  • The electrical signal-induced systemic photosynthetic response is accompanied by changes in the photochemical reflectance index in pea

    摘要: Plants can be affected by numerous environmental stressors with spatially heterogeneous actions on their bodies. A fast systemic photosynthetic response, which is connected with long-distance electrical signalling, plays an important role in the adaptation of higher plants to the action of stressors. Potentially, measurement of the response by using a photochemical reflectance index (PRI) could be the basis of monitoring photosynthesis under spatially heterogeneous stressors; however, the method has not been previously used for investigating the systemic photosynthetic response. We investigated changes in PRI and photosynthetic parameters (quantum yields of PSI and PSII and nonphotochemical quenching) in intact leaves of pea (Pisum sativum L.) after local heating of another leaf and the propagation of electrical signals through the plant body. We showed that electrical signals decreased the quantum yields of PSI and PSII and increased the nonphotochemical quenching of intact leaves in times ranging from minutes to tens of minutes; the changes were strongly connected with changes in PRI. Additional analysis showed that changes in PRI were caused by an increase of the energy-dependent quenching induced by electrical signals. Thus PRI can be potentially used for monitoring the systemic photosynthetic response connected with long-distance electrical signalling.

  • DWARF4 accumulation in root tips is enhanced via blue light perception by cryptochromes

    摘要: Brassinosteroid (BR) signalling is known to be coordinated with light signalling in aboveground tissue. Many studies focusing on the shade avoidance response in aboveground tissue or hypocotyl elongation in darkness have revealed the contribution of the BR signalling pathway to these processes. We previously analysed the expression of DWARF 4 (DWF4), a key BR biosynthesis enzyme, and revealed that light perception in aboveground tissues triggered DWF4 accumulation in root tips. To determine the required wavelength of light and photoreceptors responsible for this regulation, we studied DWF4-GUS marker plants grown in several monochromatic light conditions. We revealed that monochromatic blue LED light could induce DWF4 accumulation in primary root tips and root growth as much as white light, while monochromatic red LED could not. Consistent with this, a cryptochrome1/2 double mutant showed retarded root growth under white light while a phytochromeA/B double mutant did not. Taken together, our data strongly indicated that blue light signalling was important for DWF4 accumulation in root tips and root growth. Furthermore, DWF4 accumulation patterns in primary root tips were not altered by auxin or sugar treatment. Therefore, we hypothesize that blue light signalling from the shoot tissue is different from auxin and sugar signalling.

  • Electrochemiluminescent biosensor with DNA link for selective detection of human IgG based on steric hindrance

    摘要: A highly selective DNA-based electrochemiluminescence (ECL) based biosensor is described for the detection of human IgG. It is exploiting the effect of steric hindrance that affects the strength of the ECL signal in the presence of IgG. Digoxin-linked signalling DNA was specifically bound to IgG, and this causes steric hindrance which limits the ability of DNA to hybridize with capturing DNA attached to a gold electrode. Europium (II) doped CdSe quantum dots were covalently linked to the DNA in order to generate the ECL signal. Using this steric hindrance hybridization method, the ECL signal of the biosensor were proportional to the concentration of IgG with a wide linear range and a 14 pM detection limit. Conceivably, the method can be expanded to the detection of a wide range of proteins for which homologous recognition elements are available.

  • The impact of photocatalytic Ag/TiO2 and Ag/N-TiO2 nanoparticles on human keratinocytes and epithelial lung cells

    摘要: The potential human health risks following the exposure to inorganic nanoparticles (NPs) is a very important issue for their application in leather finishing industry. The aim of our study was to investigate the cytotoxic effect of silver (Ag)/titanium dioxide (TiO2) NPs on human cells. Photocatalytic NPs were prepared by electrochemical deposition of Ag on the surface of TiO2 and nitrogen (N)-TiO2 NPs and, subsequently, physico-chemical characterized. Then, a set of experiments have been performed to study the cytotoxicity and cell death mechanisms involved, the changes in cell morphology and the production of ROS induced in human keratinocytes (HaCaT) and human lung epithelial cells (A549) by exposure to NPs. Moreover, the changes in major signaling pathways and the inflammatory response induced by Ag/N-TiO2 NPs in A549 cells were investigated. The data showed that cell death by late apoptosis/necrosis is induced in cells as function of the dose and the type of NPs and is characterized by morphological changes and cytoskeletal disorganization and an increase in reactive oxygen species (ROS) production. The exposure of A549 cells to Ag/N-TiO2 NPs determine the activation of ERK1/2 MAP-kinase pathway and the release of pro-inflammatory mediators CXCL1, GM-CSF and MIF, known to be involved in the recruitment of circulating neutrophils and monocytes. In conclusion, due to its cyto-friendly properties as compared to Ag/TiO2 NPs, we consider Ag/N-TiO2 NPs suitable for leather surface finishing with some caution regarding the limit of dose handled, that has to be below 0.5 mg/ml.

  • Otx2 enhances transdifferentiation of Müller cells-derived retinal stem cells into photoreceptor-like cells

    摘要: Retinal Müller glial cells have the potential of neurogenic retinal progenitor cells, and could reprogram into retinal‐specific cell types such as photoreceptor cells. How to promote the differentiation of Müller cells into photoreceptor cells represents a promising therapy strategy for retinal degeneration diseases. This study aimed to enhance the transdifferentiation of rat Müller cells‐derived retinal stem cells (MC‐RSCs) into photoreceptor‐like cells and explore the signalling mechanism. We dedifferentiated rat Müller cells into MC‐RSCs which were infected with Otx2 overexpression lentivirus or control. The positive rate of photoreceptor‐like cells among MC‐RSCs treated with Otx2 overexpression lentivirus was significantly higher compared to control. Furthermore, pre‐treatment with Crx siRNA, Nrl siRNA, or GSK‐3 inhibitor SB‐216763 reduced the positive rate of photoreceptor‐like cells among MC‐RSCs treated with Otx2 overexpression lentivirus. Finally, Otx2 induced photoreceptor precursor cells were injected into subretinal space of N‐methyl‐N‐nitrosourea induced rat model of retinal degeneration and partially recovered retinal degeneration in the rats. In conclusion, Otx2 enhances transdifferentiation of MC‐RSCs into photoreceptor‐like cells and this is associated with the inhibition of Wnt signalling. Otx2 is a potential target for gene therapy of retinal degenerative diseases.

  • Shining light on plant hormones with genetically encoded biosensors

    摘要: Signalling molecules are produced, degraded, modified and transported throughout the development of higher organisms. Understanding their mode of action implies understanding these dynamics in vivo and in real time. Genetically encoded biosensors are being more and more used as tools to ‘follow’ signalling molecules and their responses inside an organism. This is the case for plants, where important progresses have been made in the development of such biosensors. Here, we summarize the main genetically encoded biosensors built for plant hormones, constructed using diverse components and steps of their signalling pathways.
