Layer-by-layer Modification of Magnetic Graphene Oxide by Chitosan and Sodium Alginate with Enhanced Dispersibility for Targeted Drug Delivery and Photothermal Therapy
摘要: In this work, graphene oxide nanosheets loaded by magnetic iron oxide nanoparticles (mGO) was synthesized and the technique of layer-by-layer (LbL) self-assembly was utilized in the successful production of chitosan/ sodium alginate functionalized mGO naocomposites for use in targeted anti-cancer drug delivery and photothermal therapy. The mGO-CS/SA nanocomposites had a diameter of ~0.5 μm and a thickness of 40-60 nm with superparamagnetic behavior. The modified nanocomposites exhibited a decrease in agglomeration and an increase in stability in biological solution following stability tests. Meanwhile, the nonspecific protein adsorption was strongly suppressed after the modification. The mGO-CS/SA nanocomposites were loaded with doxorubicin hydrochloride (DOX) via π- π stacking and electrostatic attraction with a high drug loading amount (121%, w/w). The DOX-loaded nanocomposites (mGO-CS/SA-DOX) showed improvements in function including enhanced dispersion and noticeable pH-sensitive drug release behavior. Cellular studies denoted magnetically targeted cellular uptake characteristics and excellent photothermal effect of mGO-CS/SA, as well as concentration-dependent cytotoxicity of mGO-CS/SA-DOX. Therefore the functionalization of mGO using chitosan and sodium alginate would be beneficial in biomedical applications.

Extracellular vesicles based self-grown gold nanopopcorn for combinatorial chemo-photothermal therapy
摘要: Here, we generated a popcorn-like gold nanostructure exploiting extracellular vesicles (EVs). EVs can first serve as the vehicle for chemotherapeutic drug doxorubicin (DOX). Taking advantages of EVs, gold nanoparticles can be then self-grown surrounding the EVs, assembling into popcorn-like nanostructure. The formulated nanopopcorn, consisting of self-grown gold nanoparticles and EVs encapsulated with DOX, retained the photothermal transduction from gold nanoparticle assemblies and cytotoxicity of DOX. Under external near infrared irradiation, gold nanopopcorn can produce hyperthermia to induce tumor ablation and trigger drug release, achieving combinatorial chemo-photothermal therapy. The nanoplatform demonstrated improved cellular internalization, controlled drug release, enhanced antitumor efficacy with tumor inhibitory rate up to 98.6% and reduced side effects. Our design of popcorn-like nanostructure will contribute a novel modality for facile and green synthesis of complex metal nanostructures exploiting natural properties of EVs for combinational therapy.

Ultrastructural and optical characteristics of cancer cells treated by a nanotechnology based chemo-photothermal therapy method
摘要: The current chemotherapy method demonstrates the need for improvement in terms of efficacy and safety. Given the beneficiary effect of heat in combination with chemotherapy, the purpose of this study is to develop a multifunctional nanoplatform by co-incorporating gold nanoparticles (AuNPs) as photothermal agent and cisplatin as anticancer drug into alginate hydrogel (named as ACA) to enable concurrent thermo-chemotherapy. The in vitro cytotoxicity experiment showed that the as-developed nanocomplex was able to induce greater cytotoxicity in KB human nasopharyngeal cancer cells compared to free cisplatin at the same concentration. Moreover, the interaction of ACA and laser irradiation acted synergistically and resulted in higher cell death rate compared to separate application of photothermal therapy and chemotherapy. The micrograph of KB cells also revealed that ACA was able to selectively accumulate into the mitochondria, so that laser irradiation of KB cells pre-treated with ACA resulted in intensive morphological damages such as plasma membrane disruption, chromatin condensation, autophagic vacuoles formation and organelle degeneration. Moreover, the sign and magnitude of altered optical nonlinear refractive index measured by Z-scan technique was shown to be significantly in cells exposed to irradiation. Consequently, the nanocomplex developed herein could be a promising platform to combine photothermal therapy and chemotherapy effectively, thereby achieving synergistic therapeutic outcome.

