研究不足
潜在的误差来源包括测量过程中的空气进入(尤其是在滴眼液法中)、比色皿法中缺乏受控混合、镜片厚度和暴露面积的差异,以及可能的泄漏或残留氧气。建议的改进措施包括更好的密封、使用隔膜进行采样、添加混合系统以及设计专用比色皿。
1:实验设计与方法选择:
采用3D打印部件设计了两套定制系统——滴眼液瓶法和比色皿法。两套系统均将隐形眼镜作为半胱胺溶液氧气进入的唯一通道,通过紫外-可见分光光度法测量氧化速率,基于化学计量关系计算氧气透过率。
2:样本选择与数据来源:
使用市售隐形眼镜(如安视优舒视氧、安视优欧舒适、安视优天天抛、库博光学艾维瑞等)。半胱胺溶液配制于充氮除氧的磷酸盐缓冲盐水(PBS)中。
3:实验设备与材料清单:
设备包括创想三维CR-10 3D打印机、Monoprice Select 3D打印笔、热电Genysys 10S紫外-可见分光光度计、打孔器、石英比色皿、滴眼液瓶、铝箔、强力胶及多种化学试剂(半胱胺、乙醇、PBS、维生素E)。材料包含PLA线材及特定品牌隐形眼镜。
4:实验流程与操作步骤:
滴眼液瓶法通过在瓶身打孔,用3D打印盖固定隐形眼镜后注入半胱胺溶液,滴落液用于测量;比色皿法采用改良比色皿盖固定隐形眼镜,直接盛装半胱胺溶液随时间测定吸光度。两种方法均预先通氮除氧。
5:数据分析方法:
根据紫外-可见吸收数据测量半胱胺氧化速率,利用氧化速率与镜片氧气通量的化学计量方程计算氧气透过率。
