ELDI-MSI was shown to be an effective method for diagnosing agglomeration and crystallinity content in pharmaceutical preparations with little to no sample preparation. The spatial distributions of amorphous API were similar to surfactants, while crystalline API showed agglomeration. This method provides valuable information on the form of APIs in tablets, which is crucial for understanding solubility and bioavailability.

The study was limited by the laser wavelength available (355 nm), requiring the addition of TiO2 to the tablet blend for uniform laser ablation. The horizontal elongation of analyte images due to the data acquisition method may affect the interpretation of spatial distributions.
