研究目的
To understand the mechanism(s) of age-dependent outcomes of hepatitis B virus (HBV) infection in humans and to identify factors associated with HBV clearance by investigating the hepatic myeloid cell dynamics in mice of different ages.
研究成果
Ly6C monocytes and KCs play essential roles in determining HBV clearance/tolerance, representing the resistance and tolerance arms of host defenses, respectively. Manipulation of these cells is a promising avenue for immunotherapy of HBV-related liver diseases.
研究不足
The study is limited to a mouse model, and findings may not directly translate to human HBV infection. The role of TLR4 in promoting KC depletion needs further validation due to potential genetic alterations in the C3H/HeJ strain.
1:Experimental Design and Method Selection:
Established an age-related HBV mouse model to study hepatic myeloid cell dynamics. Used hydrodynamic injection for HBV plasmid transfection.
2:Sample Selection and Data Sources:
C3H/HeN mice of two ages (6-week-old and 12-week-old) were used. Serum HBV markers and hepatic leucocyte populations were monitored.
3:List of Experimental Equipment and Materials:
HBV plasmid, CCR2 antagonist, clodronate liposomes, flow cytometry for cell analysis, ELISA for cytokine measurement.
4:Experimental Procedures and Operational Workflow:
Mice were transfected with HBV plasmid, serum markers monitored weekly, hepatic leucocyte populations examined at specific time points, and treatments administered to study roles of specific cell types.
5:Data Analysis Methods:
Flow cytometry data analysis, statistical analysis using Student t tests, cytokine measurement via ELISA.
