Investigating the impact of refractive index increment (dn/dc) on the determination of molecular weight (Mw) of hyaluronic acid (HA) by muti-angle laser light-scattering (MALS) technique and measuring the Mw variation of HA in the manufacturing process of preparations.

Each Mw of HA sample corresponds to a specific value of dn/dc, ranging from 1.38 to 1.74 L/g with the Mw increasing from 13.5 to 2840 kDa. MALS with appropriate dn/dc selected is reliable and accurate for Mw determination. The random coil structure of HA is little influenced by the sterilization operation, providing a precise method for Mw determination in medical applications and preparation industries.

The study focuses on HA in stroke-physiological saline solution and may not be directly applicable to other solvents or conditions. The impact of temperature on dn/dc parameters was investigated but only for a limited range of temperatures (20, 30, and 40 °C).
