The experimental glaucoma group included only relatively young animals, so effects of aging could not be assessed. The radial scan protocol used only 12 B-scans, which may increase variability and limit sector-based analysis. Human data were cross-sectional, not longitudinal, so temporal relationships could not be fully established.
1:Experimental Design and Method Selection:
The study used a nonhuman primate experimental glaucoma model and cross-sectional human data. Custom algorithms were developed for OCT scan analysis, including transverse scaling based on ocular biometry.

Normative data from 44 healthy rhesus monkeys and longitudinal data from 9 monkeys with induced glaucoma; cross-sectional data from 89 healthy, 74 glaucoma suspect, and 104 glaucoma human patients.

4:Experimental Procedures and Operational Workflow:
Animals were anesthetized, pupils dilated, and OCT scans acquired. Glaucoma was induced by laser scarring of the trabecular meshwork. Human subjects underwent OCT scanning after pupil dilation. Data were exported and analyzed with custom MATLAB algorithms.
5:Data Analysis Methods:
Statistical analysis using GraphPad Prism, MATLAB with Statistics Toolbox, and SigmaPlot. Nonlinear fits (exponential models) were applied where linear fits were inadequate, assessed by runs test and Akaike information criteria.
