mimic the EIT effect using artificial media, or metamaterials, which are based on coupled resonators. This chapter focuses on metamaterials that realize the storage and retrieval of electromagnetic waves in the same way as the atomic EIT system. We introduce tunable metamaterials that are loaded with variable capacitors, which realize dynamical modulation of the EIT-like effects and the control of asymmetric spectra unique to the Fano resonance. Experiments are performed to show that electromagnetic waves are stored and released coherently in a multi-layered metamaterial designed for the operation in the microwave region. In addition, we show the frequency conversion of the stored waves and the extension of the storage time using parametric amplification.

  • Amplification and cross-Kerr nonlinearity in waveguide quantum electrodynamics

    摘要: We explore amplification and cross-Kerr nonlinearity by a three-level emitter (3LE) embedded in a waveguide and driven by two light beams. The coherent amplification and cross-Kerr nonlinearity were demonstrated in recent experiments, respectively, with a V and a ladder-type 3LE coupled to an open superconducting transmission line carrying two microwave fields. Here, we consider (cid:2), V, and ladder-type 3LE, and compare the efficiency of coherent and incoherent amplification as well as the magnitude of the cross-Kerr phase shift in all three emitters. We apply the Heisenberg-Langevin equations approach to investigate the scattering of a probe and a drive beam, both initially in coherent states. We particularly calculate the regime of the probe and drive powers when the 3LE acts most efficiently as a coherent amplifier and derive the second-order coherence of amplified probe photons. Finally, we apply the Kramers-Kronig relations to correlate the amplitude and phase response of the probe beam, which are used in finding the coherent amplification and the cross-Kerr phase shift in these systems.

  • A novel intracellular signal amplification strategy for the quantification of ATP in single cells by microchip electrophoresis with laser induced fluorescence detection

    摘要: An intracellular signal amplification strategy was developed for quantification of ATP in single cells by microchip electrophoresis with laser induced fluorescence detection. By using the method proposed, intracellular ATP levels in single Hela, HepG2 and HL-7702 cell were found in the range of 30~150, 30~140, and 19~120 fmol/cell, respectively.

  • Rapid, sensitive and highly specific label-free fluorescence biosensor for microRNA by branched rolling circle amplification

    摘要: A simple fluorescence biosensor for rapid and sensitive target microRNA (miRNA) quantification by branched rolling circle amplification (BRCA) is developed in this work. Target miRNA functions as primer to recognize and hybridize with a circle DNA template, initiating rolling circle amplification (RCA) by Phi29 DNA polymerase. The introduction of reverse primers complementary to the RCA products enables isothermal BRCA, in which a large amount of deoxynucleotide (dNTP) were consumed and same number of pyrophosphates (PPi) were produced. In this study, a simple and non-expensively synthesized terpyridine-based Zn(II) complex is utilized as fluorescent probe for selective detection of pyrophosphate (PPi) over dNTP. The PPi generated in this isothermal amplification process efficiently chelates to this terpyridine-Zn(II) complex, forming a highly fluorescent complex, terpyridine-Zn(II)-PPi, whose fluorescence intensity is closely related with the initial target miRNA concentration. The utilization of the isothermal BRCA amplification and direct monitoring of the DNA polymerization by-product, i.e. PPi, for non-label fluorescence detection of miRNA greatly simplify this sensor procedure. This sensor shows a linear response between the fluorescence intensity and the target miRNA concentration from 50 to 500 fM with a detection limit of 25 fM. This much-simplified sensor offers a sensitive and easy-to-use platform for miRNA quantification, and hence may significantly enhance the utilisation of miRNAs as biomarkers in drug discovery, clinical diagnosis and life science research.

  • Surface-plasmon-coupled chemiluminescence amplification of silver nanoparticles modified immunosensor for high-throughput ultrasensitive detection of multiple mycotoxins

    摘要: A novel surface-plasmon-coupled chemiluminescent immunosensor was developed for the detection of multiplex mycotoxins with ultrahigh sensitivity, high throughput and simplicity. The immunosensor was constructed by immobilizing carboxyl modified silver nanoparticles (AgNPs) and bovine serum albumin combined antigens sequentially on the glass chip modified with amino groups. The optical properties of AgNPs could amplify the chemiluminescence (CL) generated on the chip through the surface plasmon resonance phenomenon. Using a competitive immunoassay, the CL signals from all the sensing sites on the chip were collected simultaneously by a charge-coupled device for ultrasensitive analysis of multiple mycotoxins. Citrinin, aflatoxin B1 and ochratoxins A were selected as model analytes. Under optimal conditions, the surface-plasmon-coupled chemiluminescent immunosensor-based method presented wide linear ranges over 4 orders of magnitude and much lower limits of detection than previous work. The assay results of mycotoxins in red yeast rice samples using the proposed method were in good agreement with that using the liquid chromatography-mass spectrometry method. Its high selectivity, high throughput, acceptable stability and accuracy showed broad prospects in mycotoxin monitoring and evaluation of safety on Traditional Chinese Medicine.