Ultrathin Tellurium Oxide/Ammonium Tungsten Bronze Nanoribbon for Multi-Modality Imaging and Second Near-Infrared Region Photothermal Therapy
摘要: Developing nano-photothermal agents (PTAs) with satisfied photothermal conversion efficiency (PTCE) in the second NIR window (1000-1350 nm, NIR II) holds great promise for enhanced photothermal therapy (PTT) effect. Herein, we develop a NIR-II PTA with advanced PTCE, based on a new two-dimensional (2D) ultrathin tellurium oxide/ammonium tungsten bronze (TeO2/(NH4)xWO3) nanoribbons (TONW NRs). The doped ammonia ions mediated-free electrons injection into the LUMO band of WO3 combined with the electronic transitions between W6+ ions and the lone pair of electrons in Te atoms achieve excellent NIR absorption of TONW NRs resulted from localized surface plasmon resonance (LSPR). The polyethylene glycol functionalized TONW NRs (PEG-TONW NRs) exhibit good stability and biocompatibility, displaying a PTCE high to 43.6%, surpassing many previous nano-PTAs active in the NIR II region, leading to remarkable tumor ablation ability both in vitro and in vivo. Meanwhile, advanced X-ray computed tomography (CT) and photoacoustic (PA) imaging capability of PEG-TONW NRs were also realized. Given the admirable photothermal effect in NIR II region, good biocompatibility and advanced CT/PA imaging diagnosis capability, the novel PEG-TONW NRs is promising in future personalized medicine application.

Theranostic Carbon Dots with Innovative NIR-II Emission for in Vivo Renal Excreted Optical Imaging and Photothermal Therapy
摘要: Carbon dots (CDs) with low biotoxicity, high photostability, and well-controlled small size are highly desirable imaging agents for optical bioimaging. However, most of the CDs triggered by ultraviolet (UV)/blue light present visible/first near infrared (NIR-I) emissions shorter than 820 nm, impairing their imaging applications in vivo by low penetration depth. Hence, developing novel CDs based materials with second near infrared (NIR-II) emission located in 1000-1700 nm region is an urgent task. Here, a novel NIR-II emitting CDs-based nanoprobe triggered by 808 nm laser is developed. The designed CDs with 900-1200 nm luminescence possess high quantum yield (QY-0.4%), and high biocompatibility, which have proven to be effective probes for in vivo NIR-II bioimaging. Notably, nearly 65% CDs are excreted from mouse urine within 6 h, further demonstrating the rapid renal clearance of CDs. Furthermore, the designed CDs also exhibit high photothermal efficiency (30.6%), making them ideal materials for thermal ablation of cancer. Our findings pave the way of designing multifunctional CDs-based theranostic platform for simultaneously integrating the advanced NIR-II bioimaging and photothermal therapy (PTT) of cancer.

Multifunctional nanoplatforms for photoacoustic imaging-guided combined therapy enhanced by CO induced ferroptosis
摘要: A multifunctional CO/thermo/chemotherapy nanoplatform is here reported, which is composed of mesoporous carbon nanoparticles (MCN) as near-infrared (NIR)-responsive drug carrier, doxorubicin (DOX) as chemotherapeutic drug and triiron dodecacarbonyl (FeCO) as thermosensitive CO prodrug. The nanoplatform could absorb near-infrared (NIR) light and convert it into ample heat to trigger CO release and could also release DOX in the acidic tumor microenvironment. More importantly, the generated CO molecules successfully increase cancer cell sensitivity to chemotherapeutics by the ferroptosis pathway. Subsequently, under the guidance of photoacoustic imaging, the FeCO-DOX@MCN nanoplatform demonstrates high treatment efficacies in vitro and in vivo by combination of chemotherapy, photothermal therapy and gas therapy. This multifunctional platform with excellent antitumor efficacy has great potential in precision cancer therapy.

Supramolecular Photothermal Nanomaterials as an Emerging Paradigm toward Precision Cancer Therapy
摘要: The concept of the “supramolecular photothermal effects” refers to the collection property and photothermal conversion efficiency resulting from the supramolecular assembly of molecular photothermal sensitizers. This review considers organic supramolecular photothermal materials assembled at the nanoscale via various molecular self-assembly strategies and associated with the organization of multiple noncovalent interactions. In these materials, the individual photosensitizer molecules are typically aggregated through self-assembly in a certain form that exhibits enhanced biostability, increased photothermal conversion efficiency with photoluminescence quenching, and improved photothermal therapeutic effects in comparison with those of the monomeric photosensitizer molecules. These supramolecular photothermal effects are controlled or influenced by intermolecular noncovalent interactions, especially the hydrophobic effects, which are distinct from the mechanisms of conventional sensitizer molecules and polymers and inorganic photothermal agents. A focus lies on how self-assembly strategies give rise to supramolecular photothermal effects, including polymer and protein fabrication, small molecule self-assembly, and the construction of donor–acceptor binary systems. Emphases are placed on the rational design of supramolecular photothermal nanomaterials, drug delivery, and in vivo photothermal therapeutic effects. Finally, the key challenges and promising prospects of these supramolecular photothermal nanomaterials in terms of both technical advances and clinical translation are discussed.

