1:Experimental Design and Method Selection:
The study employed a SERS-based aptasensor using double strand DNA binding bimetallic gold@silver nanoparticles as a sensing platform. Probe DNAs were embedded on gold nanoparticles via thiol modification, followed by silver shell encapsulation and hybridization with Cy3-labeled aptamer DNAs.
2:Sample Selection and Data Sources:
Milk samples were obtained from an online supermarket (Mengniu brand). KANA stock solutions were prepared and diluted for testing.
3:List of Experimental Equipment and Materials:
Chemicals included gold(III) chloride trihydrate, silver nitrate, etc., from various suppliers. Instruments included a laser confocal Raman microscope system (LabRAM HR), spectrophotometer (UV-1800), transmission electron microscope (JEM-1400 Plus and JEM-2100), and potentiometric analyzer (Zetasizer Nano).
4:Experimental Procedures and Operational Workflow:
Synthesis of Au NPs and modification with probe DNA, synthesis of Au@Ag NPs-ssDNA and modification with aptamer DNA, analysis of KANA in milk samples with preprocessing to remove proteins and fats, followed by SERS measurement.
5:Data Analysis Methods:
Raman spectra were collected and analyzed, with detection limit calculated using LOD = 3Sb/b, and selectivity assessed using R ratio.
