研究目的
To develop functionalized fluorescent carbon nanoparticles for sensitively targeted imaging of folate-receptor-positive cancer cells.
研究成果
The FA-CDs demonstrate excellent targeting specificity for FR-positive cancer cells, high biocompatibility, and low toxicity, making them a promising platform for cancer diagnosis and therapy through targeted bioimaging.
研究不足
The quantum yield of the FA-CDs is 17.3%, which may be lower than some other nanomaterials, potentially limiting brightness in imaging applications. The study focuses on in vitro cell lines; in vivo applications and long-term toxicity were not evaluated.
1:Experimental Design and Method Selection:
The study used a hydrothermal method to synthesize carbon dots (CDs) from proline and ethylenediamine, followed by covalent conjugation with folic acid (FA) using EDC and NHS for targeted bioimaging. Confocal laser scanning microscopy was employed for cell imaging.
2:Sample Selection and Data Sources:
MCF-7 cells (FR++), HepG-2 cells (FR+), and PC-12 cells (FR-) were used for comparative studies to evaluate targeting specificity.
3:List of Experimental Equipment and Materials:
Instruments included a Teflon-lined stainless autoclave, dialysis membrane (MWCO 500–1000 Da), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), Fourier transform infrared (FTIR) spectroscopy, UV-vis spectrophotometer, zeta potential analyzer, and confocal laser scanning microscope. Materials included proline, ethylenediamine (EDA), folic acid (FA), EDC, NHS, PBS buffer, and cell culture reagents.
4:Experimental Procedures and Operational Workflow:
CDs were synthesized hydrothermally at 180°C for 5 hours, then conjugated with FA via EDC/NHS coupling, dialyzed, and characterized. Cell imaging involved incubating cells with FA-CDs for 2 hours and observing fluorescence.
5:Data Analysis Methods:
Fluorescence intensity was measured, quantum yield calculated using quinine sulfate as reference, and statistical analysis performed for cytotoxicity (MTT assay) and imaging results.
