Calcination-free synthesis of high strength self-cleaning anti-reflective MgF2-TiO2 coating
摘要: In order to understand the nonlinear dynamic behavior of a planar mechanism with clearance, the nonlinear dynamic model of the 2-DOF nine-bar mechanism with a revolute clearance is proposed; the dynamic response, phase diagrams, Poincar′e portraits, and largest Lyapunov exponents (LLEs) of mechanism are investigated. The nonlinear dynamic model of 2-DOF nine-bar mechanism containing a revolute clearance is established by using the Lagrange equation. Dynamic response of the slider’s kinematics characteristic, contact force, driving torque, shaft center trajectory, and the penetration depth for 2-DOF nine-bar mechanism are all analyzed. Chaos phenomenon existed in the mechanism has been identi?ed by using the phase diagrams, the Poincar′e portraits, and LLEs. The e?ects of the di?erent clearance sizes, di?erent friction coe?cients, and di?erent driving speeds on dynamic behavior are studied. Bifurcation diagrams with changing clearance value, friction coe?cient, and driving speed are drawn. The research could provide important technical support and theoretical basis for the further study of the nonlinear dynamics of planar mechanism.

Improved immuno-PET imaging of HER2-positive tumors in mice: Urokinase injection-triggered clearance enhancement of <sup>64</sup> Cu-trastuzumab
摘要: Immuno-positron emission tomography (immuno-PET) is expected to improve the specificity of small chemical tracers such as 18F-fluorodeoxyglucose. Whole antibodies significantly accumulate in target molecule-expressing tumors but frequently persist too long in the blood circulation for imaging purposes. We investigated the utility of whole antibodies, 64Cu-labeled via a urokinase-substrate linker, and their exogenous urokinase-responsive cleavage to enhance clearance of immuno-PET probes from the blood and shorten the time required to develop adequate imaging contrast. Specifically, we used 64Cu-labeled trastuzumab in human epidermal growth factor receptor 2 (HER2)-positive tumor-bearing mice. 64Cu-labeled trastuzumab with a urokinase-cleavage site (64Cu-CB-TE1A1P-USL-trastuzumab) was synthesized using a bifunctional chelator incorporating a urokinase substrate peptide. Urokinase cleavage was analyzed in vitro by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and radio-gel permeation-high performance liquid chromatography. Improvements in radioisotope clearance and HER2-imaging by urokinase injection were evaluated by PET imaging and ex vivo biodistribution studies in A431 tumor-bearing mice. 64Cu-CB-TE1A1P-USL-trastuzumab was cleaved into smaller radioactive fragments by 20,000 IU/mL urokinase treatment in vitro at an efficacy of ~95%. The probe targeted HER2 in A431 tumors in mice within 24 h post-injection, and approximately two-thirds of the probe in the blood circulation was eliminated via renal clearance of radioactive fragments after three urokinase injections. Therefore, the tumor/blood ratio increased 3.0-fold. Without urokinase injection, the tumor accumulation of 64Cu-CB-TE1A1P-USL-trastuzumab slowly increased and the blood radioactivity decreased over 72 h. However, the tumor/blood ratios in mice after three urokinase injections were higher at 24 h than those in mice without injections at 72 h. The results indicate that our approach shortened the time required to develop adequate imaging contrast of immuno-PET by > 2 days. Therefore, this approach can benefit high-sensitivity imaging under lower radioactive decay conditions and can decrease patient radiation exposure. In addition, it could reduce other adverse effects of radioimmunotherapy.

Theranostic Carbon Dots with Innovative NIR-II Emission for in Vivo Renal Excreted Optical Imaging and Photothermal Therapy
摘要: Carbon dots (CDs) with low biotoxicity, high photostability, and well-controlled small size are highly desirable imaging agents for optical bioimaging. However, most of the CDs triggered by ultraviolet (UV)/blue light present visible/first near infrared (NIR-I) emissions shorter than 820 nm, impairing their imaging applications in vivo by low penetration depth. Hence, developing novel CDs based materials with second near infrared (NIR-II) emission located in 1000-1700 nm region is an urgent task. Here, a novel NIR-II emitting CDs-based nanoprobe triggered by 808 nm laser is developed. The designed CDs with 900-1200 nm luminescence possess high quantum yield (QY-0.4%), and high biocompatibility, which have proven to be effective probes for in vivo NIR-II bioimaging. Notably, nearly 65% CDs are excreted from mouse urine within 6 h, further demonstrating the rapid renal clearance of CDs. Furthermore, the designed CDs also exhibit high photothermal efficiency (30.6%), making them ideal materials for thermal ablation of cancer. Our findings pave the way of designing multifunctional CDs-based theranostic platform for simultaneously integrating the advanced NIR-II bioimaging and photothermal therapy (PTT) of cancer.

