研究目的
To identify MALDI-TOF MS biomarkers able to differentiate the subspecies of Staphylococcus schleiferi (S. schleiferi subsp. schleiferi and S. schleiferi subsp. coagulans).
研究成果
The present study identified a set of biomarkers able to discriminate S. schleiferi subspecies by MALDI-TOF mass spectrometry. These biomarkers could help in the determination of novel and improved targets that may assist in the identification of these species/subspecies in clinical samples.
研究不足
The study identified a set of biomarkers for discriminating S. schleiferi subspecies, but further studies are required to improve reproducibility and attest the validity of these biomarkers for this purpose.
1:Experimental Design and Method Selection:
The study used MALDI-TOF MS for the identification of S. schleiferi subspecies.
2:Sample Selection and Data Sources:
Twelve strains (eight S. schleiferi schleiferi and four S. schleiferi coagulans) were selected from a collection of 76 S. schleiferi isolates.
3:List of Experimental Equipment and Materials:
Microflex LT mass spectrometer (Bruker Daltonics), MSP 96 BC target polished steel (Bruker Daltonics, Germany), HCCA matrix (α-cyano-4-hydroxycinnamic acid), formic acid (Sigma-Aldrich), acetonitrile PA (Sigma-Aldrich).
4:Experimental Procedures and Operational Workflow:
Protein extraction protocol was performed, followed by MALDI-TOF MS runs. Spectra were analyzed with BioNumerics software v
5:Data Analysis Methods:
Spectra were processed and analyzed using default parameters, and a set of five MALDI-TOF MS biomarkers were selected to enable the discrimination of S. schleiferi subspecies.
