Reflectance NIRS can be used to monitor barrier function in stratum corneum based on changes in protein and lipid. It provides a non-invasive method to monitor changes in protein, lipid and water in stratum corneum, which can be useful for investigating skin disorders related with barrier disruption.

The study was conducted on a limited number of healthy volunteers without dermatological disorders. Further studies are required to investigate skin with disorders such as atopic dermatitis using NIRS.
1:Experimental Design and Method Selection:
Reflectance NIRS was used to investigate the effect of disruption of barrier on spectra of skin. Multivariate statistical analysis and second derivative analysis of human skin spectra were performed.
2:Sample Selection and Data Sources:
Sixteen healthy volunteers without any dermatological disorder participated. NIR skin spectra were collected using VERTEX 80 FTIR spectrometer.
3:List of Experimental Equipment and Materials:
VERTEX 80 FTIR spectrometer (Bruker, Germany), fiber optic probe (Bruker, Germany), 3M scotch tape, Dermalab Combo (Cortex Technology, Denmark), Spectralon (Labsphere, New Hampshire).
4:Experimental Procedures and Operational Workflow:
Site for analysis was marked and 3M scotch tape was used for tape stripping of stratum corneum in volar forearm to induce acute barrier disruption. Before tape stripping, TEWL was measured and NIR spectra were collected for baseline measurement. After each three consecutive tape stripping with adhesive tape on volar forearm, TEWL was measured and skin spectra were obtained.
5:Data Analysis Methods:
Standard Normal Variate (SNV) method was applied for the pre-treatment of spectra obtained from each subject. Principal component analysis (PCA) was performed for multivariate analysis of skin spectra. The second derivative of skin spectra were calculated with the Savitzky-Golay derivative.
