  • A new fluorescence probe based on diarylethene with a benzothiazine unit for selective detection of Cd2+

    摘要: A novel photochromic diarylethene derivative containing a benzothiazine unit has been synthesized. Its photochromism and fluorescent selectivity to metal ions were studied in detail in methanol solution. Under the stimulation of Cd2+ ions, the derivative showed a significant fluorescence enhancement and obvious red shift, accompanied by the fluorescent color changed from dark purple to bright blue. The 1 : 1 stoichiometry between the derivative and Cd2+ was verified by titration experiments and high resolution mass spectrometry. In addition, a molecular logic circuit was designed with the emission intensity at 476 nm as output and the stimuli of Cd2+/EDTA and UV/vis as inputs.

  • d-Glucose recognition based on phenylboronic acid-functionalized polyoligomeric silsesquioxane fluorescent probe

    摘要: We report a new strategy to synthesize hybrid fluorescent nanosensors consisting of phenylboronic acid-functionalized POSS (POSS–PBA) and diol-modified 8-anilino-1-naphthalenesulfonic acid (ANSA (a fluorescent dye)) for the detection of the biologically important D-glucose. The probe was characterized by FT-IR and 1H-NMR analyses, and the photoluminescence intensity was measured under various conditions to confirm its glucose sensing ability. Our POSS-APBA-dye probe could detect glucose at concentrations of 0–20 mg/mL, with a good linear relationship even at low glucose concentrations of 0–1 mg/mL. The properties of the POSS-APBA-dye probe were evaluated and compared with those of an APBA-dye probe. The glucose sensing ability of our POSS-APBA-dye probe was largely unaffected by the presence of interfering substances. The probe showed high sensing ability in a pH 5 environment and long-term (approximately 40 days) fluorescence stability.

  • A Colorimetric/Fluorescence Dual-channel Probe for Highly Discriminating Detection of Cysteine

    摘要: In this work, a novel fluorescence (FL) probe for selective and sensitive detection of Cys with colorimetric and FL dual signal changes was reported. The probe was synthesized by two step of sulfonamide reaction coupling between a sulfonyl benzoxadiazole (SBD) dye and dansyl chloride linked with rigid piperazine group. The probe showed a specific off-on response to Cys in aqueous solution with nanomolar LOD, and without interference by a range of amino acids and several competing analytes. Upon addition of Cys, the probe will undergo sequential substitution and intramolecular rearrangement reactions, yielding a 4-amino SBD derivative, which results in generation of strong yellow fluorescence emission at 575 nm accompanied by a two-step red shift in the absorption spectral. Moreover, it can be used for imaging of endogenous Cys in living cells.

  • Copper nanoclusters @ nitrogen-doped carbon quantum dots-based ratiometric fluorescence probe for lead (II) ions detection in porphyra

    摘要: A novel ratiometric fluorescence probe was proposed for detecting lead (II) ions (Pb2+) in porphyra, the approach was based on copper nanoclusters and nitrogen-doped carbon quantum dots (CuNCs-CNQDs). In this probe, the CuNCs delivered the response signal, the fluorescence of which was enhanced by Pb2+ due to the aggregation-induced emission enhancement (AIEE) between Pb2+ and CuNCs. The CNQDs provided the self-calibration signal, whose fluorescence remained almost unchanged in coexistence with Pb2+. According to the change of fluorescence intensity ratio between the fluorophores, CuNCs–CNQDs nanohybrid was used as ratiometric probes for the sensitive detection of Pb2+ in the range of 0.010 to 2.5 mg L-1, with a detection limit of 0.0031 mg L-1. Finally, the probe was successfully applied to detect Pb2+ in porphyra with relative standard deviations (RSDs) lower than 5%. This study provides a straightforward, stable, and sensitive approach for detecting Pb2+ in porphyra.

  • <p>A Lysosome-Targetable Fluorescence Probe Based on L-Cysteine-Polyamine-Morpholine-Modified Quantum Dots for Imaging in Living Cells</p>

    摘要: Quantum dots (QDs) are used as fluorescent probes due to their high fluorescence intensity, longevity of fluorescence, strong light-resistant bleaching ability and high light stability. Therefore, we explore a more precise probe that can target an organelle. Methods: In the current study, a new class of fluorescence probes were developed using QDs capped with 4 different L-cysteine-polyamine-morpholine linked by mercapto groups. Ligands were characterised by Electrospray ionization mass spectrometry (ESI-MS), H-Nuclear Magnetic Resonance (1H NMR) spectroscopy, and 13C NMR spectroscopy. Modified QDs were characterized by Transmission Electron Microscope (TEM), Ultraviolet and visible spectrophotometry (UV–Vis), and fluorescence microscopy. And the biological activity of modified QDs was explored by using MTT assay with HeLa, SMMC-7721 and HepG2 cells. The fluorescence imaging of modified QDs was obtained by confocal laser scanning fluorescence microscopy (CLSM). Results: Synthesized QDs ranged between 4 to 5 nm and had strong optical emission properties. UV–Vis and fluorescence spectra demonstrated that the cysteine-polyamine-morpholine were successfully incorporated into QD nanoparticles. The MTT results demonstrated that modified QDs had lesser cytotoxicity when compared to unmodified QDs. In addition, modified QDs had strong fluorescence intensity in HeLa cells and targeted lysosomes of HeLa cells. Conclusion: This study demonstrates the modified QDs efficiently entered cells and could be used as a potential lysosome-targeting fluorescent probe.