Enhanced photoacoustic and photothermal effect of functionalized polypyrrole nanoparticles for near-infrared theranostic treatment of tumor
摘要: Functionalized nanomaterials with near-infrared (NIR) responsive capacity are quite promising for theranostic treatment of tumors, but formation of NIR responsive nanomaterials with enhanced theranostic ability and excellent biocompatibility is still very challenging. Herein, PEGylated indocyanine green (ICG)-loaded polypyrrole nanoparticles (PPI NPs) were designed and successfully formed through selecting polydopamine as the linkage between each component, demonstrating enhanced NIR responsive theranostic ability against tumor. Combining in vitro cell study with in vivo assay, the formed PPI NPs were proved being fantastic biocompatible while effectively internalizing in HeLa cells and retaining in HeLa tumor demonstrated by in vitro flow cytometry/confocal measurement and in vivo photoacoustic imaging assay. With the guidance of photoacoustic imaging, successful photothermal ablation of tumor was achieved when treating with PPI NPs plus laser, which was much more effective than the group treated with NPs free of ICG. The greatly combined enhanced photoacoustic and photothermal effect is mainly ascribed to the functionalized polypyrrole nanoparticles, which could accumulate in tumor site more effectively with a relative longer retention time taking advantage of the nanomaterial-induced endothelial leakiness phenomenon. All these results demonstrate the designed PPI NPs possess enhanced NIR responsive property are to hold a great promise for tumor NIR theranostic applications.

Engineering Metal Organic Frameworks for Photoacoustic Imaging Guided Chemo/Photothermal Combinational Tumor Therapy
摘要: Imaging-guided therapy has considerable potential in tumor treatment. Different treatments have been integrated to realize combinational tumor therapy with improved therapeutic efficiency. Herein, the conventional metal organic framework (MOF) MIL-100 is utilized to load curcumin with excellent encapsulation capacity. Polydopamine-modified hyaluronic acid (HA-PDA) is coated on MIL-100 surface to construct engineering MOF nanoparticles (MCH NPs). The HA-PDA coating not only improves the dispersibility and stability of NPs but also introduces a tumor-targeting ability to this nanosystem. A two-stage augmented photothermal conversion capability is introduced to this nanosystem by encapsulating curcumin in MIL-100 pores and then coating HA-PDA on the surface, which confer the MCH NPs with strong photothermal conversional efficiency. After being intravenously injected into xenograft HeLa tumor-bearing mice, MCH NPs prefer to accumulate at the tumor site and achieve photoacoustic imaging-guided chemo/photothermal combinational tumor therapy, generating nearly complete tumor ablation. Engineering MOFs is an efficient platform for imaging-guided combinational tumor therapy, as confirmed by in vitro and in vivo evaluations.

PEGylated polyethylenimine-stabilized polypyrrole nanoparticles loaded with DOX for chemo-photothermal therapy of cancer cells
摘要: Combination of kinds of therapy modalities is promising for effective cancer treatment. Herein, a kind of multifunctional nanoparticles (NPs) was developed for cancer chemo-photothermal therapy applications. Polypyrrole (PPy) NPs were formed using a facile polymerization method using poly(ethyleneimine) (PEI) as stabilizer, followed by polyethylene glycol (PEG) modification and anticancer drug doxorubicin (DOX) loading. Showing obvious absorbance in the NIR range, the obtained PPy-PEI-PEG NPs displayed well photothermal ability with desirable photothermal stability. The release of the loaded DOX can be promoted by pH and laser stimulation. Compared with single therapy modality, the combination of chemotherapy and photothermal therapy showed higher cancer cell killing effect. The cellular internalization of the obtained NPs was proved to be effective. The developed multifunctional NPs are promising candidates for combined therapy of cancer cells.
