The engineered MCH NPs are promising candidates for imaging-guided chemo/photothermal combinational cancer therapy, as confirmed by in vitro and in vivo evaluations. The MCH NPs exhibited high curcumin-loading capability, improved stability, tumor-targeting ability, and strong photothermal conversion efficiency, leading to nearly complete tumor ablation in vivo.

The study does not address the potential long-term toxicity of the MCH NPs or their biodegradability in vivo. The photothermal conversion efficiency, while comparable to other agents, could be further optimized.
1:Experimental Design and Method Selection:
The study utilized MIL-100 MOF to load curcumin and coated it with polydopamine-modified hyaluronic acid (HA-PDA) to construct MCH NPs. The methodology included microwave-assisted synthesis of MIL-100, loading of curcumin, and coating with HA-PDA.
2:Sample Selection and Data Sources:
HeLa cells, A549 cells, CHO cells, and MRC-5 cells were used for in vitro studies, and HeLa tumor-bearing mice were used for in vivo studies.
3:List of Experimental Equipment and Materials:
Equipment included a microwave reactor, UV-Vis spectrophotometer, infrared thermographic camera, and multispectral optoacoustic tomography (MSOT) system. Materials included FeCl3·6H2O, BTC, HA, curcumin, and dopamine hydrochloride.

The procedures included synthesis of MIL-100, curcumin loading, HA-PDA coating, photothermal effect measurement, drug release behavior study, stability tests, cellular uptake and cytotoxicity assays, in vivo imaging, and tumor therapy evaluation.

Data were analyzed using statistical methods including Student’s t-test, with results presented as mean ± standard deviation.
