研究目的
Investigating the enhancement of curcumin antitumor efficacy and further photothermal ablation of tumor growth by single-walled carbon nanotubes delivery system in vivo.
研究成果
SWCNT-Cur significantly enhances the antitumor activity of curcumin in vivo by improving its bioavailability, cellular uptake, and combining photothermal therapy. The formulation shows no obvious toxicity in main organs and negligible solvent residue, making it a promising strategy for cancer therapy.
研究不足
The study focuses on the in vivo and in vitro evaluation of SWCNT-Cur's performance and characteristics, but the long-term toxicity and clinical applicability of SWCNT-Cur need further investigation.
1:Experimental Design and Method Selection:
The study involved the preparation of SWCNT-Cur, characterization of the formulation, in vitro cellular uptake studies, in vivo pharmacokinetic studies, and evaluation of antitumor efficacy in a murine S180 tumor model.
2:Sample Selection and Data Sources:
Human prostate cancer PC-3 cells and S180 mouse ascites tumor cells were used.
3:List of Experimental Equipment and Materials:
SWCNT, curcumin, phosphatidylcholine, polyvinylpyrrolidone K30, Tween 20, RPMI-1640 medium, fetal bovine serum, penicillin, streptomycin, Agilent 6890 N Gas Chromatography, Nicolet iS10 spectrometer, confocal microscopy, spectrofluorometer, Agilent 1100 HPLC system.
4:Experimental Procedures and Operational Workflow:
Preparation of SWCNT-Cur, solvent residue and FTIR spectral analysis, qualitative and quantitative cellular uptake, in vivo pharmacokinetics, animal model and treatment, histological analysis.
5:Data Analysis Methods:
Statistical analysis was performed using ANOVA followed by Dunnett’s test by SPSS17.0 statistical software.