Determination of central corneal clearance in scleral lenses with an optical biometer and agreement with subjective evaluation
摘要: Purpose: To compare three methods to measure center corneal clearance (CCC) during scleral lens wear: subjective (slit lamp), image processed (ImageJ) and with an optic biometer. The optic biometer technique was validated in comparison to an OCT in the first part of the study. Methods: Twenty-two eyes (11 subjects) with healthy corneas were recruited. Three measures of OCT with scleral lens and ten measurements of axial length (AL) with IOLMaster with and without lens were performed. For the second part, 61 eyes (35 subjects) enrolled in a clinical study were selected. Measurements of CCC were done with IOLMaster, SlitLamp and ImageJ. Results: The measurements of CCC indirectly obtained with IOLMaster had a strong correlation with AS-OCT measurements (r = 0.981), showing a mean difference of 122.18 ± 46.05 μm (higher with IOLMaster). Regarding the second part, measurements of CCC were 238.66 ± 95.94 μm, 250.16 ± 124.31 μm and 263.15 ± 90.60 μm, for the IOLMaster, SlitLamp and ImageJ, respectively. The correlations were higher for ImageJ vs Subjective measure (r = 0.891) than for IOL vs Subjective (r = 0.748) and IOL vs ImageJ (r = 0.745). Analysis of differences and correlations between SlitLamp and ImageJ through time showed a mean difference of -32.28 ± 89.95 μm (r = 0.683) and 11.57 ± 32.95 μm (r = 0.940) at V12months. Conclusions: It is possible to measure CCC with IOLMaster, considering AL measured with and without lens and lens thickness. The three methods tested have good correspondence, showing that IOLMaster and ImageJ could be objective techniques to measure CCC. Also, it is possible to improve the agreement of subjective measures when compared to objective measures trough time.

Wafer‐Scale Polymer‐Based Transparent Nanocorals with Excellent Nanoplasmonic Photothermal Stability for High‐Power and Superfast SERS Imaging
摘要: To understand the mechanism(s) of age-dependent outcomes of hepatitis B virus (HBV) infection in humans, we previously established an age-related HBV mouse model in which 6-week-old (N6W) C3H/HeN mice exhibited virus tolerance whereas 12-week-old (N12W) counterparts presented virus clearance. By investigating the hepatic myeloid cell dynamics in mice of these two ages, we aim to identify factors associated with HBV clearance. C3H/HeN mice were transfected with an HBV plasmid by hydrodynamic injection. Serum HBV markers were monitored weekly. Hepatic leucocyte populations and their cytokine/chemokine productions were examined at baseline, day 3 (D3), day 7 (D7), and day 14 after injection. C-C chemokine receptor type 2 (CCR2) antagonist and clodronate (CLD) were respectively administered to N12W and N6W mice to study the roles of lymphocyte antigen 6 complex, locus C (Ly6C) monocytes and Kupffer cells (KCs) in viral clearance. N12W mice had a significantly higher number of TNF-α–secreting Ly6C monocytes and fewer IL-10–secreting KCs at D3 in the liver than their younger N6W counterparts after HBV transfection. In addition, the elevated number of interferon-γ+ T cells at D7 was only seen in the older cohort. The enhanced Ly6C monocyte induction in N12W mice resulted from elevated C-C motif chemokine ligand 2 (CCL2) secretion by hepatocytes. CCR2 antagonist administration hampered Ly6C monocyte recruitment and degree of KC reduction and delayed HBV clearance in N12W animals. Depletion of KCs by CLD liposomes enhanced Ly6C monocyte recruitment and accelerated HBV clearance in N6W mice. Conclusions: Ly6C monocytes and KCs may, respectively, represent the resistance and tolerance arms of host defenses. These two cell types play an essential role in determining HBV clearance/tolerance. Manipulation of these cells is a promising avenue for immunotherapy of HBV-related liver diseases.

