1:Experimental Design and Method Selection:
Two custom systems were designed using 3D-printed parts: an eye drop bottle method and a cuvette method. Both systems isolate the contact lens as the only path for oxygen entry into a cysteamine solution, and UV-vis spectrophotometry measures the oxidation rate to calculate oxygen transmissibility based on stoichiometry.
2:Sample Selection and Data Sources:
Commercial contact lenses (e.g., Acuvue Moist, Acuvue TruEye, Acuvue Oasys, Dailies AquaComfort Plus, Dailies Total1, Coopervision Avaira) were used. Cysteamine solutions were prepared in phosphate-buffered saline (PBS) purged with nitrogen to remove oxygen.

Equipment includes a Creality CR-10 3D printer, Monoprice Select 3D Printing Pen, Thermospectronic Genysys 10S UV-Vis spectrophotometer, hole-puncher, quartz cuvettes, eye drop bottles, aluminum foil, super glue, and various chemicals (cysteamine, ethanol, PBS, vitamin E). Materials include PLA filament, contact lenses from specific brands.
4:Experimental Procedures and Operational Workflow:
For the eye drop bottle method, a hole is punched in the bottle, covered with a contact lens using a 3D-printed cap, filled with cysteamine solution, and drops are eluted for measurement. For the cuvette method, a modified cuvette lid holds the contact lens, filled with cysteamine solution, and absorbance is measured directly over time. Both methods involve purging with nitrogen to remove oxygen initially.
5:Data Analysis Methods:
The rate of cysteamine oxidation is measured from UV-vis absorbance data, and oxygen transmissibility is calculated using a stoichiometric equation relating the oxidation rate to oxygen flux through the lens.