  • Anti-inflammatory effect of photodynamic therapy using guaiazulene and red lasers on peripheral blood mononuclear cells

    摘要: To evaluate the anti-inflammatory effect of photodynamic therapy using guaiazulene and a red laser in peripheral blood mononuclear cells (PBMCs). Guaiazulene solutions (1, 2, 5, 25, 35, and 100 μM in 99.8% methanol) were irradiated with red laser light (625 nm, 151.5 mW/cm2) in continuous mode at 0, 4, and 8 J/cm2 in black 96-well plates. ROS were measured using spin trapping technique with electron spin resonance (ESR) spectroscopy and fluorescence. The two highest concentrations were tested using cell viability (PrestoBlue?) and anti-inflammation (RANTES and PGE2 ELISA) assay kits. Kruskal-Wallis and Dunn Bonferroni tests were used for statistical analyses with significant differences at p-value < 0.05. Guaiazulene solutions between 2 and 5 μM exposed to red laser light at 4-8 J/cm2 generated significantly more singlet oxygen compared to the no guaiazulene group (p < 0.01) and reduced RANTES and PGE2 levels in TNF-α-inflamed peripheral blood mononuclear cells without affecting cell viability. Photodynamic activation of guaiazulene generated singlet oxygen and suppressed inflammatory markers in PBMCs.

  • Synthesis of Glutathione Modified CdSe Quantum Dots and Its Application in Determination of Trace Hg2+ Ions

    摘要: The water-soluble glutathione (GSH) modified CdSe quantum dots (QDs) were prepared. Results show that the optimum NaBH4:Se:(Cd2+): glutathione molar ratio is obtained to be 3:1:3:9. Meanwhile, the glutathione modified CdSe quantum dots synthesized by the optimum process have a single face centered cubic phase. It is found that the fluorescence quenching occurred as Hg2+ ions interact with glutathione modified CdSe quantum dots. Hg2+ ions can be determined in the concentration range of 0.3-15 μg/L, which is proposed to determine the trace quantities of Hg2+ ions as fluorescence probe.

  • Quantum dots based “on-off” fluorescence probe for the selective detection of Cu2+ ion: Application to real samples analysis

    摘要: Turn ‘On-Off’ quantum dot-based fluorescent probe has been developed for the selective detection of Cu2+. This strategy achieved by consecutively fluorescence enhancement and quenching of mercapto propionic acid (MPA) capped cadmium sulphide quantum dots (MPA-CdS QDs) by the addition of D-penicillamine (D-PA) and Cu2+ respectively. After successive addition of Cu2+ in CdS QDs- (D-PA) system, the fluorescence intensity of quantum dots decrease due to the removal of D-PA molecules from the surface of QDs as well as selective ion exchange process takes place between Cu2+ and Cd2+ at the surface of QDs. At optimal conditions, probe offers a good response in the linear range between 4 ng/mL and 40 ng/mL with LOD 2.71 ng/mL. This reported strategy is a very simple and selective towards Cu2+. Hence, a new method for the potential detection of Cu2+ in real pharmaceutical samples can be developed by using QDs-based fluorescence probe.

  • Amine derivative of triphenyl ether as an optical sensor for the detection of cyanide ions and traces of water in acetonitrile supported with voltammetric studies

    摘要: A novel triphenyl ether amine (TPEA) has been synthesized to develop a customized receptor for cyanide ions. TPEA is selective for CN? ions in the existence of a number of competitive anions like HSO4?, OAc?, and halides. The proposed receptor responds linearly to CN? ions up to a concentration of 400 μM with a detection limit of 0.4 μM and with a binding constant of 4.16 × 107 M?1. The response of the receptor for CN? ions was validated by voltammetric methods. TPEA has also been studied as a probe for the detection of water in acetonitrile.

  • Real-time monitoring of newly acidified organelles during autophagy enabled by reaction-based BODIPY dyes

    摘要: Real-time monitoring of newly acidified organelles during autophagy in living cells is highly desirable for a better understanding of intracellular degradative processes. Herein, we describe a reaction-based boron dipyrromethene (BODIPY) dye containing strongly electron-withdrawing diethyl 2-cyanoacrylate groups at the α-positions. The probe exhibits intense red fluorescence in acidic organelles or the acidified cytosol while exhibiting negligible fluorescence in other regions of the cell. The underlying mechanism is a nucleophilic reaction at the central meso-carbon of the indacene core, resulting in the loss of π-conjugation entailed by dramatic spectroscopic changes of more than 200 nm between its colorless, non-fluorescent leuco-BODIPY form and its red and brightly emitting form. The reversible transformation between red fluorescent BODIPY and leuco-BODIPY along with negligible cytotoxicity qualifies such dyes for rapid and direct intracellular lysosome imaging and cytosolic acidosis detection simultaneously without any washing step, enabling the real-time monitoring of newly acidified organelles during autophagy.