Evaluation of Safety and Efficacy of Laser Hair Removal With the Long‐Pulsed 755?nm Wavelength Laser: A Two‐Center Study With 948 Patients
摘要: Background and Objectives: Laser hair removal is the most common laser therapy and the third most commonly performed procedure with more than one million treatments in United States in 2016. This retrospective study was conducted to assess long‐term efficacy and safety of the 755 nm laser for hair removal. Study Design/Materials and Methods: Nearly, 3,606 laser treatments were performed with the long‐pulsed 755 nm wavelength laser equipped with an epidermal cooling device between 1997 and 2005 and were followed till 2013. Standardized assessments were conducted by two treating physicians and patients at two follow‐up intervals. At first follow‐up, clearance was assessed by two physicians and clearance and satisfaction by patients. At the second follow‐up, patients were assessed if hair clearance sustained compared with the first follow‐up. Results: Nine hundred and forty‐eight patients with Fitzpatrick skin types I–IV were treated with a total of 3,606 laser treatments in this study. The mean age at the beginning of the study was 35 years (±11), 95.1% of patients were female (n = 902) and 4.9% male (n = 46). Five hundred and seventy‐four patients received a minimum of three treatments and an average of 5.31 (3–16) treatments on axilla, back, bikini, breast, abdomen, face, lower extremity, or upper extremity region. First, follow‐up was conducted 3.9 (±1.5) years after the laser treatment. Seventy‐four percent of these final patients received 75–100% clearance as reported by the physician and 48% clearance as reported by the patient. Fifty‐two percent of patients reported slower hair growth and 42% change in hair texture. Ninety percent of patients treated on axilla, 82% treated on the bikini area, and 79% treated on lower extremities experienced 75% or more clearance after three treatments. Facial, as well as breast and abdomen treatments, only showed a 66% and 62%, respectively, after three treatments. For these locations, five and more treatments were needed to achieve a quote of 79% (face) or 80% (breast and abdomen) for a 75–100% clearance. Upper extremity and back treatments did not have enough physician ratings to draw conclusions. Long‐term adverse events were minimal and were all located on the face (one patient scar, four patients herpes infection). Second follow‐up of 173 patients was conducted after 11.5 years (±2.0) and 87.9% of patients reported that their improvement sustained. Conclusions: The long‐pulsed 755 nm alexandrite laser is a safe and efficacious treatment for the reduction of unwanted body hair with permanent results and high patient satisfaction.

Ablative fractional laser‐assisted photodynamic therapy for lentigo maligna: a prospective pilot study
摘要: Background Lentigo maligna (LM) is an in situ form of melanoma carrying a risk of progression to invasive lentigo maligna melanoma (LMM). LM poses a clinical challenge, with subclinical extension and high recurrence rates after incomplete surgery. Alternative treatment methods have been investigated with varying results. Photodynamic therapy (PDT) with methylaminolaevulinate (MAL) has already proved promising in this respect. Objectives To investigate the efficacy of ablative fractional laser (AFL)-assisted PDT with 5-aminolaevulinic acid nanoemulsion (BF-200 ALA) for treating LM. Methods In this non-sponsored prospective pilot study, ten histologically verified LMs were treated with AFL-assisted PDT three times at 2-week intervals using a light dose of 90 J/cm2 per treatment session. Local anaesthesia with ropivacaine was used. Four weeks after the last PDT treatment the lesions were treated surgically with a wide excision and sent for histopathological examination. The primary outcome was complete histopathological clearance of the LM from the surgical specimen. Patient-reported pain during illumination and the severity of the skin reaction after the PDT treatments were monitored as secondary outcomes. Results The complete histopathological clearance rate was 7 out of 10 LMs (70%). The pain during illumination was tolerable, with the mean pain scores for the PDT sessions on a visual assessment scale ranging from 2.9 to 3.8. Some severe skin reactions occurred during the treatment period, however. Conclusions Ablative fractional laser-assisted PDT showed moderate efficacy in terms of histological clearance. It could constitute an alternative treatment for LM but due to the side effects it should only be considered in inoperable cases.
